Cytostatic effect of specific thioredoxin reductase 1 inhibitor in mammalian cells

Abstract

The thioredoxin (Trx) and Trx reductase (TrxR) system is a key antioxidant system for maintaining reactive oxygen species (ROS) balance. High ROS levels are produced because of abnormal proliferation and survival signaling. The expression of Trx/TrxR enzymes increases to eliminate ROS. To date, several small-molecule inhibitors that target the Trx/TrxR system have been used to treat cancer cells. In this study, I characterize a new compound (225-C8) that inhibits cytosolic TrxR1 and suppresses mammalian cell proliferation. I observed that 225-C8 specifically inhibits cytosolic TrxR1 via the use of recombinant proteins. I also demonstrated that 225-C8 has cytotoxic effects on both normal and cancer cells. Lastly, I observed that cancer cells uptake 225-C8, which decreased TrxR activity. Together, these data suggest that 225-C8 specifically inhibits cytosolic TrxR1, and it affects the viability of both cancer and normal cells.;Thioredoxin system 은 항산화 시스템 중의 하나로 세포 내의 활성산소 (ROS)의 양을 조절한다. 암세포의 경우, 세포 내에서 일어나는 비정상적인 증식과 생존 경로에 의해 활성산소가 증가하고, 증가한 활성산소를 조절하기 위해 항산화 시스템 또한 증가한다. 따라서 지금까지 항암효과를 내기 위한 thioredoxin 억제제와 thioredoxin reductase 억제제가 많이 발견되고 또 개발되어 왔지만, 세포질의 TrxR1과 미토콘드리아의 TrxR2를 구분하는 억제제는 알려진 것이 없다. 따라서 본 논문에서는 우리가 발견한 화합물(225-C8)이 세포질의 TrxR1을 특이적으로 억제하는지를 확인하고, 더 나아가 세포질의 TrxR1을 억제함으로 항암효과를 볼 수 있는지 관찰하였다. 먼저, TrxR 의 재조합 단백질을 이용하여 225-C8이 세포질의 TrxR1을 특이적으로 억제하는지 확인하였고, 이 화합물을 여러 가지 암세포에 처리했을 때, 암세포에서 세포독성이 나타나는 것을 확인하였다. 또한 225-C8이 암세포로 들어가는 것을 HPLC 를 통해 확인하였고, 이 화합물이 암세포에서 TrxR 을 억제하는 것을 확인하였다. 하지만 화합물은 또한 다양한 조직에서 얻은 정상세포도 유사하게 독성을 나타냄으로서 암치료제보다는 다른 질환에 적용할 수 있기를 기대하고 있다.
