- 파골세포는 오래된 뼈를 분해하는 역할을 통해 뼈의 재형성과정에 중요한 역할을 한다. Receptor activator of NF-κB ligand (RANKL)은 단핵/대식세포로부터 파골 세포로 분화하는데 필수적이다. 단핵/대식세포에 RANKL 자극을 주면 세포 내에 활성산소종 (ROS, reactive oxygen species)이 생성되고, 생성된 ROS는 파골세포의 분화를 촉진시킨다. 하지만, 파골세포의 분화에서 ROS의 작용 기전은 많이 연구 되지 않았다. 본 연구에서는 단핵/대식세포에서 ROS의 작용을 밝히고자, 대식세포 계통인 RAW264.7에서 RANKL에 의해 ROS가 생성되는 것을 관찰하였으며, nicotinamide adenine dinucleotide phosphate (NADPH) oxidase 1 inhibitor인 diphenylene iodonium를 전처리하였을 때 RANKL에 의해 생성된 ROS가 감소되는 것을 확인하였다. 또한, RANKL에 의해 생성된 ROS에 의해 SHP-1이 산화되는 것을 확인하였다. RANKL자극에 의해 나타나는 spleen tyrosine kinase (SYK)과 bruton’s tyrosine kinase (BTK)의 tyrosine 인산화가 ROS에 의존적으로 조절되고, 이들 두 kinase들이 SHP-1의 기질로 작용하는 것도 확인하였다.
이상의 결과들을 종합해 볼 때, 파골세포 전구세포에 RANKL 자극에의해 ROS 생성이 증가되고, 생성된 ROS에 의해 SHP-1의 활성부위에 존재하는 cysteine이 산화되어 활성이 억제됨으로써, 파골세포의 분화를 촉진시키는 인자로 작용할 수 있다는 가능성을 확인하였다.;Osteoclasts play a critical role in bone remodeling by resorbing the bone matrix. Receptor activator of NF-κB ligand (RANKL) is necessary for osteoclast differentiation from monocyte/macrophage lineage cells. RANKL-induced generation of intracellular reactive oxygen species (ROS) modulates signaling cascades responsible for osteoclast differentiation, but how ROS affect signaling pathways is largely unknown. Here, we show that RANKL-stimulated production of ROS cause inhibition of SH2 domain-containing protein tyrosine phosphatases-1 (SHP-1) by oxidation of its catalytic cysteine residue. Treatment of cells with an inhibitor of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase prevents ROS production, SHP-1 oxidation, and osteoclastogenesis. Furthermore, RANKL-induced tyrosine phosphorylation of spleen tyrosine kinase (SYK) and bruton’s tyrosine kinase (BTK) is regulated by a redox-dependent manner. In addition, SHP-1 regulated RANKL-stimulated tyrosine phosphorylation of BTK. Taken together, our results suggest that RANKL-mediated ROS production leads to oxidation of SHP-1 that is followed by the promotion of osteoclast differentiation.
