dc.description.abstractGlutamate excitotoxicity, oxidative stress, and inflammatory response have all been implicated in the pathogenesis of many neurodegenerative disorders. Although numerous effective therapeutic agents for the treatment of neurodegenerative diseases have been developed over the past several decades, there are still lots of demands for developing new anti-neurotoxic agents. In part Ⅰ, to search the new neuroprotective compounds involved in neurodegenerative diseases, the novel benzylidene compounds (JC1-6) were synthesized and their biological activities were evaluated on the basis its ability to inhibit neurotoxicities. All compounds (JC1-6) showed considerable effects on free radical scavenging, the suppression of lipopolysaccharide (LPS)-induced nitric oxide (NO) generation in microglia, and the inhibition of glutamate-induced neurotoxicity. (2E)-3-(4-hydroxy-3-methoxyphenyl)phenylpro-2-en-l-one (JC3), in particular exhibited the most potent anti-neuroprotective effect in ischemia model. Based on the above results, the mechanisms of biological action of JC3 which exhibited potent anti-excitotoxic and anti-inflammatory effects were determined using cortical neurons, microglia, organotypic hippocampal culture, and in vivo ischemia model consisting of middle cerebral artery occlusion (MCAO). Compound JC3 exhibited a neuroprotective effect on oxygen-glucose deprivation (OGD) or hydrogen peroxide (H₂O₂)-induced cytotoxicity in cultured cortical cells. In addition, compound JC3 suppressed the generation of NO, pro-inflammatory cytokines, and reactive oxygen species (ROS) in LPS-induced inflammation. This study demonstrated that the action of JC3 is initiated by a set of intracellular signaling cascades that includes Janus tyrosine kinase/signal transducers and activators of transcription (Jak/STAT) and mitogen-activated protein kinase (MAPK) pathways. These results suggest that benzylideneacetophenone derivatives could be useful anti-neurotoxic agents. Lectins are proteins that exert profound effects on mammalian cell metabolism, which involves various mechanisms of cell recognition, adhesion, signal transduction, mitogenic stimulation, cytotoxic, and apoptosis. These effects were demonstrated using plant lectins that cross-link cell surface glycoproteins thereby initiating various cellular responses. The lectin, concanavalin A (Con A) have been the most extensively investigated member of the lectin family of plant proteins. In part Ⅱ, the cellular responses of Con A in neuronal cells, particularly, focused on its mitogenic effect and cytotoxicity. The process of gene regulation via protein phosphorylation by Con A was examined in cortical cells in order to investigate the mitogenic effect of Con A. Con A induced tyrosine phosphorylation and N-methyl-D-aspartate (NMDA) receptor subunit expression in cortical cells. In addition, α-amino-3-hydroxy-5-methyl-isoxazole-4-propionate (AMPA)/ kainate (KA) receptor antagonists were used to investigate whether Con A, which blocks the desensitization of the AMPA/KA receptors, exerts its mitogenic effect in cortical cells. The results showed that the AMPA/KA receptors antagonists suppressed the phosphorylation of the protein as well as the expression of NMDA receptor subunits in Con A-treated cortical cells. Furthermore, the expression of NMDA receptor subunits was modulated in a region-specific manner in microinfusion of Con A into the cerebroventricle. Slot blot assay was performed with culture to examine the presence of endogenous Con A which is known to exert a number of mitogenic and cytotoxic effects. Interestingly, the presence of Con A was strongly detected on the medium of microglia exposed to certain stress conditions such as serum-deprivation or oxygen-deprivation/reoxygenation in a time-dependent manner. To further determine the mechanisms of Con A cytotoxicity, the cultured cortical cells were treated with a high concentration of Con A. The Con A induced neurotoxicity and astrocyte activation on a dose-dependent manner. The elevation of tumor necrosis factor-α (TNF-α) by activated astrocytes led to the elevation of intracellular ROS and the activation of tyrosine kinase in cortical neuronal cells. In addition, it was demonstrated that the process of cell death in these neurons exhibited the typical features of apoptotic, including the actvation of p38/MAPK, the down-regulation of Bcl-xL, the activation of caspase-3, and the elevation of ceramide in the neurotoxic condition after Con A treatment. In addition, it was also confirmed that Con A induced neuronal toxicity and the activation of astrocytes in brain cortical areas infused with Con A. Finally, based on the previous report that the action of Con A resembles that of α-amyloid (Aα), the cellular localization of Con A was investigated in the APPsw mouse model of Alzheimer's disease (AD). Interestingly, Con A was especially detected on cerebral neurons in APPsw mice as well as in astrocytes, and ceramide was elevated in the cortex of APPsw mice, as shown in the Con A-treated cortical cells. Therefore, this study primarily demonstrated the possibility that Con A has the potential to be a useful diagnostic marker of neurodegenerative diseases such as AD. The results of this study showed that Con A induced the expression of neuronal receptor such as NMDA receptor through protein phosphorylation and that it may be released from activated microglia under deteriorative conditions. In addition, treatment with a high concentration of Con A induced neurotoxicity and astrocyte activation in cultured neuronal cells and in the cortical area of rat brain. These results suggest that Con A could be an interesting target lectin in protein expression and neurotoxicity. Furthermore, the detedtion of Con A in the culture medium and the Con A binding observed in Alzheimer's-prone mice suggest that Con A may play an important role in neurodegenerative diseases.;본 연구에서는 benzylideneacetophenone 유도체의 신경보호작용과 당에 결합하는 단백질인 Concanavalin A (Con A)의 세포독성이 신경기능에 어떻게 영향을 주는지 규명해 보았다. Part Ⅰ에서는 benzylideneacetophenone 유도체의 신경보호작용을 살펴보았다. 