dc.description.abstractThis study was performed to characterize five mulberry leaf extracts in terms of hypoglycemic activity in vitro and in vivo as well as chemical composition and to find the most appropriate dose for the selected mulberry leaf extract in healthy subjects. After obtaining five different mulberry leaf extracts by using different extraction solvents (water, 25% ethanol, 50% ethanol, 75% ethanol, or 100% ethanol) the levels of 1-deoxynojirimycin, quercetin and dietary fiber, probable functional ingredients for the hypoglycemic effect of mulberry leaf extract, were determined by High Performance Liquid Chromatography and a method of AOAC. Those of the other phytochemicals were profiled by 15T FT-ICR Mass Spectrometer. Then, hypoglycemic activities of each extract were evaluated in vitro and in vivo: α-glucosidase inhibition in an in vitro cell-free system, glucose uptake inhibition in a Caco-2 cell system, and oral glucose tolerance activity in Sprague Dawley (SD) rats. All of the five mulberry leaf extracts showed α-glucosidase inhibition activity with no significant differences between groups. In case of glucose uptake inhibition on Caco-2, [3H]-2-deoxyglucose uptake was significantly reduced in all mulberry leaf extract treated groups (P<0.05) except for the 75% EtOH group. Especially water extract group showed the strongest inhibition on [3H]-2-deoxyglucose uptake. We determined the uptake of glucose after administration of sugars with each of five mulberry leaf extract in SD rats. After overnight fasting, maltose alone or maltose with each mulberry leaf extract was administered to SD rats by gavage feeding. Although not statistically significant, the incremental area under the curve (IAUC) showed a tendency of decrease in five treatment groups compared with the control (P=0.0582). Water extract, which was selected as having the most potent hypoglycemic effect based on in vitro and in vivo studies, was administered in four different doses (0, 1.25, 2.5, or 5g) to healthy subjects aged 20-50 years (n=10/group) simultaneously with 75g maltose. Additionally to determine the appropriate intake time of mulberry extract, another group was administered 5g of the water extract 30 min before the administration of 75g maltose. Postprandial levels of glucose were measured up to 2 hours and IAUC, peak height, and peak time were calculated. The postprandial glucose levels of IAUC were dose-dependently decreased, although not statistically significant. Excursion and peak height were significantly decreased from 2.5g compared with the control group. However, there was no difference between the two groups differing in administration time.;본 연구에서는 다섯 가지 뽕잎추출물에서의 화학적 구성을 분석하고 in vitro와 in vivo 시험을 통해 혈당강하효과를 평가하여 정상인을 대상으로 하는 인체적용시험에 가장 적당한 뽕잎추출물을 선정하였다. 각각 다른 용매 (물, 25% 에탄올, 50% 에탄올, 75% 에탄올, 100% 에탄올)를 이용하여 다섯 가지 뽕잎 추출물을 추출한 후에, 혈당강하효과를 기대할 수 있는 기능성 물질인 DNJ (1-deoxynojirimycin), quercetin, 식이섬유를 고성능 액체 크로마토그래피 (HPLC)와 식품공전의 방법을 통해 분석하였다. 이 외의 추출물에 포함되어있는 다른 피토케미컬은 15T FT-ICR 질량분석기 (Mass Spectrometer)를 통해 프로파일링하였다. 그 후, 각 추출물에서의 혈당강하효과를 in vitro와 in vivo를 통해 측정 하였다 : cell-free 실험인 α-glucosidase 저해작용과 Caco-2 세포를 이용한 glucose 흡수저해 실험, 그리고 Sprague Dawley 쥐에서의 경구당부하 실험을 수행하였다. 다섯 가지 추출물 모두에게서 α-glucosidase 저해 효과가 나타났지만 추출물 간의 유의적인 차이는 보이지 않았다. Caco-2 세포를 이용한 glucose 흡수저해실험의 경우, [3H]-2-deoxyglucose의 흡수가 75%에탄올 추출물을 제외한 모든 추출물에서 유의적으로 저해되었다. 특히 물 추출물에서 가장 강한 저해효과가 나타났다. 또한, SD쥐에게 당과 각 추출물을 함께 투여한 후의 당 흡수 정도를 관찰하였다. 12시간 공복 후, maltose 또는 maltose와 각 추출물을 함께 SD 쥐에게 경구투여 한 결과, incremental area under the curve (IAUC)는 다섯 가지의 추출물 군들이 대조군 보다 감소되는 경향을 보였다(p=0.0582). In vitro 와 In vivo 연구를 통하여 가장 큰 혈당강하효과가 기대되는 추출물로 물 추출물이 선정되었으며 4가지 다른 용량 (0, 1.25, 2.5, 5g) 으로 20세에서 50세 사이의 정상인(군 당 10명)에게 75g의 maltose와 함께 섭취하게 하였다. 추가적으로 적절한 추출물 섭취시간을 알아보기 위해 다른 한 그룹은 5g의 물 추출물을 75g의 maltose를 섭취하기 30분 전에 섭취하게 하였다. 식후혈당과 인슐린은 섭취 후 2시간 동안 관찰하였고, IAUC, peak height, peak time을 계산하였다. 식후 혈당의 IAUC는 용량에 비례하여 낮아지는 경향을 보였으며 excursion 과 peak height는 추출물을 2.5g섭취한 군에서부터 대조군과 유의적인 차이를 보였다. 그러나, 섭취시간을 다르게 한 두 군간에는 차이를 보이지 않았다.-
