Abstract
Furan, trihalomethane, benzene은 식품의 조리, 가공 중 생성될 수 있는 유해물질이다. 국제암연구센터에 의해 furan은 그룹 2B (발암 우려 물질), trihalomethane 중 chloroform은 그룹 2A (발암 가능 물질), benzene은 그룹 1 (인체 발암 물질)로 분류되었으며 그 위해성이 주목된 바 있는 위해 물질이다. 따라서 최근 국제적으로 식품 내 furan, benzene, trihalomethane의 섭취에 관한 관심이 증가되어왔으나, 국내 식품 내 furan, benzene, trihalomethane의 함량 분석 및 저감화 기초 연구는 미흡한 실정이다. 본 연구에서는 국내 축산물 및 그 가공식품 내 furan, benzene, trihalomethane의 동시 분석법 확립 및 333 종의 시료에 대한 정성 및 정량 분석하였다. 또한, 탄수화물을 염소와 브롬으로 처리 시 염소 이온과 브롬 이온의 노출 농도, pH, 가열 시간에 따른 trihalomethane 생성에 미치는 영향에 대해 분석하였다. 축산물 및 그 가공식품 내 furan, benzene, trihalomethane를 정성 및 정량 분석하기 위해 우유, 마요네즈, 삶은 달걀 흰자, 소고기 안심 매트릭스에서 headspace-solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) 분석법을 확립하였다. 분석법은 직선성, 검출 한계, 정량 한계, 일간 및 일내 정밀도와 정확도 항목에서 검증되었고 분석에 대한 측정불확도를 측정하였다. Furan은 0.1~8.8 %의 정밀도와 83.1~118.1 %의 정확도를 나타내었다. Chloroform은 0.1~12.0 %의 정밀도와 80.6~114.9 %의 정확도를 나타내었다. Bromodichloromethane은 0.2~8.4 %의 정밀도와 79.3~115.7 %의 정확도를 나타내었다. Dibromochloromethane은 0.1~16.5 %의 정밀도와 80.9~119.1 %의 정확도를 나타내었다. Bromoform은 0.1~14.6 %의 정밀도와 85.6~115.4 %의 정확도를 나타내었다. Benzene은 0.2~9.5 %의 정밀도와 80.8~111.6 %의 정확도를 나타내었다. 본 연구 결과 해당 분석법은 축산물 중 furan, benzene, trihalomethane 측정에 신뢰성이 있음을 보여주었다. 탄수화물 8종 (Chitin, Fructose, Galactose, Glucose, Glyceraldehyde, Maltose, Ribose, Starch)과 염소 이온 및 브롬 이온으로 이루어진 model system에서 100 ℃로 30 분, 1 시간, 2 시간 가열시켜 생성된 trihalomethane 생성 수준을 비교하였다. Trihalomethane을 정성 및 정량 분석하기 위해 headspace-gas chromatography-mass spectrometry (HS-GC-MS) 분석법을 사용하였다. 탄수화물과 염소 이온, 브롬 이온을 가열 반반 시켰을 때 chloroform과 bromoform의 생성에 미치는 영향을 확인하기 위해 염소 이온과 브롬 이온의 농도, 가열 시간, pH를 다르게 하여 반응 연구를 진행하였다. 그 결과 염소 이온의 농도를 100 μg/mL과 200 μg/mL로 연구를 진행했을 때 chloroform은 N.D. ~ 491.8 μg/L 수준으로 생성되었다. 또한, 브롬 이온의 농도는 1 μg/mL과 2 μg/mL로 연구를 진행했을 때 bromoform은 N.D. ~ 594.7 μg/L 수준으로 생성되었다. 탄수화물의 종류에 따른 chloroform과 bromoform의 생성 수준을 확인한 결과 glucose보다 다른 탄수화물의 생성 수준이 10배 정도 높았다. 염소 이온과 반응하여 chloroform을 생성한 연구의 경우 가열 시간과 pH에 따른 생성 수준 변화를 보이지 않았다. 그러나 브롬 이온과 반응하여 bromoform을 생성한 연구의 경우 pH가 높을수록 가열 시간이 길수록 더 높은 수준의 bromoform이 생성되었다. 본 연구를 토대로 수행된 가열 조건에서 chloroform과 bromoform이 다량 생성됨을 확인할 수 있었다. 또한, 상온에서 진행된 기존의 연구 보다 가열 조건에서 더 높은 수준의 chloroform과 bromoform이 생성됨을 확인하였다. 결론적으로 식품에 널리 존재하는 탄수화물은 염소 이온과 브롬 이온이 포함된 물로 가열 반응 시 상당한 수준의 트리할로메탄을 생성할 수 있는 것으로 확인되었다. ;Furan, benzene, and trihalomethane can be formed during the preparation and processing of some foods. In the present study, validation was carried out for the simultaneous determination of furan, benzene, and trihalomethane [chloroform(CF), bromodichloromethane(BDCM), dibromochloromethane(DBCM), and bromoform(BF)] in livestock products. Headspace-solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) was used to determine the linearity in the range of 0~100 μg/L, limit of detection (LOD), and limit of quantification (LOQ), inter- and intra-day precisions (RSD %), and accuracy. Results were obtained for LOD (0.05~1.71 μg/L) and LOQ (0.16~5.18 μg/L) in beef tenderloin, LOD (0.09~1.30 μg/L and LOQ (0.28~3.22 μg/L) in mayonnaise, LOD (0.07~1.30 μg/L and LOQ (0.22~3.94 μg/L) in boiled egg white and LOD (0.17~1.05 μg/L) and LOQ (0.51~3.17 μg/L) in milk. Recovery for all compounds fell within 80.0~120.0 % and all compounds showed to have R.S.D. values ≤ 20 %. The results indicated that this quantitative method was reliable for the measurement of furan, benzene, and trihalomethane in livestock products. In this study, model system consisting of 8 kinds of carbohydrates (chitin, fructose, galactose, glucose, glyceraldehyde, maltose, ribose, starch), chloride ion and bromide ion, the level of trihalomethane produced by heating at 100 ℃ for 30 minutes, 1 hour and 2 hours were compared. This study compared the changes of trihalomethane formation in heating model system consisting of carbohydrates, chloride ion and bromide ion. Chitin, fructose, galactose, glucose, glyceraldehyde, maltose, ribose and starch were used as carbohydrates, and the reaction was performed at 100℃ for 30 minutes, 1 hour and 2 hours. Chloroform and bromoform were extracted using headspace microextraction (HS) and then analyzed using gas chromatography-mass spectrometry (GC-MS). In order to confirm the effects of carbohydrates, Cl- and Br- on the formation of chloroform and bromoform, this study was conducted with different carbohydrates, different concentrations of Cl- and Br-, heating time and pH. As a result, when chloride ion concentrations were 100 μg/mL and 200 μg/mL, chloroform was produced in the range of N.D. to 491.8 μg/L. On the other hand, when bromide ion concentrations were 1 μg/mL and 2 μg/mL, bromoform was produced in the range of N.D. to 594.7 μg/L. The levels of chlorform and bromform formed with glucose were 10 times lower than those of other carbohydrates. Also, regarding the formation of chloroform, there was no change in its level according to heating time and pH. However, the formation of bromoform increased with according to pH and heating times. In conclusion, carbohydrates widely present in foods are found to be capable of producing a considerable level of trihalomethane when reacted with water containing Cl- and Br-at 100 ℃.
