dc.description.abstractIt has been reported that traditional Korean diet pattern is plant-based and nutritionally balanced, and is containing various minerals and vitamins possessed in the diet. Although several studies have investigated the effect of plant-based single Korean food on inflammation, there is lack of evidence on the effect of whole-Korean food. In the present study, representative food groups in Korean diet were selected and generated as plant extracts based on the Fifth Korean National Health and Nutrition Examination Survey. Among the eleven plant-based groups from Korean diet, we selected vegetable (Veg) and fruit (Fruit) groups based on high antioxidant capacity as well as all mixed 10 groups (Total). The production and expression of inflammatory mediators induced by lipopolysaccharide (LPS) were examined. Nitric Oxide (NO) production was inhibited in LPS or LPS/Interferon-gamma (IFN-γ)–activated RAW 264.7 macrophages without a cytotoxic effect at concentration from 2 mg/ml to 8 mg/ml of plant extracts from Korean foods (PEK). The expressions of inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2) were down-regulated and phosphorylation of inhibitory kappaBα (p-IκBα) were significantly degraded by those three extract groups. Furthermore, plant extracts from Korean foods (PEK) suppressed inflammatory mediators, including tumor necrosis factor-α (TNF-α), interleukin-6 (IL-6), and prostaglandin E2 (PGE2) production in these cells. Total group showed higher anti-inflammatory effect compared with either Veg group or Fruit group. Taken together, these findings suggest that PEK may have beneficial effect on inflammation-mediated chronic diseases by suppressing pro-inflammatory cytokines.;본 연구에서는 한식의 식물성 추출물이 대식세포의 염증반응에 미치는 항염 효능과 그 기전에 대해 알아보았다. 전통적인 한국의 식이 유형은 식물성 성분에 기초하고 있고, 식이 내에 다양한 비타민과 무기질을 함유하고 있어 영양학적으로 균형적이라고 보고되어 왔다. 그러나, 한 가지 식품만이 아니라 여러 식품을 함께 섭취하였을 때의 염증 예방 기능을 확립하도록 수행된 연구는 거의 없는 실정이다. 본 연구에서는 5기 국민건강영양조사의 분석에 따라 한국의 식이 내에서의 1% 이상 소비되는 다소비 대표 식품군들이 선택되었고 식물성 추출물로 제조되었다. 한식의 식물성 추출물 10가지 군들 중에서, LPS로 자극된 염증성 매개물 활성과 발현을 세 군 – 10군을 모두 합한 군 (Total), 채소군 (Veg), 과일군 (Fruit) – 에서 측정하였다. 2 mg/ml 에서 8 mg/ml까지의 농도로 세포 독성이 없는 상태에서 진행된 실험에서 LPS와 LPS/IFN-γ 로 활성화된 RAW 264.7 대식세포에서의 NO 생성 저해를 확인하였다. iNOS, COX-2 발현, 그리고 IκBα의 인산화가 세 군에서 유의적으로 감소하였다. 뿐만 아니라, 한식의 식물성 추출물은 이 세포에서 TNF-α, IL-6 와 같은 염증성 사이토카인과 PGE2의 생성을 감소시켰다. Total 군은 Veg 군이나 Fruit 군에 비해 항염 효과가 더 높았다. 이로써 PEK가 염증 유발 매개물을 억제하여 염증에 관련된 여러 만성 질환들을 예방할 수 있다는 점을 시사한다.-
