Ten New Natural Products from Two Marine-derived Streptomyces spp.

Abstract

Enormous microorganisms and bacteria which have not yet been fully researched still exist in marine environments. The secondary metabolites they produce usually have possessed a new structure or activity. At present, 70% of the total clinical used compounds are derived from natural sources. Many new natural products have been continuously identified from Streptomyces strain which is representative actinomycete. In this paper, eight and two new natural products were isolated from two species of Streptomyces sp. respectively. Meroindenone (1) and merochlorin E-K (2- 8) were isolated from CNH-189 obtained from marine sediments from San Diego. Antarone (9) and antaroide (10) were isolated from Antarctic marine sediments. The chemical structures of ten secondary metabolites were elucidated by LC/MS, IR, CD and NMR spectroscopic data (1H, 13C, COSY, HMBC, HSQC and NOESY).;해양생태계는 생물학적 다양성이 높아 이곳에서 서식하는 미생물을 배양하여 2차대사산물의 구조를 규명하면 기존에 알려지지 않은 새로운 구조의 물질일 가능성이 높다. 또한 이러한 물질의 생리활성을 시험해 보았을 때 활성을 가지는 신규 천연물인 경우가 많다. 이러한 이유들로 해양미생물은 신약 선도 물질의 중요한 자원으로 큰 잠재성이 있다. 전체 신규 천연물의 45%가 해양 방선균 중 하나인 스트렙토마이스 (Streptomyces)로부터 얻어 졌을 정도로 스트렙토마이스 에서는 최근 여러 종류의 다양한 생리활성을 가진 신규 천연물이 발견되었다. 따라서 스트렙토마이스의 연구는 새로운 천연물 의약소재 개발을 위해 그 중요성이 상당하다. 본 연구에서는 해양 유래 방선균인 2종의 스트렙토마이스 종으로부터 10개의 신규 천연물을 분리하고 구조를 규명하였다. CNH-189 균주로부터 8종의 신규물질인meroindenone과, merochlorins E-K를 분리•정제하여 구조를 규명하고 생리활성을 시험하였다. 이 중 6개의 물질에서 항박테리아 활성이 있는 것을 확인하였고 그 중 merochlorins E, F, K에서 매우 강한 활성이 있음을 밝혔다. 또한 SCO-736 균주로부터 2종의 신규천연물인 antarone과 antaroide를 분리•정제하여 구조를 규명하고 다양한 생리활성 시험을 진행한 결과, 미백 활성 시험에서 antaroide의 미백 활성이 대조군인 알부틴에 비해 같은 농도에서 45% 높게 나타나 미백 활성이 뛰어남을 확인하였다.
