The plasma membrane redox enzyme, NQO1, up-regulates mitochondrial function and has a cytoprotective role

Abstract

세포 내에서 해독작용을 하는 효소인 NQO1은 Nrf2/Keap1 pathway에 의해 조절되고 산화환원 반응의 항상성을 유지시키는 것과 연관되어 있다. 세포질이나 세포막에서 NQO1이 항산화 작용에 관여한다는 많은 연구결과가 보고되었다. 하지만 NQO1이 미토콘드리아의 기능을 조절하는 기작에 대해서는 아직 밝혀지지 않았다. 우리는 NQO1을 과발현시킨 세포 주(SH-SY5Y)를 만들어서 몇몇 미토콘드리아의 기능을 측정하였다. 일반적인 세포 배양 조건에서 실험한 결과, NQO1을 과발현시킨 세포에서 대조군에 비해 미토콘드리아의 complex 기능과 ATP 생성은 크게 증가하였지만 ROS 생성은 감소하였다. 또한 NQO1이 과발현 된 세포는 미토콘드리아 억제제인 rotenone과 antimycin A에 대하여 저항성을 가진다. 우리의 실험결과에 따르면, 대조군과 비교하였을 때 NQO1을 과발현시킨 세포가 미토콘드리아 독소에 대하여 더 잘 견디는 것으로 보여졌다. 이러한 결과들을 종합해보면 미토콘드리아의 기능이 NQO1에 의해 증가되고, NQO1이 세포를 보호할 수 있다. 더 나아가 NQO1은 신경보호 기능을 가지고 이것은 치료적인 대상이 될 수 있다.;NQO1 is a detoxifying enzyme regulated by the Nrf2/Keap1 pathway and involved in maintenance of redox homeostasis in cells. Antioxidant functions of NQO1 in the cytosol and plasma membranes have been reported in many studies. However, the mechanism how NQO1 modulate mitochondrial functions is yet to be investigated. We generated human neuroblastoma cell lines overexpressing NQO1 and measured several mitochondrial functions. Mitochondrial complex I activity and levels of ATP production were significantly increased in SH-SY5Y cells transfected with NQO1 under normal culture conditions. However, levels of ROS production were attenuated by overexpression of NQO1 under normal culture conditions. The redox enzyme made cells more resistant to mitochondrial inhibitors, rotenone and antimycin A. In conclusion, mitochondrial functions are up-regulated and protected by NQO1, suggesting that NQO1 has a neuroprotective role and can be a therapeutic target.
