Abstract
Neuroblastoma (NB) originated from immature neuronal cells has poor clinical outcome. NB cells possess cancer stem cells (CSCs) characteristics that facilitate the initiation of a tumor, as well as its metastasis. The human bitter taste receptors (referred to as TAS2Rs) are one of five basic taste receptors and belong to a family of G-protein coupled receptors (GPCRs). Recent reports about the expression of taste receptors in non-gustatory tissues suggest their additional functions distinct from taste perception. While it is generally admitted that the recognition bitter taste may be associated with a self-defense system against the ingestion of poisonous food compounds or disease-related function in human body, the anti-cancer stemness and invasion effects of TAS2Rs on cancer cells remain poorly understood. In the present study, TAS2R8 and TAS2R10 were endogenously expressed in the two NB cells, SK-N-BE(2)C [BE(2)C] and SH-SY5Y [SY5Y]. These receptors are more expressed in more differentiated SY5Y cells. Both TAS2Rs were up-regulated in retinoic acid-induced neuronal differentiated cells. In addition, ectopic transfection of the TAS2Rs induced neurite elongation in BE(2)C cells, and down-regulated CSCs markers (including DLK1, CD133, Notch1, and Sox2) expression, and suppressed self-renewal capacity and tumorigenicity. Furthermore, inhibition of cell migration and invasion as well as inhibition of matrix metalloproteinase activity and expression were examined by overexpression of TAS2Rs. A well-known regulator of tumor metastasis, hypoxia-inducible factor-1α (HIF-1α) and its downstream targets, vascular endothelial growth factor (VEGF) and glucose transporter-1 (GLUT-1) expression were suppressed by TAS2Rs. Taken together, our novel finding suggests that TAS2Rs targets CSCs by suppressing cancer stemness characteristics and invasion in NB, thereby highlighting new chemotherapeutic potential of bitter taste receptors.;신경모세포종은 신경으로 분화되지 않은 미성숙한 상태의 신경모세포에서 유래하는 미분화 종양으로, 영·유아기에 주로 발생한다. 악성 신경모세포종의 종양 세포는 암 줄기세포적 특성을 가지고 있는데, 이는 종양의 발생부터 전이까지 영향을 미쳐 환자의 좋지 않은 예후를 야기시키는 항암관련 연구의 주된 표적 중 하나이다. 사람의 쓴 맛 수용체(TAS2Rs)는 G단백질 연결 수용체(GPCR)군에 속하며, 혀의 미뢰를 구성하는 미각세포 말단에 위치하여 쓴 맛을 감지한다. 더불어, 이는 입을 통해 잠재적으로 유해한 독성 물질이 체내로 유입되는 것을 막아주는 1차적 방어기능을 가진다. 최근에는 일련의 연구들을 통해 이러한 쓴 맛 수용체가 미각기관이 아닌 체내의 다른 조직에도 분포되어 있음이 밝혀졌으며, 미각기능 외의 추가적인 역할에 대한 가능성이 시사되어 다양한 관련 연구가 진행되고 있다. 하지만, 쓴 맛 수용체의 항암 기능과 관련된 연구는 거의 이루어지지 않아 지금까지 밝혀진 정보가 미흡한 실정이다. 따라서 본 연구에서는 쓴 맛 수용체가 신경모세포종 종양 세포의 암 줄기세포적 특성과 암 전이적 특성에 미치는 영향을 세포주 배양을 통해 관찰하였다. 쓴 맛 수용체 중 TAS2R8과 TAS2R10의 유전자 발현을 확인한 결과, SK-N-BE(2)C와 SH-SY5Y 세포에서 모두 내재적으로 발현되었는데, 세포 분화가 더 많이 진행된 SY5Y에서 더 강하게 발현되고 있음을 확인하였다. 또한, 레티노산에 의한 BE(2)C의 신경 분화된 상태에서 그 발현 정도가 증가함으로써, 세포의 분화적 특성과의 관련성이 확인되었다. 나아가, BE(2)C 세포에 쓴 맛 수용체를 형질 주입을 통해 세포 내 과발현 상태로 유도한 뒤, 세포 자체의 형태 및 암 줄기세포적 특성의 변화를 확인하였다. 그 결과, 쓴 맛 수용체가 과발현된 세포에서 형태적으로는 신경돌기가 발생하고, 더불어 암 줄기세포 마커인 DLK1, CD133, Notch1, Sox2의 발현과 자기 재생 능력이 억제되었다. 이와 함께, 종양의 전이와 관련된 HIF-1α및 그의 후속 타겟 유전자인 VEGF, GLUT1의 발현이 낮아졌으며, 세포기질을 분해하는 MMP효소 활성 및 유전자 발현의 감소, 세포의 이동성 및 침습성 억제를 확인할 수 있었다. 이와 같이, 쓴 맛 수용체가 세포의 신경분화를 유도하고 암 줄기세포적 특성과 암 전이적 특성을 조절함으로써 종양의 발생 예방 및 치료에 긍정적으로 활용될 수 있음을 시사한다.
