dc.description.abstract본 연구는 암컷 쥐의 엽산 영양상태와 임신경험이 해마조직의 엽산 함량, 혈장 호모시스테인 농도와 인지기능에 미치는 영향을 알아보고자 하였다. Sprague-Dawley종 암컷 흰쥐를 FS×NP (엽산 보충군×임신 무경험군), FS×P (엽산 보충군×임신 경험군), FD×NP (엽산 결핍군×임신 무경험군), FD×P (엽산 결핍군×임신 경험군)으로 나누었다. 쥐의 인지기능 실험은 39주령에, 희생은 45주령에 시행되었다. 해마조직의 엽산 함량의 경우 유의적으로 엽산 영양상태와 임신경험에 영향을 받았다. 혈장 호모시스테인의 농도는 엽산 영양상태, 임신경험 및 엽산 영양상태와 임신경험간의 상호작용에 영향을 받았다. 인지 기능 테스트 결과, 모리스 수중미로 검사 결과, 3 사분면에서 출발한 probe test에서 유의적으로 유의적으로 엽산 영양상태에 영향을 받았다. 이 결과는 엽산 영양이 결핍되면 probe test에서 공간 기억이 저하되는 것을 의미한다. 감각운동 및 정서 검사 (전 인지검사)에서 불안감을 평가하기 위한 개방장 검사의 몸치장 행동 (grooming) 빈도는 엽산 영양상태와 임신 경험간의 상호작용에 유의적으로 영향을 받았다. 한편, 혈장 호모시스테인의 농도는 해마조직의 엽산 함량, 3 사분면에서 출발한 probe test의 도피대 위치 영역에 머문 기간과 유의적으로 음의 상관관계를 보였다. 결론적으로, 엽산 영양상태와 임신 경험은 정상적인 해마조직의 엽산 함량, 혈장 호모시스테인 농도 및 인지기능 (특히 공간기억)에 중요한 영향을 미치는 것으로 사료된다.;This study investigated the effect of dietary folate level and pregnancy on the hippocampus folate content, plasma homocystiene level and cognitive function in the female rat. Sprague-Dawley rats were grouped as folic acid supplemented x non-pregnant (FS × NP), folic acid supplemented x pregnant (FS × P), folic acid deficient x non-pregnant (FD × NP) and folic acid deficient x pregnant (FD × P). A cognitive test was performed at 39 weeks and rats were sacrificed at 45 weeks. The hippocampus folate content was significantly influenced by dietary folate level and pregnancy. The plasma homocyseine level was significantly influenced by dietary folate level, pregnancy, interaction between dietary folate level and pregnancy. In the morris water maze test, the result of 3rd quadrant starting probe test was significantly influenced by dietary folate level but not pregnancy. The result indicates that dietary folic acid deficient was leads to decrease spatial memory in the probe test. In the open field test, motor activity was not affected by dietary folate level and pregnancy. But, grooming frequency was significantly affected by the interaction between dietary folate level and pregnancy. In this study, it showed that the acoustic startle response and contextual fear conditioning test was not affected by dietary folate content and pregnancy. Moreover, significant negative correlations were found between plasma homocysteine level and hippocampus folate content (p=0.024) and target duration in the 3 quadrant start probe test (p=0.006). In conclusion, the impact of dietary folate level and pregnancy might be as important to the normal hippocampus folate content, plasma homocystiene level and cognitive function (especially spatial memory) in the female rat.-
