- 삼중 결합 물질중의 하나인 methyl propiolate의 아민 첨가 반응에서, 반응 메카니즘에 대한 체계적 연구를 수행하기 위하여 methyl propiolate와 여러 가지 이차 아민의 첨가반응 속도를 분광광도법으로 측정하였다. 6원고리의 아민들은 Broensted 상관 관계를 잘 만족시키나, 구조가 다른 아민의 경우 직선 관계로부터 벗어나는 것으로 보아 methyl propiolate에 아민의 공격이 속도 결정 단계이며 친핵체인 아민의 구조에 의해서 반응성이 크게 영향 받는다는 것을 알 수 있다. Broensted β 값이 작고, primary isotope effect나 general acid/base catalysis가 없다는 실험 결과로부터 반응은 methyl propiolate에 아민의 공격단계가 속도 결정 단계인 stepwise mechanism을 통해서 일어난다고 볼 수 있다.
Morpholine, piperidine, pyrrolidine이 methyl propiolate에 첨가된 enamine의 가수분해반응의 속도가 pH 3.0-9.5의 범위에서 측정되었다. 그 결과, 염기성 용액이나 중성 용액에서는 general acid로부터 enamine의 β-탄소로의 양성자 이동이 속도 결정 단계임을 알 수 있었고, 강산성 용액에서는 빠르게 생성된 iminium ion에 대한 물의 공격이 반응의 속도를 결정함을 알 수 있었다.; Second-order rate constants have been measured spectrophotometrically for addition reaction of various secondary amines to methyl propiolate in order to investigate the reaction mechanism. Six-membered cyclic amines show a good Broensted cerrelation while acyclic and five-membered cyclic amines exhibit significant deviations from linearity. The result of the Broensted plot implies that nuclmphilic attack of amine would be the rate-detemining step and the steric factor of amines plays an important role in the reaction system. Broensted β_(nuc) value is observed to be small and primary isotope effect and general acid/base catalysis to be absent in the present reaction system. The kinetic results have led to a conclusion that the reaction proceeds via a stepwise mechanism with a rate determining attack of amines to methyl propiolate.
Rate constants for the hydroysis of the morpholine, piperidine and pyrrolidine enamines from methyl propiolate have been investigated in pH range of 3 - 10 at 25℃. In weakly acidic and basic solutions, the rate-limiting step is proposed to be general acid catalyzed protonation of the β-carbon atom of the free enamine. In strongly acidic solutions, attack of water on iminium ion is the rate-limiting.
