dc.description.abstractNucleosides are glycosylamines consisting of a nucleobase bound to a ribose or deoxyribose sugar via a β-glycosidic linkage. Examples of nucleosides include cytidine, uridine, adenosine, guanosine, thymidine and inosine. Nucleosides can be phosphorylated by specific kinases in the cell on the sugar's primary alcohol group (-CH2-OH), producing nucleotides, which are the molecular building-blocks of DNA and RNA. In medicine several nucleoside analogues are used as antiviral or anticancer agents. These compounds are activated in the cells by being converted into nucleotides, they are administered as nucleosides since charged nucleotides cannot easily cross cell membranes. MLN4924, which is in clinical trials as an anticancer agent, was stereoselectively synthesized from D-ribose via a route involving stereoselective reduction, regioselective cleavage of an isopropylidene moiety, and selective displacement of a cyclic sulfate moiety as key step. Promising biological activity in a number of therapeutic areas has been reported for 3′-C-hydroxymethyladenosine and 2′-modified nucleosides. In particular, disubstitution at the C-2′ position of nucleosides has resulted in significant activity against the hepatitis C virus (HCV). 3′-C-hydroxymethy-2′-C-substituted-purine nucleosides were designed and synthesized from D-xylose.;뉴클레오사이드는 핵염기가 리보스나 데옥시리보스 당에 -글라이코시딕 본드로 연결된 글라이코실아민이다. 뉴클레오사이드에는 싸이티딘, 유리딘, 아데노신, 구아노신, 싸이미딘과 이노신이 있다. 뉴클레오사이드는 당의 첫번째 알코올 그룹이 세포 내 특이 카이네이즈에 의해 인산화되어 DNA와 RNA의 분자 기본 단위인 뉴클레오타이드를 만든다. 약으로써 몇몇 뉴클레오사이드 유도체는 항바이러스제나 항암제로 이용된다. 전하를 띤 뉴클레오타이드는 쉽게 세포막을 통과할 수 없기 때문에 뉴클레오사이드 상태로 통과하여, 세포 내에서 뉴클레오타이드로 변형되면서 활성화된다. 항암제로써 임상시험 중인 MLN4924는 D-ribose로부터 입체선택적 환원과 아이소프로필리딘의 위치선택적 분할, key step으로써 사이클릭 황산의 선택적 대체를 거쳐 입체선택적으로 만들어진다. 3′-C-하이드록시메칠아데노신과 2′-뉴클레오사이드 치환체가 여러 분야에 걸쳐 생물학적 활성을 보인다고 보고된 바 있다. 특히 뉴클레오사이드의 2′-C-치환기가 HCV치료에 중요한 활성을 보인다. 따라서 3′-C-하이드록시메칠-2′-C-퓨린뉴클레오사이드치환체가 D-xylose로부터 디자인되고 합성되었다.-
