Abstract
Sprouts have been related to numerous foodborne outbreaks worldwide. Previous studies that foodborne bacteria can contaminate from seeds and survive up to sprouts suggested the importance of investigating microbial changes during sprout growth, but little is known about the shifts in microbial communities from seeds to sprouts. This study aimed to identify bacterial changes during sprout production procedures of alfalfa, radish, and rapeseed. Samples were collected from seven production procedures (seed, soaking, germination 1, germination 2, sprouting, unwashed, and washed sprouts) and subjected to microbiome with 16S rRNA sequencing as well as microbial quality analyses (n = 105). The results showed that bacterial counts rapidly increased within one day and alfalfa sprout had the highest number of bacteria among other types. Total aerobic bacteria (TAB) and coliforms grew up to 7-9 log CFU/g at the final procedure of the production. E. coli O157:H7, Salmonella Typhimurium, and Listeria monocytogenes were not detected in all samples. Microbiome analysis revealed that Proteobacteria (phylum) and Enterobacteriaceae and Pseudomonadaceae (family) were the most dominant strains. Pseudomonadaceae grew predominantly only in radish and rapeseed at germination 2 and sprouting procedure, respectively. Alfalfa had the significantly highest value of alpha diversity in seed among other types but its community structure became simplified due to the rapid growth of Enterobacteriaceae (73.8%) at the soaking procedure. Considering the fact that no significant growth was observed in TAB count at the soaking procedure, this result indicates an apparent growth of Enterobacteriaceae. Since Enterobacteriaceae is widely used as a hygiene indicator to ensure food safety, managing soaking procedure in alfalfa could be effective for controlling sprout production. Beta diversity confirmed that washing could not change the bacterial community and the patterns of ecology changes were differed by seed type. This study is the first study to examine microbiome profile during sprout growth and the results could be used for establishing effective control points in sprout production.;새싹채소로 인한 식중독은 전 세계적으로 꾸준하게 보고되고 있어 새싹채소의 미생물학적 안전성 확보가 요구되고 있다. 다수의 연구는 식중독 세균이 종자를 오염시킬 수 있고 새싹채소까지 살아남아 문제가 될 수 있음을 보이며 재배 과정 중 미생물 생태 변화를 구명하는 것의 중요성을 시사하였으나 이와 관련된 연구는 미흡한 실정이다. 본 연구는 배양법 기반의 미생물 분석(총 세균과 대장균군의 정량분석 및 주요 식중독세균의 정성분석)과 high throughput sequencing(HTS) 분석을 통한 16S rRNA 마이크로바이옴 분석을 이용하여 종자부터 새싹채소까지의 성장 과정 중 미생물학적 변화를 심층적으로 분석하였다. 실제 새싹채소 생산 공정을 고려하여 재배 7단계(씨앗, 침지, 발아 1, 발아 2, 싹 틔움, 미세척, 및 세척 새싹)를 설정하였고 새싹채소 세 종류(알팔파, 무순, 유채)에 대한 재배를 진행하여 단계별로 시료를 채취하였다(총 105건). 배양기반의 분석 결과, 씨앗에는 종자 종류별로 2~4 log CFU/g의 총 세균수, 불검출~2 log CFU/g의 대장균군이 존재하다가 재배 후 1~2일 내로 급격하게 증가하며 최종 새싹채소에서는 총 세균수와 대장균군이 최대 7~9 log CFU/g까지 나타났다. 알팔파는 세 종류의 새싹 중 가장 높은 총 세균수와 대장균군 수를 나타냈으며 E. coli O157:H7, Listeria monocytogenes, Salmonella Typhimurium은 모든 샘플에서 불검출되었다. 마이크로바이옴 분석 결과, 새싹 종류별로 우점종과 군집 변화 양상이 다르게 나타났다. 전반적으로 문(phylum) 수준에서는 Proteobacteria가, 과(family) 수준에서는 Enterobacteriaceae(최종단계의 알팔파: 80.97%, 무순: 37.72%, 유채: 24.11%)와 Pseudomonadaceae(최종 단계의 알팔파: 8.74%, 무순: 32.31%, 유채: 61.04%)가 우점종으로 나타났다. 이중 Pseudomonadaceae는 무순과 유채에서 우세하게 나타났으며 우점 시기는 각각 발아 2와 싹 틔움 단계였다. 알팔파 씨앗은 다른 종류와 비교하여 유의적으로 높은 수치의 알파 다양성 값을 나타내며 풍부한 종 다양성을 보였으나 침지 단계부터 Enterobacteriaceae가 급증(73.8%)하며 우점종으로 나타났다. 총 세균수, 대장균군, Enterobacteriaceae는 위생지표세균으로 널리 활용되고 있으며, 본 실험 결과 다른 새싹채소에 비해 알팔파에서 세 가지 수치가 모두 높게 나타났다. 이는 알팔파에서 식중독 사건이 주로 발생하는 원인에 대한 과학적 근거가 될 수 있다. 알팔파의 경우 침지 단계에서 Enterobacteriaceae의 급격한 성장을 보였으며, 이는 침지 단계를 관리하는 것이 알팔파의 미생물학적 안전성 확보에 효율적이며, 마이크로바이옴 분석이 효과적인 위생관리지점을 설정하는 데 활용될 수 있음을 시사한다. 베타 다양성 분석 결과, 미생물 군집 변화 패턴은 종자 종류에 따라 상이하였고 세척은 새싹 군집을 유의적으로 변화시키지 못하였다. 본 연구는 배양법 기반의 미생물 분석과 16S rRNA 염기서열해독을 통해 종자부터 새싹채소까지의 미생물학적 군집 변화를 규명하고자 한 첫 연구로, 연구 결과는 새싹 종류별로 효과적인 관리 지점을 설정할 때 기초 자료로 활용될 수 있다.
