dc.description.abstractThis study was performed to investigate the anti-inflammatory activities of ethanolic extract of Acanthopanax koreanum Nakai (AE) in carbon tetrachloride (CCl4)-induced liver injury in rats. Male Sprague-Dawley rats were randomly divided into 4 groups to receive the following experimental diets and CCl4 (2.0 mL/kg B.W) intraperitoneally for 8 weeks (n=8 per group): Group I, CCl4 control (CON); Group II, CCl4 + AE 1% (AE1); Group III, CCl4 + AE3% (AE3) and Group IV, CCl4 + acanthonic acid 0.037% which is equivalent to AE3% (AA). The alanine aminotransferase activities was significantly decreased by AE and AA treatment in a dose-dependent manner (p=0.0063), whereas the aspartate aminotransferase activities showed a tendency of decrease (p=0.1384). Hematoxylin-eosin and Masson’s staining showed that CCl4-induced liver damage which was remarkably improved by the administration of the high-dose AE treatment. AE3 in albumin levels were more effective than in AA (p=0.0076). Tumor necrosis factor-alpha and Interleukin 6 production were more inhibited in AE3 than in AA (p=0.0016, p=0.0002, respectively). Taken together, the results suggested that ethanolic extract of Acanthopanax koreanum Nakai had hepatoprotective effects by increasing reducing of IL-6 and TNFα production. In addition, the effect of AE was superior to single compound AA.;본 연구는 사염화탄소(CCl4)로 유도된 흰쥐의 간 손상에 대한 탐라오갈피 에탄울 추출물(AE)의 항염증 활성을 검증하기 위해 수행되었다. Sprague-Dawley계 수컷 흰쥐(200g)를 무작위로 CCl4 컨트롤(CON); 저용량군(CCl4 + AE1% (AE1)), 고용량군(CCl4 + AE3% (AE3)), 아칸토산 군(CCl4 + 0.037% acanthoic acid (AA)) 총 4 군이 되도록 나누었다. 그 중에 AE3군에 아칸토산 함량이 AA군과 등량한다. 8주 동안 위에 과 같은 실험식이 자유로게 주고 그리고 사염화탄소(2.0 mL/kg B.W.)을 복강투여을 시행하였다. 사염화탄소로 인한 간 손상 및 기능 회복 정도를 알아보기 위하여 ALT/AST를 측정하였는데, ALT에서는 CON군 비해 AE 와 AA군 유의적으로 감소 하였고, AST에서는 같은 경향보있다(p = 0.1384). H&E staining 과 Masson’s staining등 조직염색중에 사염화탄소에 의한 간 손상을 보였다. 알부민 수준의 AE3군 는 AA군 보다 더 효과적이었다(p = 0.0076). TNFαand IL-6를 측정하였다. 감소하는 경향를 보였다 (각각 p=0.0016, p=0.0002). 이상결과를 종합해분때, 분 실험결과가 탐라오갈피 에탄올 추출물이 항 염증 활동의 증가를 통해서 간 보호 효과가 있다고 제안했다. 또한, 분 실험에서 AE의 효과는 단일 화합물 AA보다 우수 하였다.-
