Abstract
급성 폐 손상(ALI)에 대한 연구가 지속적으로 진행됨에 따라 병리 및 임상적 이해가 높아지고 있음에도 불구하고, ALI는 여전히 사망률이 매우 높은 치명적인 질병이며 효과적인 약물 치료법이 부재한 상황이다. 한편, 전사체 분석을 통해 약물의 작용 기전을 알아내는 것은 사전 지식 및 적용 데이터 범위 측면에서 강력한 장점을 가진다. 최근에는 세포의 이질성에 크게 영향을 받을 수 있는 약물 반응을 해석하기 위해 단일세포 전사체 분석 방법을 활용하는 흐름이 가속화되고 있다. 본 연구에서는 급성 폐 손상에서의 SIRT1 억제제(Sirtinol)의 효과를 전사체 수준의 변화에 집중하여 분석하였다. bulk 전사체 분석에서는 LPS에 의해 유도된 면역반응이나 폐의 구조적 손상, 특히 산화 스트레스를 Sirtinol이 완화시킴으로써 ALI에 대한 치료 효과를 가지는 것으로 보였다. 단일세포 전사체 분석에서는 세포 비율 변화, 차등 발현 유전자의 개수, SIRT1 유전자의 발현 변화에 따라 주요 세포 타입으로 호중구를 선정하였으며, 이는 Il23a, Il27, Il12b와 같은 리간드들과 Hif1a, Nfkb1, Stat3 전사인자들을 통해 염증성 단핵구와 상호작용하는 것으로 나타났다. 따라서, SIRT1 억제제는 ALI 마우스 모델에 대한 효과적인 치료제로서의 잠재력을 가진다. 이는 주로 LPS에 의한 과도한 산화 반응을 완화시키면서 작용하고, 호중구와 염증성 단핵구의 상호작용에 의해 매개된다. 이번 연구의 결과는 ALI 치료 타겟으로서의 SIRT1 유전자에 대한 새로운 관점을 제시하였으며, 나아가 ALI의 효과적인 치료법 발굴에 대한 단초가 될 것이다.;Despite the growing understanding of acute lung injury (ALI) as studies continue, ALI is still a life-threatening disease with a high mortality rate and no effective treatment available. Transcriptomic analysis, as a method to reveal the mechanism of action of drugs, has important advantages: namely, it does not require prior knowledge and wide range of data applicability. In recent years, it is becoming important to use single-cell transcriptome analysis methods to interpret drug reactions that are greatly affected by cell heterogeneity. In this study, we investigated the potential therapeutic effect of SIRT1 inhibitor (Sirtinol) in ALI, especially focusing on transcriptional-level changes. Bulk analysis showed that Sirtinol can have a therapeutic effect on ALI by relieving LPS-induced immune response, structural damage, and oxidative stress. In scRNA-seq analysis, we selected neutrophil as a major cell type among 20, based on changes in cell frequency, number of DEGs, and expressional changes of the SIRT1 gene. In addition, we found that neutrophils may interact with cytotoxic monocytes through ligands such as Il23a, Il27, and Il12b and transcription factors such as Hif1a, Nfkb1, and Stat3. We conclude that SIRT1 inhibitors have the potential as effective treatments for ALI models. Their action alleviates LPS-induced oxidative reactions and is mediated by the interaction between neutrophils and cytotoxic monocytes. This study presents a new perspective on the SIRT1 gene as a treatment target, and represents a starting point for the discovery of more effective treatments for ALI.