글루타메이트의 흥분독성, 산화적스트레스, 염증반응 등은 수많은 신경퇴행성 질환의 발병원인이다. 수 십년 동안, 많은 화합물 및 촌연물이 연구되어왔으나 아직 효능이 우수한 물질은 개발되지 못한 실정이다. 신경퇴행성 질병을 줄이기 위해 본 연구에서는 새로운 benzylidene 약물을 합성하고 신경 독성 억제에 대한 생물학적인 활성을 측정하였다. 우선, benzylideneacetophenone 유도체 (JC1-6)를 합성하고 자유라디칼 억제, 항염증효과, 항흥분독성의 효과를 측정하였다. 대부분의 약물 (JC1-6)이 자유라디칼, 소교세포에서 LPS로 유도된 NO생성 억제, 그리고 대뇌피질 세포배양에서 글루타민으로 유도된 신경 독성을 억제하는 반면, (2E)-3-(4-hydroxy-3-methoxyphenyl)phenylpro-2-en-1-one) (JC3)는 허혈성 모델에서도 강력한 신경보호 효과를 나타냈다. 이와 같은 결과를 바탕으로, 대뇌피질 세포, 소교세포, 해마 조직 배양 그리고 middle cerebral artery occlusion (MCAO)을 이용한 허혈성 모델에서 강력한 흥분독성과 염증 반응을 억제하는 효과를 나타낸 JC3에 대한 생물학적 작용의 기작을 살펴보았다. JC3는 L-글루타메이트로 유도된 세포 독성, oxygen-glucose deprivation (OGD), 혹은 H₂O₂로 인한 대뇌피질세포 독성에서 신경보호 작용을 나타냈다. 또한 LPS로 인한 염증반응에서 NO, 염증성 사이토카인, 활성산소종의 생성을 억제하였다. 이 기작은 세포내 신호 전달계인 janus tyrosine kinase/signal transducers and activators of transcription (Jak/STAT)과mitogen-activated protein kinase (MAPK) 경로에 의해 시작됨을 확인하였다. 본 연구 결과 benzylideneacetophenone유도체인 JC3가 신경독성 억제 약물로 유용할 것으로 사료된다. Part Ⅱ에서는 Con A의 세포독성이 신경기능에 미치는 영향에 대해 살펴보았다. Lectin은 인식, 접착, 신호전달, mitogenic stimulation, 세포독성, 그리고 apoptotic 기작을 수반한 포유동물의 세포대사에서 상당한 영향를 미치는 단백질이다. 다양한 lectins중에서, Con A는 식물 단백질의 lectin family의 하나로 가장 광범위하게 관찰된다. 그러므로, 본 연구에서는 Con A를 이용하여 신경세포에서 나타나는 반응 기작을 규명해보고자 하였다. 특히, Con A의 mitogenic effect와 세포 독성이 뇌세포 기능에 미치는 영향에 관해 살펴보았다. 첫 번째로, Con A에 의한 단백질 인산화를 통한 유전자 조절을 확인하기 위해, 대뇌피질 세포, 성상교세포, 그리고 소교세포를 배양하여 Con A를 처리하였다. 우선, Con A는 tyrosine 인산화와 NMDA 수용체의subunits의 발현을 유도하였다. 또한, AMPA 수용체의 길항제가 Con A로 인한 tyrosine 인산화와NMDA subunits의 발현에 영향을 미치는지를 확인하였고 AMPA 수용체 길항제는 Con A로 인한 단백질 인산화와 NMDA수용체 subunits의 발현을 억제하였다. 게다가, Con A를 직접 뇌로 주입한 후 살펴본 결과에서, NMDA 수용체 subunits의 발현은 뇌부위 특이적인 방식으로 조절됨을 확인하였다. 두 번째로, mitogenic 효과와 세포독성 효과를 갖고 있는 내인성 Con A의 존재를 확인하기 위하여, Con A가 신경세포에서 혈청 제거, 산소 제거와 산소 재개와 같은 해로운 상황 하에서 분비되는 지를 확인하였다. 흥미롭게도, Con A는 이와 같은 상황에서 대뇌피질 뉴런이나 성상교세포보다 소교세포의 배지에서 시간에 따라 상당히 증가함이 탐지되었다. 그러므로, Con A의 세포독성효과를 스트레스 상황과 같은 조건에서 확인할 수 있었다. 위의 결과를 바탕으로, 뇌에서 Con A로 인한 세포 독성 기작을 확인하기 위해서, 비교적 높은 농도의 Con A를 대뇌피질 세포에 처리하였다. Con A는 신경독성과 성상교세포의 활성을 유도하였다. 활성화된 성상교세포에 의한 TNF-α의 증가는 대뇌피질 뉴런세포에서 세포 내의 활성산소종의 생성과 tyrosine kinase의 활성의 결과를 가져왔다. 또한, Con A에 의한 신경독성은 p38/MAPK의 활성, Bcl-xL의 감소, cleaved caspase-3의 증가, 그리고 ceramide의 증가가 유도되었다. 또한, Con A를 뇌의 대뇌피질로 주입한 결과에서도, 뉴런 독성과 성상교세포의 활성을 유도한다. 마지막으로, 알츠하이머 모델인 APPsw를 이용하여 Con A의 결합유무를 확인하였다. Con A는 특이적으로 APPsw의 대뇌피질 뉴런에서 탐지되었다. 흥미롭게도, APPsw의 대뇌피질에서 성상교세포는 활성화되고 ceramide의 증가는 이전에 언급한 Con A 처리시 나타난 대뇌피질 세포에서의 결과와 일치한다. 그러므로, 본 연구는 Con A가 알츠하이머와 같은 퇴행성신경계 질환의 진단 지표로 유용할 것이라고 제시해주었다. 요약해서, Con A는 단백질 인산화와 NMDA와 같은 신경세포의 수용체의 발현을 유도하고 활성화된 소교세포로부터 방출된다. 하지만, 비교적 높은 양의 Con A는 신경세포와 동물의 대뇌피질로 주입한 결과, 모두에서 신경독성과 성상교세포의 활성이 나타내었다. 본 연구 결과는 Con A가 단백질 발현과 신경독성에 관여함을 제시하였다.-
