dc.description.abstract방사선 사고가 발생하면 1차적으로 피해자의 인체 흡수 방사선량을 측정하는 것이 시급하며 2차적으로 이 흡수 선량 정도에 따라 치료 방침을 결정하게 된다. 이러한 인체 방사선 피폭 선량을 측정하는 것이 방사선생물계측법인데 1985년 소련 체르노빌 원폭 사고 후 그 유용성이 재평가되어 많은 자료가 축적되고 있다. 이에 국제 원자력 기구 (International Atomic Energy Agency) 에서는 각 국가마다 이러한 생물 계측 체계의 확립을 권고하고 있어 본 연구에서는 기존의 방법인 세포질 분열 차단 소핵분석법(Cytokinesis-blocked micronucleus method), Giemsa 염색법에 의한 불안정 염색체 이상(예; dicentric, ring, acentric fragment) 분석법 외에 최근에 개발되어 그 활용도가 증가하고 있는 Fluorescence in situ hybridization (FISH)법을 확립하여 방사선 생물 계측 분야에 이용코자 하였다. 또한 방사선 방어 및 방사선 항암치료시 중요한 인자중 하나인 방사선 선량율의 영향을 Giemsa 염색법과 FISH법을 사용하여 분석하여, 방사선에 의해 유발되는 불안정 및 안정 염색체 이상의 특성과 그 형성기작을 분석코자 하였으며, 최근에 인간의 노화 및 종양 유발기작과 밀접한 관계가 있는 것으로 밝혀지고 있는 telomere와 telomerase를 염색체 이상 유발과 연관하여 FISH법과 polymerase chain reaction (PCR)을 이용한 telomeric repeat amplification protocol (TRAP)을 사용하여 분석하였다. 사람 임파구 세포의 in vitro 감마선 조사(0.5, 1, 2, 3, 4 Gy)후 배양하여 유발되는 염색체 이상 빈도와 방사선량과의 관계를 세포질 분열 차단 소핵분석법, Giemsa 염색법에 의한 불안정 염색체 이상 분석법과 사람 염색체 1, 3, 4, 8번 염색체에 대한 DNA probe를 사용하여 FISH법을 시행하여 안정 염색체 이상(예; translocation)과 불안정 염색체 이상을 제1차 세포 분열 중기 임파구인 M1 세포에서 분석하였다. 세포 분열 주기는 FPG (fluorescence plus Giemsa) 방법으로 측정하였고 FISH법 시행시 염색체 이상의 분석은 새로운 분류 체계인 Protocol for Aberration Identification and Nomenclature (PAINT) system에 따라 translocation, dicentric, ring, acentric fragment 뿐만 아니라 color junction으로 분류하여 시행하였다. 방사선량에 따른 염색체 이상 유발 정도는 전형적인 linear-quadratic 양상을 보였으며 안정 염색체 이상 유발 정도가 불안정 염색체보다 1.2에서 2.6배 정도로 높게 나타났으며 DNA양이 적은 4, 8번 염색체가 상대적으로 DNA양이 많은 1, 3번에 비해 더 빈번하게 안정 및 불안정 염색체 이상 형성에 관여하였다. 이는 염색체에 따라 방사선에 대한 민감도가 다르다는 것을 시사한다. 방사선 선량율의 영향을 분석키 위해 급성 피폭의 모형으로 0.57 Gy/min의 고선량율과 만성 피폭의 경우에는 0.01 Gy/min의 저선량율을 사용하였다. 방사선 선량율의 영향은 불안정 염색체 이상인 경우는 1 Gy 이상의 선량에서 나타났으며 안정 염색체 이상인 경우는 더욱 명백하여 0.5 Gy 이상의 선량에서 약 2배 정도의 차이를 보였다. Centric ring 염색체 대 dicentric 염색체 발생 비율은 만성 피폭에 비해 급성 피폭에서 약 2배 정도로 높았으나 centric ring 염색체 발생 빈도는 만성과 급성 피폭간에 차이가 없었다. 이것은 centric ring이 dicentric 형성기작과는 달리 선량에 따라 비례하는 single hit phenomenon 기작에 의해 형성된다는 것을 제시한다. Telomere란 모든 염색체 말단에 존재하는 DNA절편으로 (TTAGGG)n 의 염기 서열을 가지고 있으며 비정상적인 DNA 재결합을 억제하고 새포내에 존재하는 mitotic clock으로서 발암 억제 등과 밀접한 관계가 있는 중요한 부분이다. 이것이 interstitial telomeric sequence로 존재 시의 특성을 규명키 위해 centromere와 인접한 위치에 telomeric repeats를 가지고 있는 Chinese hamster ovary (CHO) 와 Chinese hamster embryo (CHE) 세포를 전리 방사선과 같은 형식으로 염색체 이상을 유발하는 제한 효소를 처리하여 분석하였다. CHO 세포에서는 이러한 (TTAGGG)n repeats가 염색체의 centromere부분에 가까이 있는 반면 CHE세포에서는 염색체의 끝과 centromere부분 양쪽에 존재하는 것으로 밝혀졌다. 제한 효소 처리로 유발된 염색체이상중 22% 부터 39%가 telomeric repeat signal을 가지고 있었으며 일부 세포는 telomeric repeat sequences가 합성되었음을 나타내는 강한 signal을 보여주고 있어 이 부분이 더욱 염색체 절단이나 misrepair에 민감함을 나타낸다. Telomere를 어떤 일정 길이로 유지시켜주는 ribonuclear DNA polymerase 효소가 telomerase인데 사람의 종양 세포 대부분과 immortalized 세포주 (cell line) 에서 높은 활성이 측정되었다. 또한 종양 세포에서는 염색체의 숫자적 형태적 이상 빈도가 높으므로 telomerase 효소 활성의 발현에 따른 염색체 이상 형성과의 관계를 밝히기 위해 사람 colon carcinoma cell line (SW480)과 nonpolyposis colorectal carcinoma cell line (NA 50600, NA 59와 NA 61)을 2, 4 Gy X-선에 조사시켜 PCR-based TRAP 법으로 telomerase 활성도를 측정하였고 Giemsa 염색법과 FISH법으로 염색체 이상을 분석하였다. 