Abstract
본 연구에서는 유산균 Lactobacillus paracasei와 Lactobacillus plantarum을 각각 접종시킨 귀리를 24시간 동안 발효시켜 나타나는 휘발성 성분의 차이를 규명하고자 하였다. 고체상미세추출법 (solid-phase microextraction)을 이용하여 시료의 상부공간에 평형 상태로 있는 휘발성 성분을 추출하고, 기체 크로마토그래피-질량분석법 (gas chromatography-mass spectrometry)으로 귀리발효물의 휘발성 성분을 분석하였다. 시료에 따른 차이는 주성분분석법 (principal component analysis)을 수행하여 시각화하고 분석하였다. 총 61 가지의 성분들이 L. paracasei을 접종시킨 귀리발효물에서 검출되었고, L. plantarum을 접종시킨 시료에서는 62 가지의 성분들이 검출되었다. 발효가 진행될 수록 acetic acid, 3-methylbutanoic acid, hexanoic acid와 같은 산이 증가하였는데 다른 시료와 24시간 발효된 시료의 차이에 주요 성분으로서 기여하였다. L. plantarum으로 발효된 귀리 시료들이 L. paracasei으로 발효된 귀리 시료들보다 control과의 차이가 더 적었다. 특히 L. plantarum 발효 초기 (2, 4시간)의 귀리발효물과 control은 PCA plot상에서 큰 상관관계를 나타내었다.;This study investigated volatile metabolite profiles of oat fermented by Lactobacillus paracasei or Lactobacillus plantarum. Gas chromatography-mass spectrometry (GC/MS) coupled with head-space solid-phase microextraction (HS-SPME) method was applied for qualitative and quantitative analysis of the metabolites. A total of 61 and 62 compounds were identified in oat fermented by L. paracasei and L. plantarum, respectively. In addition, principal component analysis (PCA) was utilized to discriminate fermented oat samples. Initial (including the control) and later fermentation stage could be discriminated in PCA mainly by acids (acetic acid, 2-methylpropanoic acid, 3-methylbutanoic acid, hexanoic acid, pentanoic acid, heptanoic acid, octanoic acid, and butanoic acid), aldehydes (hexanal and 3-methylbutanal), and ketones (2-heptanone, 2-octanone, 3-hydroxy-2-butanone, 2-nonanone, and 2-undecanone). The contents of acids, ketones, 2-pentylfuran, and pyruvate metabolism products generally increased over fermentation times in both species, where acids contributed the highest to samples fermented for 24 hours. On the other hand, samples fermented with L. plantarum did not differ significantly from the control as compared to those fermented by L. paracasei. Samples at initial fermentation stage (2 and 4 hours) of L. plantarum were particularly closely correlated to control in PCA plot.
