- INO80 는 ATP 를 가수분해 할 때 나오는 에너지를 이용해서 nucleosome 구조를 바꿀 수 있는 swi/snf based chromatin remodeling complex 의 family 중 하나이다. INO80 은 최근 효모에서 밝혀진 것으로 여러 개의 단백질로 이루어진 complex 를 이루고 있다. 효모에서 밝혀진 INO80 은 다른 ATP-dependent chromatin remodeling complex 와 마찬가지로 전사 조절에 관계될 뿐 아니라, DNA 이중 나선 절단을 복구하는데 있어서도 직접적으로 관련 있다. 포유류 genome 은 효모 INO80 유전자의 homolog (mammalian INO80) 를 가지고 있지만 mammalian INO80 의 기능에 대해서는 알려진 바가 거의 없다.
이런 이유에서, 본 논문에서는 DNA 손상 시 일어나는 세포내 반응에서와 세포주기 조절에 있어서 human INO80 의 기능을 알아보았다. 먼저, human INO80 이 siRNA 에 의해 조절 받는 세포들을 사용하였고, 이런 세포들은 성장속도가 느린 것을 관찰할 수 있었다. 좀더 세부적으로 세포주기를 분석한 결과, human INO80 은 S 기에서 G2 기로 세포주기가 진행될 때 중요한 역할을 하고 있음을 알 수 있었다. siRNA 에 의해서 human INO80 이 조절 받는 세포에서는 DNA 가 손상 받았을 경우 S checkpoint 가 어느 정도 결점이 있음을 알 수 있었지만 G2/M checkpoint 를 발동시키거나 mitosis 를 행함에는 문제가 없음을 보았다. Affymetrix gene chip 분석을 통해서 human INO80 에 의해 조절 받는 유전자들에 대해 알아볼 수 있었고, S 기를 진행하는데 관련된 많은 유전자들이 human INO80 에 의해 조절 받고 있음을 알게 되었다. 또한 siRNA 에 의해 human INO80 의 정상적인 발현이 억제된 세포에서는 cyclin B 단백질의 발현이 감소되고, S 기에서 Cdc25C 의 조절이 비정상적으로 이뤄지고 있는 점으로 보아 human INO80 이 cyclin B 와 Cdc25C 단백질 발현을 조절함으로 S 기에서 G2 기의 진행을 조절하는 것임을 알게 되었다. 물론, human INO80 이 S 기에서 G2 기로 세포주기를 진행함에 있어서 어떻게 관여하는지는 좀더 면밀히 이해하는 것이 필요하다.
본 논문은 mammalian INO80 이 DNA 손상 시 나타나는 세포 내 반응과 세포주기 조절에 있어서 어떤 기능을 하는지를 처음으로 밝힌 것이라 할 수 있다. 이것은 새롭게 알려진 ATP-dependent chromatin remodeling complex 인 human INO80 의 기능을 이해하기 위한 중요한 단서를 제공한다고 볼 수 있다.;INO80 is a member of the family of the swi/snf-based chromatin remodeling complexes that are capable of altering nucleosome structure by using the energy of ATP hydrolysis. INO80 was recently identified from yeast and shown to exit as a multiprotein complex. Yeast INO80 is not only involved in transcriptional regulation like other ATP-dependent chromatin remodeling complexes, but it also has been shown to be directly implicated in DNA double strand break (DSB) repair. Mammalian genome possesses a homolog of yeast INO80 gene, however, little is known about the function of mammalian INO80.
In this thesis, I investigated, for the first time, the function of human INO80 focusing on its potential roles in DNA damage responses and the regulation of cell cycle progression. For this, I used HeLa cells in which hINO80 is downregulated by the stable expression of specific siRNA. First, I found that the cells downregulated for hINO80 show a retarded growth to a large extent. By detailed cell cycle analysis, I then found that hINO80 plays an important role in the progression of cell cycle from S to G2 phase. Consistent with these results, the cells downregulated for hINO80 show some defect in S checkpoint after DNA damage. However, hINO80 is not required for mitosis or G2/M DNA damage checkpoint activation. I then performed Affymetrix gene chip analysis to determine the genes that are regulated by hINO80. A number of genes implicated in S phase progression were turned out to be regulated by hINO80. Finally, I found that the cells knock downed for hINO80 exhibited a defect in Cdc25C down regulation during S phase as well as decreased cellular levels of cyclin B, suggesting that hINO80 regulates the S to G2 progression by controlling Cdc25C and cyclin B. More experiments will be necessary to understand in detail how hINO80 contributes to the S to G2 cell cycle progression.
This is the first characterization of mammalian INO80 with respect to the functions in DNA damage responses and cell cycle control. I believe that the results of this work provide important clues for understanding the functions of this newly identified ATP-dependent chromatin remodeling complex.
