Abstract
Cancer metastasis is the primary cause leading to cancer morbidity and mortality so far. Metastasis is initiated by epithelial-mesenchymal transition (EMT) through which cancer cells acquire the ability to migrate and invade. EMT is regulated by diverse factors including growth factors, receptor tyrosine kinases, and transcription factors as well as non-coding RNAs (e.g., microRNAs and long non-coding RNAs). Among them, ZEB1 is a well-known transcription factor promoting EMT in diverse epithelial cancer cells. In this dissertation, non-coding RNAs regulated by ZEB1 were identified and their functions and regulation mechanisms were investigated in terms of EMT and metastasis. ZEB1 and miR-200 form a double-negative feedback loop: ZEB1 is a transcriptional repressor of miR-200, and, in turn, miR-200 suppresses ZEB1 via direct targeting. miR-200 is well-known as an EMT suppressor; therefore, the first part of my dissertation deals with the identification of novel target genes of miR-200 and the investigation of the roles of the target gene in head and neck and lung cancer cells. Through the bioinformatic analysis, QKI, an RNA binding protein, was finally selected as a target gene for miR-200. The expression levels of QKI showed a negative correlation with those of miR-200 family members in head and neck cancer cells and lung cancer cells. Targeting of QKI by miR-200 was confirmed through a 3´-UTR assay. QKI-knockdown (KD) increased invasion, tumorigenesis, and metastasis of cancer cells, suggesting that QKI is a tumor suppressor. In the whole transcriptomic analysis using RNA sequencing, QKI-KD enriched genes related to cytokine production, inflammatory response, and interferon-gamma response, implying that QKI regulates pro-inflammatory gene signatures. Furthermore, patients with high QKI expression showed better survival rates than those with low QKI expression. Collectively, QKI is a direct target of miR-200 and promotes cancer cell invasion, cancer progression, and metastasis by forming a negative feedback loop with miR-200. The second part of my dissertation was designed to identify long non-coding RNAs (lncRNAs) regulated by ZEB1 and to elucidate their transcriptional regulation and intracellular working mechanisms. Numerous lncRNAs are abnormally expressed in cancer cells and act in diverse roles to regulate cellular processes such as chromatin remodeling, transcriptional control, epigenetic regulation, and miRNA inhibition. Through RNA sequencing, lnc-NR2F1 was identified as an upregulated lncRNA induced by ZEB1 and showed a positive correlation with ZEB1 in TCGA LUAD data. Given that lnc-NR2F1 is located at the antisense strand of the neighbor gene, NR2F1, the relationship between lnc-NR2F1 and NR2F1 was examined. NR2F1 acted as a transcription factor of lnc-NR2F1, and ZEB1 also transcriptionally upregulated NR2F1. lnc-NR2F1 increased