Abstract
지방세포에서 인슐린에 의한 GLUT4의 이동기작을 이해하고자 인슐린의 신호전달과정중 ras를 경유하여 활성화된다고 알려진 kinase가 세포 내 insulin dependent glucose transporter pool인 LDM 막의 단백질을 인산화시 킬 수 있는가를 알아보았다. 지방세포로 분화된 3T3-L1 세포에 인슐린을 처리하면 GLUT4가 세포 내 vesicle로 부터 세포막으로 이동하는 것을 형광염색법으로 확인하였다. 지방세포에 인슐린을 처리한 후 ras에 대한 항체로 얻은 면역침강물에 MEK, ERK가 존재하며 인슐린에 의해 이들 kinase가 활성화됨을 관찰할 수 있었다. GLUT4가 존재함을 확인한 LDM을 항 ras 항체 면역침강물로 in vitro에서 인산화를 시켜 본 결과, 인슐린에의해 활성화된 kinase에 의해 LDM 막의 62,46,42와 33.5KDa 단백질에 인산화가 일어남이 관찰되었다. 이로써 인슐린에 의해 GLUT4가 세포막으로 이동하는 현상은 인슐린 처리시 ras의해 활성화된 kinase가 LDM 막 단백질을 인산화시키는 것과 연관될 것이라고 생각된다.;To understand a possible insulin mediated translocation mechanism (If the insulin dependent glucose transporter(GLUT4) from intracellular vesicle to plasma membrane, we studied that kinases related to ras signaling system could phosphorylate the membrane proteins of low density microsome(LDM), internal GLUT4 pool. When 373-L1 adipocytes were treated with insulin, the increase of GLUT4 translocation from intracellular vesicle to plasma membrane was observed by immunofluorescence microscopy. Activated GAP, MEK and ERK existed in the anti-ras antibody immunoprecipitates from the insulin treated adipocytes. Several species of LDM membrane proteins(62,46,42 and 33.5KDa) were phosphorylated when LDM membrane were incubated with the anti-ras immunoprecipitates from insulin treated adipocytes. These results suggest that the phosphorylation of LDM membrane proteins by ras-activated kinases may have a role in insulin regulated translocation of the GLUT4 molecules from intracellular vesicle to plasma membrane.
