dc.description.abstractThis study was performed to investigate the effects of dietary protein level and food restriction on protein and Ca metabolism and bone growth in rats. Male rats of Sprague-Dawley strain weighing 100.5g were divided into 3 groups. Rats in Control group were fed 16% case in diet. ad libitum, those in Low-protein group were fed 9%, those in Restriction group were given 70% of feed consumed by control group. The animals sacrificed at 1mo. & 4mos. after experiment begins. Food consumptions, body weight gain of the rats in Control group were significantly higher than those in Low-protein and Restriction groups. Even though rats in Low-protein group were fed ad libitum, their food consumptions were lower than Control group, and similar to Restriction group. There were no significant differences in DNA & RNA content & RNA/DNA ratio of brain among experimental groups. RNA content, RNA/DNA ratio of the rats in Low-protein group were lower than other groups after 1mo. but this tendency were disappeared after 4 mos. Protein levels of liver & hind limb muscles, urinary Ca & fecal Ca excretion showed no significant differences among experimental groups through the experimental period. The skeleton growth (femur & scapulus) were affected not only by dietary protein levels, but also by food restriction. The weight of femur & scapulus increased with increased level of dietary protein. There were no differences in bone composition among experimental diet groups, but changes of bone composition were found with increase of age; increased ash proportion, decreased water proportion and increased ash VS. organic material ratio were found after 4 mos. period as compared with after 1 mo. In conclusion, by dietary protein content and food restriction growth and bone growth of rats were retarded, RNA content of brain in 1 mo. period was affected, but not affected at 4mos, indicating the adaptation might be occurring with age.;본 연구에서는 실험 기간을 1개월과 4개월로 달리하여 젖 뗀 흰쥐에게 Control군은 16% Casein식이를 자유로, Low-Protein군은 Casein 9%식이를 자유로 섭취케하고, Restriction군은 Control군의 식이 섭취량의 70%를 공급하여 사육하였을때 체내 단백질대사, Ca 대사 및 뼈의 성장에 미치는 영향을 조사하였다. 식이 섭취량, 체중 증가량은 단백질의 수준에 따라 차이를 보여 Control군이 Low-Protein군보다 많았으며, Low-Protein군은 식이를 자유섭취 했음에도 불구하고 식이제한을 받은 Restriction군과 식이섭취량이 거의 비슷했다. 뇌의 DNA와 RNA함량, RNA/DNA ratio는 식이 섭취기간과 식이에 따른 유의적 차이는 없었지만, 1개월 사육한 쥐에서 뇌의 RNA 함량과 RNA/DNA ratio는 Control군이 Low-Protein군보다 높았으며, 4개월사육한 쥐에서는 식이에 따른 차이가 없었다. 이는 식이섭취기간이 지나면서 실험군이 식이에 따라 적응하는 것으로 보인다. 간 단백질함량, 근육의 단백질함량은 식이 섭취기간과 식이에 따른 유의적 차이가 없었다. 뇨 Ca 배설량, 변 Ca 배설량도 식이 섭취기간과 식이에 따른 차이를 보이지 않았다. 뼈 (대퇴골과 견갑골)는 단백질 수준 및 식이 제한에 의해 영향을 받아 Control군의 뼈무게, 회분함량이 다른 두군에 비해 많았다. 골격의 구성성분 비율은 식이에 따른 차이는 보이지 않았지만, 식이 섭취기간에 따라서 수분 함유율은 낮아졌고 회분 함유율은 증가하였으며 뼈의 마른 무게중의 무기물 대 유기물 비율이 높았다. 이는 연령의 증가에 따라 나타나는 골격 성분의 변화로 볼 수 있을 것이다. 이상의 결과로 볼 때, 본 연구의 단백질 수준 및 식이 제한은 동물의 성장과 뼈의 성장을 지체시켰고, Low-Protein군과 Restriction군의 체내 대사에 미치는 영향은 거의 비슷한 것으로 나타났으며, 본 연구에서 조사한 모든 항목에서 Control군보다 저조했다.-
