Abstract
막걸리는 우리나라를 대표하는 전통주 중 하나이다. 본 연구는 Saccharomycopsis fibuligera와 Aspergillus oryzae를 접종한 koji로 제조한 막걸리의 휘발성 및 비휘발성 대사산물을 프로파일링 하기 위해 수행되었다. 이 때, 서로 다른 종류의 koji는 쌀의 proteins, starch, lipids와 같이 장차 Saccharomyces cerevisiae에 의해 대사되는macromolecules의 분해에 주요한 영향을 미친다. 따라서, koji의 enzyme activity가 막걸리의 대사산물 생성에 미치는 영향 역시 함께 밝히고자 하였다. 그 결과, 18종의 amino acids, 15종의 carbohydrates, 20종의 organic acids, 12종의 fatty acids를 포함한 총 65종의 비휘발성 성분이 유도체화되어, gas chromatograph time-of-flight mass spectrometer로 분석되었다. 그 중, alanine과 succinic acid, palmitic acid는 S. fibuligera CN2601-09 koji로 제조한 막걸리에서 가장 높은 함량으로 검출된 반면, galactose는 S. fibuligera KJJ81 koji로 제조한 막걸리에서 가장 높은 함량으로 검출되었다. 또한, 18종의 amino acids는 S. fibuligera CN2601-09 koji로 제조한 막걸리에서 그 함량이 높게 나타났으며, A. oryzae CN1102-08과 함께 접종하여 이용했을 때, 그 함량이 증가하는 경향을 나타내었다. 그러나 대부분의 fatty acids는 A. oryzae CN1102-08과 함께 접종하여 이용했을 때, 오히려 그 함량이 감소하는 경향을 나타내었다. 한편, 4종의 acids, 11종의 alcohols, 7종의 aldehydes, 3종의 benzene derivatives, 49종의 esters, 1종의 furan derivative, 2종의 hydrocarbons, 1종의 ketone, 1종의 lactone, 5종의 phenols을 포함하는 총 84종의 휘발성 성분이 solid phase micro-extraction으로 추출되어 gas chromatograph-mass spectrometer로 분석되었다. 그리고 fusel alcohols, acetate esters와 함께 fatty acids의 ethyl esters가 주요 성분으로 검출되었다. 대부분의 fusel alcohols와 acetate esters는 가장 높은 protease 활성도를 보인 S. fibuligera CN2601-09 koji로 제조한 막걸리에서 높은 함량으로 검출되었다. 또한 이들은 A. oryzae CN1102-08과 함께 접종하여 이용했을 때, 그 함량이 유의적으로 증가하는 경향을 나타내었다. 한편, 17종의 ethyl esters 중, ethyl hexadecanoate에 이어 ethyl decanoate와 ethyl octanoate가 S. fibuligera koji를 이용해 제조한 막걸리에서 높은 함량으로 검출되었다. 이 때, ethyl hexadecanoate의 함량은 ethanol 함량이 높은 S. fibuligera KJJ81 koji로 제조한 막걸리에서 가장 높게 나타난 반면, ethyl decanoate와 ethyl octanoate의 함량은 lipase의 활성도가 가장 높은 S. fibuligera CN2601-09 koji로 제조한 막걸리에서 높게 나타났다. 그러나 위 성분들 모두 A. oryzae CN1102-08과 함께 접종하여 이용했을 때, 그 함량이 유의적으로 증가하는 경향을 나타내었다. 비휘발성 성분의 PCA plot에서 S. fibuligera koji로 제조한 막걸리는 A. oryzae를 함께 접종하여 제조한 막걸리와 분리되어나타났다. 그리고 이 때, 모든 amino acids와 xylose, 2-methylbenzoic acid, oxalic acid와 같은 일부 대사산물이 A. oryzae 혹은 이를 S. fibuligera와 함께 접종한 막걸리에 주요하게 작용하는 것을 알 수 있었다. 반면 휘발성 성분의 PCA plot에서는, S. fibuligera KPH12와 A. oryzae CN1102-08을 함께 접종한 koji를 이용해 제조한 막걸리가 다른 막걸리 샘플과 뚜렷하게 분리되어 나타났다. 그리고 여기에는 butanoic acid, 1-butanol, nonanal, benzaldehyde, 4-ethyl-2-methoxy-phenol, 4-ethylphenol과 함께 butanoic acid와 1-butanol에서 유래한 esters가 주요하게 영향을 미쳤다.;Makgeolli is one of the most popular traditional alcoholic beverages in Korea. This study investigated volatile and non-volatile metabolite profiles of makgeolli fermented by koji inoculated with Saccharomycopsis fibuligera and/or Aspergillus oryzae. The different types of koji could mainly affect the degradation of macromolecules such as proteins, starch and lipids in rice, which was further metabolized in combination with Saccharomyces cerevisiae. Therefore, activities of enzymes in koji were also examined to determine their effects on the formation of metabolites in makgeolli. A total of 65 non-volatile metabolites, including 18 amino acids, 15 carbohydrates, 20 organic acids and 12 fatty acids, were identified by gas chromatograph coupled with time-of-flight mass spectrometer after derivatization. Among them, makgeolli fermented by S. fibuligera CN2601-09 koji had the highest levels of alanine, succinic acid and palmitic acid, while makgeolli fermented by S. fibuligera KJJ81 koji had the highest levels of galactose. In addition, the contents of all 18 amino acids were highest in makgeolli fermented by S. fibuligera CN2601-09 koji, and they tended to increase after combination with A. oryzae CN1102-08. However, the contents of most fatty acids showed a tendency to decrease after combination with A. oryzae CN1102-08. On the other hand, a total of 84 volatile compounds, including 4 acids, 11 alcohols, 7 aldehydes, 3 benzene derivatives, 49 esters, 1 furan derivative, 2 hydrocarbons, 1 ketone, 1 lactone, and 5 phenols, were extracted by solid phase micro-extraction and analyzed by gas chromatograph-mass spectrometer. The major volatile compounds were fusel alcohols, acetate esters, and ethyl esters of fatty acids. The contents of most fusel alcohols and acetate esters were highest in makgeolli fermented by S. fibuligera CN2601-09 koji as enzymatic activity of protease was highest in koji inoculated with it. Also, they tended to increase significantly after combination with A. oryzae CN1102-08. On the other hand, among the 17 ethyl esters, ethyl hexadecanoate was most abundant in makgeolli fermented by S. fibuligera koji followed by ethyl decanoate and ethyl octanoate. The level of ethyl hexadecanoate was highest in makgeolli fermented by S. fibuligera KJJ81 koji which has the highest content of ethanol, while ethyl decanoate and ethyl octanoate were highest in makgeolli fermented by S. fibuligera CN2601-09 koji as enzymatic activity of lipase was highest in koji inoculated with it. However, they all tended to increase significantly after combination with A. oryzae CN1102-08. On PCA plot of non-volatile metabolites, makgeolli samples fermented by S. fibuligera koji were discriminated from others those were produced in cooperation with A. oryzae. Also, all amino acids and some metabolites such as xylose, 2-methylbenzoic acid, and oxalic acid contributed mainly to the differentiation of makgeolli samples fermented by koji inoculated with A. oryzae and/or S. fibuligera. However, on PCA plot of volatile compounds, makgeolli sample fermented by koji inoculated with both S. fibuligera KPH12 and A. oryzae CN1102-08 was separated clearly from other makgeolli samples. Butanoic acid, 1-butanol, nonanal, benzaldehyde, 4-ethyl-2-methoxy-phenol, 4-ethylphenol and some esters that derived from 1-butanol and butanoic acid were mainly responsible for the discrimination.
