Abstract
Translationally controlled tumor protein (TCTP)는 히스타민 유리 활성을 가지는 단백질로 잘 알려져 있으나, 최근 Na,K-ATPase의 3^(rd) cytosolic loop에 결합하여 그 활성을 저해하는 기능도 밝혀졌다. 하지만 cytosol에 많이 존재하는 것으로 알려진 TCTP가 Na,K-ATPase와 결합하기 위해서는 membrane으로 translocation되는 과정이 필요한데 아직 그 기전은 명확히 밝혀지지 않았다. 본 실험에서는 TCTP가 인산화/탈인산화됨에 따라 cell내의 위치가 달라질 것으로 예상하고 어떤 형태의 TCTP가 Na,K-ATPase와 결합하는지 살펴보았다. TCTP의 인산화 위치로는 plk에 의해 인산화된다고 이미 알려진 46, 64번 serine residue에 초점을 맞추었고, 이 부분을 alanine으로 치환한 TCTP mutant constructs를 가지고 실험하였다. HeLa cell을 이용하여 ^(86)Rb^(+)-uptake를 한 결과, TCTP에 의해 저하된 Na,K-ATPase의 활성이 탈인산화형인 S46?64A TCTP에 의해서는 다시 회복되었으며 immunoprecipitation을 시행한 결과 TCTP의 탈인산화형인 경우 Na,K-ATPase와의 결합정도가 저하되었음을 알 수 있었다. 탈인산화형 TCTP인 경우 membrane으로 translocation이 적게 될 것이라는 예상을 확인하기 위해 membrane fractionation, confocal microscopy등을 이용해 membrane으로 이동한 TCTP constructs를 확인한 결과 탈인산화형 TCTP인 경우 membrane에 존재하는 양이 감소한 것을 알 수 있었다. 또한 TCTP의 signal pathway가 Na,K-ATPase의 저해제로 알려져 있는 ouabain과 비슷할 것으로 예상하여 ouabain처럼 EGFR, MAPK의 활성을 유도하는지 살펴보았다. 그 결과 TCTP가 과발현되면 EGFR의 활성에는 큰 변화가 없었으나 MAPK는 활성화 됨을 알 수 있었다. TCTP에 의해 유도된 MAPK의 활성은 탈인산화형인 TCTP에 의해서는 다시 회복됨을 확인할 수 있었다. 이런 여러가지 결과로 미루어 봤을 때, TCTP의 인산화/탈인산화 조절은 TCTP의 여러 기능에 영향을 미칠 것으로 예상되고 Na,K-ATPase 활성조절에 관여함을 고찰할 수 있었다.; Extracellular translationally controlled tumor protein(TCTP) was reported to have a histamine releasing activity, whereas intracellular TCTP interactas with the third cytoplasmic domain of the α subunit of Na,K-ATPase and inhibits Na,K-ATPase activity. Phosphorylation sites in TCTP by plk were mapped to two serine residues(S46,S64) and its phosphorylation was known to be involved in the translocation mechanism from nucleus to cytoplasm. In this thesis, I investigated that phosphorylated form of TCTP by plk could translocate to plasma membrane and interact with Na,K-ATPase to inhibit its activity. First of all, I performed ^(86)Rb^(+)-uptake assay to understand the role of TCTP wild-type and TCTP mutants on the regulation of Na,K-ATPase activity. The results demonstrated that dephosphorylated TCTP at both S46A and S64A mutant has a considerable recovery effect to inhibitory function of TCTP, suggesting that phosphorylated TCTP at both S46 and S64 is able to suppress the Na,K-ATPase activity. Secondly, immunoprecipitation was done to confirm the interaction between TCTP mutant and Na,K-ATPase. When double mutant(S46A and S64A) construct was overexpressed, the interaction between the TCTP and Na,K-ATPase was remarkably decreased, suggesting that phosphorylation of TCTP might be involved in its binding to Na,K-ATPase. I also investigated the intracellular distribution of TCTP by subcellular fractionation and confocal microscopy. The results make clear that dephosphorylated TCTP doesn’t bind to Na,K-ATPase as TCTP wild-type does. Finally, I attempted to characterize the downstream signal pathway of TCTP compared with ouabain. Interaction of ouabain with Na,K-ATPase causes activation of tyrosine phosphorylation of the EGFR and p42/44 MAPK. As expected, wild-type TCTP activated p42/44 MAPK, whereas dephosphorylated TCTP couldn’t activate. Therefore, I propose that the phosphorylation and dephosphorylation of TCTP may regulate the activity of Na,K-ATPase and play a role in the process of signal pathway in HeLa cell.
