우리나라산 임면수어 (Pleurogrammus azonus)의 간에 존재하는 L-ornithine aminotransferase 에 관한 연구

Abstract

우리나라산 해수어인 임면수어 (pleurogrammus azonus)의 간에 존재하는 Ornithine aminotransferase의 활성도를 측정하여 enzyme의 분포와 그 물리화학적 성질을 연구하였다. 그 결과는 다음과 같다. 1. 임면수어의 간부위를 cell fractronation하여 enzyme activity를 측정 하였더니 ornithine aminotransferase는 mitochondrial fraction에 70.1 %, cytosol fraction에 17.9%가 들어 있었으며, nuclear에는 미량 존재하였다. 2. Ammonium sulfate fractionation와 DEAE-cellulose에 의해 정제된 enzyme의 optimum pH는 7.3이었으며, Michaelis constant는 L-ornithine에 대해 5.52mM, α-Ketoglutarate에 대해 0.424mM 이었다.;The activity of ornithine aminotransferase was measured in measured in internal organs of Korean Pleuroqrammus azonus. The results are as follows. 1. As the activity of the enzyme was measured in internal organs by means of the cell fractionation, mitochondria fraction had contained OAT about 70%, and cytosol fraction had contained OAT about 18%. 2. The Ph optimum of the partially purified liver enzyme by means of ammonium sulfate fractionation and anion exchange chromatography of DEAE-cellulose was found to be approximate 7.3. The Michaelis constant of OAT for L-ornithine and α-Keto-glutarate were found to be 5.52mM and 0.42mM, respectively.