모든 세포주에서 telomerase 활성이 4 Gy 에서 비조사한 세포보다 3∼7배 증가하였으며 dicentric, translocation, fragment 등의 염색체 이상 발생빈도도 증가하였다. 이와 같은 결과는 telomerase 활성이 있는 상태에서나 혹은 그 활성도가 증가한다 하여도 염색체 이상의 형성을 제어하지 못한다는 것을 나타내며 X-선 조사가 in vitro 상태에서 telomerase 활성을 유발할 수 있다는 것을 나타낸다. 이는 가장 가능성 있는 종양 치료 방법으로 제시되고 있는 telomerase 억제제의 개발과 병행하여 정상 세포나 immortalized 세포주 혹은 종양 세포에서 telomerase 활성을 유발하는 인자들에 대한 연구도 필요함을 제시한다. 본 연구 결과는 방사선 생물 계측 분야, 방사선 방어 분야, population monitoring 분야, 세포 유전학 분야, 종양 세포 치료분야의 유용한 자료로 이용될 수 있으리라 사료된다. ; Conventional chromosome aberration analysis (CA) method, cytokinesis blocked micronucleus (MN) method, and fluorescence in situ hybridization (FISH) technique were employed to improve the accuracy of dose estimation in case of radiation exposure and to increase the amount of experimental data on the relative biological effectiveness for cytogenetic damage including especially stable chromosomal aberrations. Blood lymphocytes were irradiated in vitro with gamma rays (doses of 0.5 up to 4 Gy). After stimulation with phyto- haemagglutinin (PHA), structural chromosomal aberrations were analyzed either in Giemsa stained preparations (micronuclei, dicentrics and rings) or following FISH using human whole chromosome specific DNA libraries (numbers 1, 3, 4, and 8) to detect stable translocations. The Protocol for Aberraion Identification and Nomenclature Terminology (PAINT) system which was newly developed for classifying chromosome aberrations detected by FISH technique was used to clarify all translocaions. Several advantages and disadvantages of these systems are discussed. Gamma ray irradiation increased the frequencies of micronuclei and chromosomal aberrations (dicentrics and trans- locations) in a linear-quadratic manner and the results indicate that radiation induces stable aberrations (translocations) rather than unstable aberration (dicentrics). The ratio of translocations per dicentric was found to be 1.2 to 2.6 fold. Though the DNA content of chromosomes #4 and #8 is much lower than chromosomes #1 and #3, the frequencies of dicentrics and translocations detected for #4 and 8 are much higher. To examine the dose rate effect which is one of the important factors in radiobiology and radiotheraphy, lymphocytes were irradiated with low (chronic exposure ; 0.01 Gy/min) or high (acute exposure ; 0.57 Gy/min) dose rate gamma rays. Dose rate effects began to appear at doses above 1 Gy in the frequency of dicentrics. For translocations, the reduction was more evident (about 2-fold) at dosed above 0.5 Gy. The ratio of dicentrics to centric rings observed following acute on gamma-ray irradiation was twice than that of chronic exposure. Moreover, for ring chromosomes, no difference was found between chronic and acute irradiation in the dose