migration, invasion, tumorigenesis, and metastasis of lung cancer cells. lnc-NR2F1 interacted with the TWIST2 genomic region and enhanced the transcription of the TWIST2 gene, suggesting that lnc-NR2F1 promotes the migratory and invasive ability of cancer cells by transcriptionally upregulating TWIST2. Moreover, high expression of lnc-NR2F1 was associated with a poor prognosis in patients with lung cancer. In conclusion, lnc-NR2F1 is induced via the ZEB1/NR2F1 axis and promotes migration, invasion, tumorigenesis, and metastasis of lung cancer cells by controlling the transcription of TWIST2. Taken together, ZEB1 is a crucial transcription factor regulating the expression of miRNAs and lncRNAs which have a significant impact on cancer progression and metastasis. ;암 전이는 암 이환률과 사망률의 대부분을 차지하는 주요 원인이며, 암세포의 침윤성과 이동성을 획득하는 과정인 EMT에 의해 개시된다. EMT는 성장 인자, 수용체 타이로신 인산화효소 및 전사인자와 같은 다양한 인자에 의해 조절되며, microRNA와 긴 비번역 RNA와 같은 비번역 RNA에 의해서도 유도된다. 그 중에서 ZEB1은 다양한 상피암세포에서 EMT를 촉진하는 전사인자로 잘 알려져 있다. 본 논문에서는 ZEB1에 의해 조절되는 비번역 RNA (microRNAs와 긴 비번역 RNAs)를 규명하고 이들이 EMT, 암 발생 및 전이과정에 미치는 영향을 연구하였다. ZEB1과 miR-200은 서로 억제하는 feedback 고리를 형성한다. 즉, ZEB1은 miR-200의 발현을 감소시키고 다시 miR-200은 ZEB1을 직접 타겟하여 억제한다. 본 논문의 첫 번째 파트에서는 miR-200의 새로운 타겟 유전자를 찾고 두경부암에서의 역할에 대해 연구했다. miRNA의 타겟 유전자 예측 사이트와 암 환자의 TCGA 데이터를 이용하여 miR-200의 새로운 타겟 유전자로서 RNA 결합 단백질인 QKI를 선정하였다. 두경부암과 폐암 세포에서 QKI의 발현과 miR-200의 발현은 음의 상관관계를 보였다. 3´-UTR assay를 통해서 miR-200이 QKI의 3´-UTR를 타겟하는 것을 확인하였다. QKI의 발현 억제는 암 세포의 침윤, 암 발생 및 전이를 촉진하는데, 이러한 결과는 QKI가 종양 억제자임을 의미한다. 세포 내에서 QKI의 조절기전을 찾기 위해 RNA sequencing을 실시하였고, 이를 통해 QKI의 억제는 사이토카인 생산, 염증 반응 및 인터페론-감마 반응과 관련된 신호전달 경로를 활성화시킨다는 것을 밝혔다. 이는 QKI가 염증 유발 유전자의 발현을 조절한다는 것을 시사한다. 또한 두경부암과 폐암환자의 TCGA 데이터에서 QKI의 발현이 낮은 환자보다 QKI의 발현이 높은 환자의 생존율이 높았다. 종합하면, 첫 번째 파트에서는 QKI가 miR-200의 타겟 유전자이며 miR-200과 negative feedback을 형성함으로써 암세포의 침윤, 암 진행 및 전이를 촉진한다는 것을 증명하였다. 두 번째 파트에서는 ZEB1에 의해 조절되는 긴 비번역 RNA(lncRNA)를 찾고 그 lncRNA의 전사 조절과 세포 내 기능 조절 기전에 대해 연구하였다. 많은 lncRNA는 암 세포에서 비정상적으로 발현되며 염색체 리모델링, 전사 조절, 후성유전학적 조절 및 miRNA 억제와 같은 다양한 세포 내의 과정을 조절한다. lncRNA sequencing을 통해서 ZEB1에 의해서 유도되는 lncRNA인 lnc-NR2F1을 선별하였는데, 폐암 환자 TCGA 데이터에서 lnc-NR2F1과 ZEB1은 양의 상관관계를 보였다. lnc-NR2F1은 염색체 상에서 이웃 유전자인 NR2F1의 antisense strand에 존재하는데, 이 둘은 프로모터 부위를 공유하므로 NR2F1이 전사인자로서 lnc-NR2F1의 발현을 조절할 것이라고 예상하였다. 실제로 NR2F1은 lnc-NR2F1의 프로모터 부위에 직접 결합하여 lnc-NR2F1의 발현을 촉진한다는 것을 확인하였다. 또한 ZEB1과 NR2F1의 관계를 찾기 위해 luciferase assay와 ChIP assay를 진행하였고 그 결과, ZEB1이 전사인자로서 NR2F1의 프로모터 부위에 결합하여 전사를 유도한다는 것을 밝혔다. lnc-NR2F1은 폐암의 이동, 침윤, 암 발생 및 전이를 증가시켰으며, TWIST2 유전자의 DNA 부위에 결합하여 전사를 촉진했다. lnc-NR2F1를 억제한 세포에 TWIST2의 발현을 다시 증가시켜주면 감소된 암세포의 이동성과 침윤성이 회복되었는데, 이러한 결과는 TWIST2가 lnc-NR2F1에 의한 암세포의 이동성과 침윤성 증가과정을 매개한다는 것을 시사한다. 더불어, 폐암 환자의 TCGA 데이터에서 lnc-NR2F1의 발현이 높은 환자는 상대적으로 예후가 좋지 않았다. 두 번째 파트를 정리해보면, lnc-NR2F1은 ZEB1과 NR2F1에 의해서 발현이 유도되며 TWIST2의 전사를 조절함으로써 폐암의 이동, 침윤, 암 발생 및 전이를 활성화시킨다. 결론적으로, 본 논문에서는 ZEB1이 암 진행과 전이에 상당한 영향을 주는 miRNAs와 lncRNA의 발현을 조절하는 중요한 전사인자라는 것을 증명하였다.
