Abstract
극도로 산성인, 금속 염이 풍부한 환경에서는 철 산화 능력을 가진 제련 미생물이 불 수용성의 metal sulfide를 산화시키는 역할을 한다. 광석 및 폐기물에 함유된 유기금속을 회수하기 위해서는 이러한 환경에서 우수 제련 미생물을 분리하여 산업적으로 유용하게 이용하는 것이 필요한데 대부분의 제련 미생물들 각각을 분리하고 그 존재여부를 판별하는 것은 매우 어렵다. 이러한 유용한 미생물들을 효과적으로 탐색하기 위하여 가장 유력한 제련 미생물인 철 산화 세균과 황 산화 세균의 ribosomal DNA 서열을 이용했다. 3,827 bp의 철 산화 세균의 rDNA가 PCR 반응과 그에 뒤따른 염기서열 분석에서 밝혀졌으며 이것은 1,540 ㅠㅔ의 16S rDNA와 422bp의 16S/23S ISR 그리고 1.845 ㅠㅔ으로 이루어져있었다. 황 산화 세균에서는 1,340 bp의 16S rDNA 서열이 밝혀졌다. 이와 같이 밝혀진 16S rDNA와 ISR 서열을 이용해서 다른 철 산화 세균과 황 산화 세균의 염기서열과 align함으로써 비교를 하였고, Maximum Likelihood method와 Parsimony method를 이용해서 철 산화 세균과 황 산화 세균 사이에서의 유연 관계를 밝혔다. 이 실험에서 사용된 철 산화 세균은 다른 철 산화 세균과 상당히 가까운 진화적 유연 관계를 가지고 있었으며 (G+C)% content눈 56.2%로 Thiobacillus의 다른 strain의 평균값(56.5%)에 근접하는 수치를 보였다. 황산화 세균은 다른 strain의 황 산화 세균과는 다소 먼 위치에 있지만 (G+C)% content는 다른 Thiobacillus와 비슷한 값을 보였다. 이런 수치들은 비제련 미생물의 (G+C)% content와는 특이적인 차이를 나타냈다. 황산화세균은 T. thiooxidans (M79401)로부터 분지했으며 T.thiooxudns (X72851)과 밀접한 관련이 있으며 철 산화 세균은 황 산화 세균으로부터 분지했다. 이 연구에서 이용된 철 산화 세균과 황 산화 세균의 rDNA서열은 높은 상동성 때문에 정확한 분지정도와 진화적 거리를 알기는 힘들었으나 그들의 진화적 위치를 파악하는데 도움이 되었으며 기존의 제련미생물에 대한 유연 관계와 일치했다 16S rDNA와 ISR 부위의 Thiobacillus에만 특이적인 염기서열들은 유용한 제련미생물을 탐색하는 방법을 고안하는 토대가 될 것이다.;Bioleaching is performed by the oxidation of insoluble metal sulfide by the specialized iron-oxidizing lithotrophic bacteria in acidic, metal-rich environments. And most of these precesses are carried out by genus Thiobacillus. A PCR-mediated approaches were performed for the detection of rDNA of T.ferroxidans and T.thiooxidans. Total 3,827 bp of rDNA of T.ferroxidans and 1,340 bp of rDNA of T.thiooxidans were amplified by PCR amplification. The 16S/23S intergenic spacer region of T.ferroxidans includes two distinct tRNA, HistRNA and LeutRNA. The size of the ISR was consistent with those of other Thiobacillos preciously revealed. Phylogenetic trees were defined by comparing the relationships of rDNA sequences using species of Thiobacillus by Maximum Likelihood and Parsimony Methods. As a result, T.ferroxidans (ATCC19859) falls into the group of T.ferroxidans and T.thiooxidans(AZ11) is somewhat distant from other T. thiooxidans. These two Thiobacillus species, however, have almost identical G+C content which is coincident with other Thiobacillus strain. Significantly conserved regions, longer than 30 bp, were found in several parts of rDNA only in genus Thiobacillus. These sequences might be used as probes for detecting bioleaching-associated bacteria eventually.
