dc.description.abstractNa^(+),K^(+)-ATPase는 모든 진핵 세포의 세포막에 존재하며 세포의 생존에 중요한 역할을 담당한다. Na^(+),K^(+)-ATPase가 제대로 작용하지 않을 경우 다양한 질병 상태가 유발되며 따라서 Na^(+),K^(+)-ATPase는 이러한 질환의 잠재적인 치료 목표물이 될 수 있다. 혈관 평활근 세포 내 Na^(+),K^(+)-ATPase 활성이 억제되면 혈관 평활근의 수축력에 대한 영향을 통해 고혈압이 유발된다고 알려져 왔다. 우리는 최근 Translationally Controlled Tumor Protein (TCTP)가 Na^(+),K^(+)-ATPase α1 subunit의 세포질내 3번째 도메인과 상호결합하여 Na^(+),K^(+)-ATPase의 활성을 저해함을 확인하였다. 따라서, in vivo 상에서 TCTP를 과발현시키면 Na^(+),K^(+)-ATPase 활성 억제를 통해 고혈압이 유발될 것이라는 가설을 세웠다. 이 연구에서 본인은 TCTP를 과다발현하는 형질전환 마우스를 제작하였으며 이 마우스에서 생후 약 6주령 이후부터 전신 혈관성 고혈압이 유발됨을 관찰하였다. TCTP 형질전환 마우스는 대조 마우스군에 비해 혈관 수축제에 대한 혈관 평활근의 수축력이 증가하였고 혈관 이완제에 대한 혈관 평활근의 이완력은 감소하였다. 또한 혈관 평활근 세포내 Na^(+),K^(+)-ATPase 활성이 억제되어 있었으며 세포내 칼슘 이온의 mobilization이 증가되어 있었다. 이러한 결과는 TCTP 과다발현을 통해 혈관 평활근내 Na^(+),K^(+)-ATPase 활성이 억제되며 이 결과 세포내 칼슘 이온의 증가를 통해 혈관 수축력이 증가하여 형질전환 마우스에서 전신성 고혈압이 유발됨을 예측할 수 있었다. 이 연구는 in vivo상에서 Na^(+),K^(+)-ATPase의 활성의 alteration과 고혈압 유발의 연결 고리를 제공하며 TCTP가 이러한 고혈압의 치료 목표물이 될 수 있음을 시사한다. Na^(+),K^(+)-ATPase는 눈의 렌즈 세포 내 나트륨 및 칼륨 이온 농도의 항상성을 유지하는데 필수적인 역할을 담당한다. 렌즈 Na^(+),K^(+)-ATPase 활성이 억제되면 렌즈 내 나트륨 이온과 칼슘 이온 농도가 비정상적으로 올라가고 이어서 삼투압 항상성이 깨어지며 칼슘 이온에 의해 활성화되는 단백질 분해 효소의 작용에 의해 세포에 damage를 주게 되어 결과적으로 렌즈의 혼탁화, 즉 백내장 상태를 초래하게 된다. 실제 여러 가지 형태의 백내장에서 디기탈리스 유사 물질과 같은 Na^(+),K^(+)-ATPase의 저해제의 존재로 인해 렌즈의 Na^(+),K^(+)-ATPase 활성이 억제된 경우가 많이 발견되었다. 이번 연구에서 사람의 렌즈 상피 세포주에서 Na^(+),K^(+)-ATPase의 저해제로 작용하는 TCTP를 과다발현시켰을 때 Na^(+),K^(+)-ATPase 활성이 억제되었으며 이로 인해 세포내 칼슘 이온의 mobilization이 증가함을 확인하였다. 이러한 결과를 통해 TCTP가 Na^(+),K^(+)-ATPase 억제작용을 통해 백내장 유발 인자로 작용할 수 있음을 예측할 수 있다.;The Na^(+),K^(+)-ATPase exists in the plasma membrane of all eukaryotic cells and plays essential roles in cell survival. Failure of Na^(+),K^(+)-ATPase causes various pathogenic states and this enzyme can be a potential target for therapeutic treatments. Inhibition of Na^(+),K^(+)-ATPase has been implicated in the pathogenesis of hypertension via its effect on smooth muscle reactivity and myocardial contractility. We recently demonstrated that Translationally Controlled Tumor Protein (TCTP) interacts with the 3rd cytoplasmic domain of Na^(+),K^(+)-ATPase α1-subunit and acts as its cytoplasmic repressor. Therefore, it was hypothesized that repression of Na^(+),K^(+)-ATPase by over-expressed TCTP might underlie the development of hypertension. In the present study, I confirmed that transgenic mice over-expressing TCTP developed systemic arterial hypertension at about six weeks after birth. Vascular smooth muscle of TCTP-overexpressing transgenic mice also displayed augmented contractile response to vasoconstrictors and attenuated relaxation response to vasodilators. These responses seem to be caused by reduced Na^(+),K^(+)-ATPase activity and increased intracellular calcium mobilization, suggesting that the inhibition of Na^(+),K^(+)-ATPase by over-expression of TCTP is involved in the pathogenesis of hypertension. This study provides link between alteration of the sodium pump activity and hypertension in vivo, and suggests that TCTP might be a therapeutic target for the treatment of the hypertension. Also, the regulation of Na,K-ATPase activity plays a key role in maintenance of homeostasis of sodium and potassium in lens cells. The inhibition of Na,K-ATPase finally causes opacification of the lens through the abnormal increases of sodium and calcium levels, disturbances of the osmotic equilibrium and activation of proteolytic enzymes and cell damage. In various types of cataracts, the lens Na,K-ATPase acitivity was reduced by inhibitor of Na,K-ATPase such as digitalis-like compounds was found. In the current study, I confirmed that the over-expression of TCTP represses the Na,K-ATPase activity and increases cytoplasmic calcium mobilization in lens epithelial cells. These results suggest that the up-regulation of TCTP levels in lens cells might induce cataratogenesis through the inhibition of lens Na,K-ATPase and calcium mobilization.-
