dc.description.abstractDouble strand breaks (DSBs) of chromosomal DNA, the most destructive form of DNA damage, are generated by exposure to ionizing radiation and chemotherapeutic agents, and also naturally occur during the cellular processes such as DNA replication, meiosis and V(D)J recombination. If not properly repaired, DNA DSBs can result in loss of genetic information, gene mutations and the genome instabilities including chromosomal translocations, all of which are the major causes of cancer development. One of the early events of the cellular response to DNA DSBs is the phosphorylation of histone H2A variant X at Ser 139 in the C-terminal tail (γ-H2AX). Although the importance of γ-H2AX in DSB repair has been well documented, its exact role remains unclear. As a step towards understanding the function of γ-H2AX, I investigated the effects of chromatin alterations on γ-H2AX induction and DSB repair focusing on histone acetylation and ATP-dependent chromatin remodeling by SWI/SNF. First, I demonstrated that DNA-PK can be activated by nucleosomes and phosphorylates H2AX in the context of chromatin, and that this phosphorylation is largely stimulated by histone acetylation. I also provided evidence that this acetylation effect occurs within the cells; treatment of the cells with trichostain A, an HDAC inhibitor, increases the levels of γ-H2AX after DNA damage. Next, using the NIH 3T3 cells in which SWI/SNF can be conditionally inactivated by expressing dominant negative forms of BRG-1, the ATPase-containing core subunit of the SWI/SNF complex, I demonstrated that SWI/SNF activity is required for efficient induction of γ-H2AX following DNA damage. Further, I provided evidence that SWI/SNF contributes, probably by facilitating the phosphorylation of H2AX, to DNA repair and thereby resistance of cells to DNA damage-induced apoptosis. These results suggest that H2AX phosphorylation and DSB repair can be regulated by two general mechanisms of chromatin alterations, histone acetylation and ATP-dependent chromatin remodeling, providing the first functional link between chromatin remodeling and DNA repair in mammalian cells.;세포 내의 DNA는 doxorubicin과 같은 화학 물질 또는 방사선과 같은 외적 원인, DNA 합성이나 V(D)J recombination 등과 같은 내적 원인에 위해 손상 받을 수 있고 특히 ‘이중 나선 절단 (DSBs)’은 이중 가장 위험한 종류의 손상이다. DSB가 복구되지 않았을 경우 유전 정보가 손실되고 유전자의 손상, 돌연변이 등을 일으킬 수 있으며 이들은 암의 중요한 원인이 될 수 있다. 따라서 손상 받은 DNA의 치유가 매우 중요한데, 우선 히스톤(histone) 단백질인 H2A의 한 종류인 H2AX의 C말단에 위치한 Serine139가 인산화 되면서 치유 과정은 시작된다. DNA의 손상 치유 과정에서 H2AX의 인산화가 중요한 역할을 하고 있음이 밝혀져 있으나 아직 어떠한 기능을 하는지는 밝혀지지 않았다. 따라서 이 논문에서는 크로마틴 (chromatin)의 구조 변화가 H2AX의 인산화 과정과 DNA손상 치유에 어떤 영향을 주는지를 히스톤 아세틸화(acetylation) 및 크로마틴 리모델링(remodeling)에 중점을 두어 알아보았다. 우선 nucleosome에 의해 활성화된 DNA-PK가 H2AX를 인산화시킬 수 있고, 이는 히스톤의 아세틸화에 의해 매우 촉진됨을 알 수 있었다. 또한 히스톤 디아세틸화를 막아주는 TSA를 이용해 실제 세포 내에서의 영향을 보았을 때 역시 같은 결과를 볼 수 있었다. 다음으로 NIH3T3세포에서 DNA가 손상 받았을 경우 SWI/SNF 리모델링 복합체가 H2AX의 인산화에 매우 중요한 역할을 함을 dominant-negative기법을 이용해 알아보았다. 그리고 이러한 SWI/SNF 리모델링 복합체는 DNA손상으로 인한 세포사멸과 손상 복구에도 매우 중요한 영향을 미침을 알 수 있었다. 이러한 결과들은 H2AX의 인산화 및 DNA 손상 복구가, 크로마틴 구조를 변화시킬 수 있는 히스톤 아세틸화와 리모델링 복합체라는 두 가지 메커니즘에 의해 조절될 수 있음을 보이고 있고, 포유류에서는 처음으로 크로마틴의 구조와 DNA 손상간의 기능적인 관계를 밝혔다는 점에서 의의가 있다고 할 수 있다.-
