Abstract
Syndecan-2 (SDC2) is a cell surface heparan sulfate proteoglycan, which is known to play important tumorigenic role in colorectal cancer cells. SDC2 is highly expressed in colon cancer cells and participates in cancer cell adhesion, migration, and invasion. Among various functions of syndecan-2, the extracellular domain of syndecan-2 serves as a docking receptor for pro-form of matrix metalloproteinase-7 (MMP-7) secreted from colon cancer cells. Previous studies reported that a synthetic peptide (S2-P) corresponding to a portion of the human syndecan-2 sequence bound to the pro-domain of MMP-7 to inhibit colon cancer activities. Since S2-P had a relatively weak binding affinity for the MMP-7 pro-domain, in this study, the amino acid sequence of S2-P was modified to improve the anticancer potential. Based on the interaction structure of S2-P and MMP-7, four peptides were generated by replacing amino acids near Tyrosine residue 51 (Tyr 51), which is critical for the interaction. The syndecan-2-mimetic peptides harboring an Ala-to-Asp substitution at the C-terminal side of Tyr 51 (S2-D) or with an Asp-to-Phe substitution at the N-terminal side of Tyr 51 and an Ala-to-Glu substitution at the C-terminal side of Tyr 51 (S2-FE) showed improved interaction affinities for the MMP-7 pro-domain. Compared to S2-P, S2-D and S2-FE were better able to block the syndecan-2-MMP-7 interaction in vitro. In addition, both peptides inhibited the cell surface localization of MMP-7 and the cancer activities (cell migration, invasion, and colony-forming activity) of human HT29 and HCT116 colon cancer cells as well as the gelatin degradation activity of MMP-7. In vivo, both peptides inhibited the primary tumor growth and lung metastasis of CT26 mouse colon cancer cells in a xenograft mouse model. Together, these data suggest that S2-D and S2-FE could be useful therapeutic anticancer peptides for colon cancer.;Syndecan-2는 세포 표면에 존재하는 heparan sulfate proteoglycan 단백질 중 하나로 대장암 발병 시 발현이 증가되며 암의 진행에 기여한다고 알려져 있다. Syndecan-2의 다양한 기능 중, syndecan-2의 세포 외부 도메인이 대장암 세포로부터 proenzyme 형태로 분비된 matrix metalloproteinase-7(MMP-7)가 결합하는 수용체의 역할을 수행한다. 선행 연구에서는 syndecan-2 와 MMP-7이 결합하는 부위의 시퀀스와 동일한 합성 펩타이드 (S2-P) 가 MMP-7의 pro-domain에 결합하여 대장암 활성을 억제하였다고 밝혔다. 본 연구에서는, S2-P 펩타이드가 MMP-7 pro-domain에 대한 약한 결합력을 가졌다는 점을 토대로, 이를 개선한 펩타이드를 이용하여 더 높은 대장암 활성 억제 효과를 갖는 SDC2 펩타이드를 찾고자 하였다. 4가지 펩타이드는 S2-P의 아미노산 시퀀스를 바탕으로 하여 S2-P와 MMP-7 간의 결합 구조 형성에 필수적인 51번 Tyrosine 주변에 위치한 아미노산들을 교체함으로써 총 4가지의 펩타이드를 제작하였다. Syndecan-2 모방 펩타이드 중 Tyr 51의 C-terminal에 위치한 Alanine을 Aspartic acid으로 바꾼 S2-D와 Tyr 51의 N-terminal에 위치한 Aspartic acid를 Phenylalanine으로 C-terminal에 위치한 Alanine을 Glutamic acid로 바꾼 S2-FE가 MMP-7 pro-domain에 더 강한 결합력을 보였다. S2-D 와 S2-FE는 S2-P보다 효과적으로 SDC2와 MMP-7 pro-domain 간의 interaction을 저해하였다. 또한 S2-D 와 S2-FE가 HT29, HCT116 두 대장암 세포주에서 세포의 이동, 침습, 콜로니 형성 등 암 활성을 억제하였고, 대장암 세포 주변으로 분비된 MMP-7의 젤라틴 분해능력을 억제하였다. CT26 마우스 대장암 세포를 사용한 이종이식 마우스 모델에서는 두 펩타이드 모두 원발암의 성장과 폐로의 전이를 억제하였다. 결론적으로, 본 실험 결과들은 S2-D와 S2-FE 두 펩타이드가 MMP-7의 pro-domain에 대한 S2-P 보다 뛰어난 결합력으로 syndecan-2-MMP-7 결합을 저해하며, 대장암 세포의 활성과 MMP-7 효소 활성을 효과적으로 억제함을 보여준다. 따라서 본 논문은 펩타이드를 통해 SDC2와 MMP-7간의 결합을 억제함으로써 대장암에서 선택적인 MMP-7의 억제가 가능함을 보여주며, S2-D와 S2-FE가 효율적인 대장암 항암 펩타이드로서 기능할 수 있는 가능성을 시사한다.
