- Abstract
- 암은 인간의 생명을 위협하는 가장 대표적인 질병이다. 과학기술의 발전으로 치료법이 다양해졌음에도 불구하고 암의 발생률과 암으로 인한 사망률은 계속해서 증가하고 있다. 그중 암으로 사망하는 원인 중 90%가 암의 전이이다. 따라서 암을 치료하고 극복하기 위해서는 암의 전이과정을 이해하는 것이 중요하다. 암세포가 혈관을 따라 다른 조직으로 이동하는 것을 암의 전이라 하는데 이때, 종양 미세 환경의 다양한 요소들은 암세포에게 이동 및 침윤의 기능을 부여한다. 또한 종양 미세 환경에서의 암 진행 및 이동은 세포 유형 및 3차원(3D) 매트릭스와 관련이 있다. 따라서 3D 매트릭스를 포함한 생체
모방 종양 모델을 개발하는 것은 암 환경을 이해하는 데 필수적인 역할을 할 수 있다. 본 연구에서는 인간 지방 유래 중간엽 줄기 세포(hADSC)와 유방암세포(MDA-MB-231)를 3차원적으로 공배양하여 다세포 스페로이드와 mPEG-PA와 히알루론산(HA)을 이용하여 자가조립 구조를 형성하는 3D 매트릭스를 제조하였다. 이후 3D 매트릭스 내에서 스페로이드의 세포 이동성을 관찰한 결과 종양 미세 환경에서의 침습성 및 전이성 암과 같은 양상을 관찰하였다. 이러한 결과는 생체모사 세포외 기질 개발을 통해 조직 미세 환경을 이해하며 암 전이 연구를 위한 체외 조직 모델 개발의 가능성을 제시하였다.;Cancer progression and migration in the tumor microenvironment are related to cell types and three dimensional (3D) matrices. Therefore, developing biomimetic tumor models, including co-culture systems and a tunable 3D matrix, could play an essential role in understanding the cancer environment. Here, multicellular spheroids using human adipose-derived mesenchymal stem cells (hADSCs) and breast cancer cells (MDA -MB 231) within the 3D matrix were used as a tumor microenvironment (TME) mimicking platform. The amphiphilic peptide block copolymer and hyaluronic acid (HA) formed a self-assembled structure, which provides a biocompatible 3D environment for the cells. Multicellular spheroids were formed on the optimized plate and were observed as cell migration from a spheroid within a 3D matrix, like invasive and metastatic cancer of TME. This study suggests a new 3D platform using polymer complexes and the importance of
tumor complexities, including various cell types and microenvironment.
