Mitochondrial reactive oxygen species in TNFα-induced necroptosis

Abstract

세포 사멸은 세포 내에서 필수적인 과정이지만 무분별하게 발생할 경우, 괴사나 자가 면역과 같은 질병을 일으킬 수 있다. 반면, 손상된 세포를 제거하지 못하면 암을 유발할 수 있기 때문에 세포 사멸을 조절하는 것은 매우 중요하다. 활성 산소는 세포 사멸의 과정에서 중요한 역할을 한다. 하지만 세포 사멸에 관여하는 활성 산소의 종류에 대해서는 아직 명확히 밝혀지지 않은 부분이 있다. 이번 연구에서는 TNFα에 의한 세포자멸괴사 과정에서 발생하는 활성 산소에 대해 조사했다. 각 종류의 활성 산소를 특이적으로 검출할 수 있는 형광 염색을 이용해 관찰한 결과, TNFα에 의한 세포자멸괴사 과정에서 미토콘드리아 과산화물음이온이 동반된다는 것을 발견했다. 특히 이러한 과산화물음이온은 RIP3에 의해 조절되며, NADPH 산화 효소가 미토콘드리아 과산화물음이온 발생의 원인이라는 것을 확인했다. 또한, 세포자멸괴사에서 발생하는 미토콘드리아 과산화물음이온이 세포자멸괴사에 직접적으로 관여하는 것이 아니라 세포자멸괴사의 결과물로서 생성되는 것임을 알 수 있었다.;The role of Reactive Oxygen Species (ROS) is crucial in the regulation of various cell death pathways. However, it is still unclear as to which specific type of ROS is involved in each pathway, leading to arguments arising about the classification of ROS in necroptosis. In particular, mitochondrial ROS has been thought to have a vital role in necroptotic pathway. Here, I show that tumor necrosis factor-α (TNFα)-induced necroptosis accompanying with mitochondrial superoxide anion. Moreover, mitochondria superoxide anion is known to be regulated by a necroptosis kinase, receptor-interacting protein 3 (RIP3). Since the level of mitochondrial superoxide anion increases in TNFα-induced necroptosis, I examined antioxidant systems for their functions in eliminating intracellular ROS and generation systems of ROS. More importantly, I have showed that the inhibition of mitochondrial superoxide anion production does not contribute to necroptosis directly. Overall, this study implicates the role of mitochondrial superoxide anion in TNFα-induced necroptosis to be the product of TNFα-induced necroptosis.
