dc.description.abstractSubcritical water may be an excellent alternative medium for extracting flavanones compared to ethanol due to its temperature-dependent selectivity, safety, efficiency of recovery, and low cost. The aim of this study was to perform qualitative and quantitative analyses of specific flavanones in Satsuma mandarin peel (SMP) using high performance liquid chromatography (HPLC) at various subcritical water extraction (SWE) conditions. Thin-layer chromatography was utilized to the identification of the main flavanones, and HPLC was used to determine the extraction yields of flavanones. The SWE conditions were as follows: temperature, 110-200℃; static extraction time, 5-20 min; and pressure, ?100 atm. Hesperidin and narirutin glycosides were the main flavanones found in Satsuma mandarin peel (SMP). The maximum yields of hesperidin and narirutin were obtained with SWE at 160oC for 10 min, and amounted to 71.36±5.93 and 11.92±0.68 mg/g of dried SMP, respectively. The extraction efficiency was higher for the SWE method than for other more conventional methods such as ethanol, methanol, and hot-water extraction.;추출용매로서 순수한 물을 사용하는 아임계수 추출법을 이용하여 제주 온주 밀감Satsuma mandarin(Citrus unshiu Marc.) 과피에 함유된 hesperidin과 narirutin을 추출하기 위한 최적조건을 설정하였고, 기존의 유기용매 추출법 및 열수 추출법과 추출효율을 비교 분석하였다. 추출 용매는 증류수를 사용하여 압력 (100 atm)은 고정하고 추출온도 (110-200℃)와 추출시간 (5-20 min)을 변화시켰고, 시료1 g과 규조토 2 g (Diatomaceous Earth, DE)을 혼합한 후 아임계수 추출장치 (DIONEX, Accelerated Solvent Extractor (ASE) 100)를 이용하여 추출하였다. 추출된 용액은TLC를 이용하여 주요 밀감 과피에 존재하는flavonone을 정성 분석하였고, 함유된 hesperidin과 narirutin 함량을 알아보기 위해 HPLC를 이용하여 정량분석 하였다. 본 연구에서의 최적 추출 조건160℃/10분 에서hesperidin은 71.93±5.93 mg/g SMP 와 narirutin: 11.92±0.68 mg/g SMP로 추출되었다. 에탄올, 메탄올, 그리고 열수 추출과 비교했을 때, 아임계 추출법이 더 높은 hesperidin, narirutin 함량이 나왔다. 이는 기존의 전통적 추출법에 비하여hesperidin, narirutin을 추출하는데 매우 적합한 방법임을 제시하고 있다.-