range of 1 to 4 Gy. These results may indicate that centric rings are formed by a single hit phenomenon in a linear manner with the dose. The interstitial telomeric sequences have been suggested to be more susceptible to chromosome breakage and rejoining. In the present study, this possibility was tested by analyzing the behaviour of intra-chromosomal telomeric sequences in restriction enzyme-treated chinese hamster ovary (CHO) and chinese hamster embryo (CHE) cells using FISH technique. Because restriction enzymes induce chromosomal aberrations in the same manner as ionizing radiations and these cell lines show large blocks of internal telomeric repeats adjacent to the centromeric regions of the chromosomes. In CHO cells, (TTAGGG)n repeats are localized only near the centromeric regions of many of the chromosomes while in CHE cells the telomeric repeat sequences are found at both the terminal and centromeric regions of the chromosomes. In CHO cells, 26% of the total aberrations induced by AluⅠ and 22% of those induced by HinfⅠwere found to be involved with internal telomeric repeat sequences. In CHE cells, which possess telomeric repeats at both the terminal and interstitial regions, 39% of the aberrations induced by AluⅠand PvuⅡ showed telomeric repeat signals. The proportion of acentric fragments with a telomeric repeat signal was higher in CHE than in CHO cells. Some of the damaged cells displayed an intense signal indicating the possible amplification of these repeat sequences are more prone to chromosome breakage and misrepair. Telomerase is a ribonucleic DNA polymerase that maintains telomere length at certain level in eukaryotes. High levels of telomerase activity have been demonstrated in immortalized cells and in the majority of human cancers. Numerical and structural chromosome aberrations are commonly observed in tumour cell lines. To study the expression and regulation of telomerase activity and its influence on the formation of chromosomal aberrations, human colon carcinoma cell line (SW480) and human nonpolyposis colorectal carcinoma cell lines (NA50600, NA59 and NA61) were exposed to 2 Gy or 4 Gy X-rays. Cells were assayed for telomerase activity using the PCR-based telomeric repeat amplification protocol (TRAP) and chromosomal aberrations were analyzed by Giemsa staining and FISH with chromosome specific DNA libraries at 24 hrs postirradiation. Increased telomerase activity was observed in all the cell lines examined; 3 to 7 fold increases were seen at 4 Gy dose. Cytogenetic rearrangements, such as dicentrics, translocations and breaks/fragments, were also increased significantly (p<0.05) following X-irradiation. The present results indicate that presence or upregulation of telomerase activity do not prevent the formation of chromosome aberrations in exposed cells and telomerase activity can be induced by X-irradiation in vitro.-
