Abstract
글루타민은 아미노산 합성과 핵산의 합성에 아미노기를 전달하는 아미노산이다. 또한, 글루타민을 이용한 보충경로(Anaplerotic reactions)는 당이 부족하거나 저산소 환경에서도 TCA 회로가 작동할 수 있게 한다. 따라서, 빠르게 성장하는 암세포에서 글루타민은 필수적인 아미노산이다. 인간의 글루타민 수송체 중 SLC1A5는 대표적인 글루타민 수송체로 세 변이체 v1, v2, v3가 있다. v1은 간암, 폐암, 유방암 전립선암 등의 암세포에서 발현이 높아진다. 그리고, 최근 v3가 암세포의 미토콘드리아 내막에서 작동하여 미토콘드리아에 Gln을 제공한다고 밝혀졌다. 이에 본 연구에서는 SLC1A5의 세 가지 단백질 변이체를 대장균에서 발현시켜 연구하였다. 인간 유전자를 대장균에서 이형 발현시키기 위해, 서로 다른 장단점을 가진 네 가지 발현벡터 pBAD30, pColdI, pET30b, pQE80L를 선정하였다. SLC1A5의 가장 작은 변이체인 두 번째 변이체(v2)를 우선적으로 클로닝하고 발현을 최적화하였다. 온도와 유도 시점(OD600), 유도물질의 농도를 다르게 배양하여 웨스턴 블랏을 통해 최적화한 결과, pBAD30은 30℃, OD600 0.8, L-arabinose 40mM 유도일 때, pColdI은 OD600 0.8, IPTG 0.5mM, pET30b은 37℃, OD600 0.8, IPTG 0.5mM 유도일 때 가장 높은 발현을 보였다. 반면, pQE80L에서는 발현되지 않았다. 앞선 실험 결과 pET30b, pColdI, pBAD30 순으로 두꺼운 밴드를 얻었으나, [14C]Gln을 이용한 1분 수송 실험 결과 L-arabinose 40mM로 유도한 pBAD30 클로닝에서만 유효한 수송 값을 얻을 수 있었다. 최종적으로 pBAD30을 통해 인간의 SLC1A5 v2가 대장균에서 활성을 가진 단백질로 발현되었음을 확인하였다. 이에 근거하여 v1과 v3를 pBAD 시스템을 통해 발현시키고 웨스턴 블랏을 통해 확인하였다. 본 연구에서 pBAD30이 대장균에서 SLC1A5를 활성을 가진 단백질로 발현할 수 있는 벡터로 확인되었다. pBAD30을 통한 SLC1A5 변이체 발현으로 대장균에서 SLC1A5 변이체의 억제제 스크리닝이 가능할 것으로 기대된다. 구조 시뮬레이션을 통해 예측된 SLC1A5 억제제가 정말 기능하는지 확인할 수 있다. 또한, v1의 구조와 v2 및 v3의 예측 구조가 다름에도 불구하고 모두 글루타민을 수송한다는 점에서 세 변이체의 기질과 구조에 관한 연구가 필요할 것으로 보인다.;Glutamine is an amino acid that delivers amine groups to the synthesis of amino acids and nucleic acids. In addition, the anaplerotic reactions using glutamine allow TCA cycle to operate even in hypoxia or lack sugar environments. Therefore, glutamine is an essential amino acid in rapidly growing cancer cells. Among human glutamine carriers, SLC1A5 is a representative glutamine carrier and has three variants v1, v2, and v3. v1 increases expression in cancer cells such as liver cancer, lung cancer, breast cancer, and prostate cancer. In addition, it was recently found that v3 works in the mitochondrial inner membrane of cancer cells to provide Gln to mitochondrial. Therefore, in this study, three variants of SLC1A5 were expressed in E. coli for study. In order to expression the human gene in E. coli, four expression vectors pBAD30, pColdI, pET30b, and pQE80L with different advantages and disadvantages were selected. The second variant(v2), which is the smallest variant of SLC1A5, was first cloned and the expression was optimized. As a result of western blot, culturing different temperature, induction point(OD600), and concentration of inducer, pBAD30 showed the highest expression when 30℃, OD600 0.8, L-arabinose 40mM induction, pColdI showed the highest expression when OD600 0.8, IPTG 0.5 induction and pET30b showed the highest expression when 37℃, OD600 0.8, and IPTG 0.5mM induction. On the other hand, it was not expressed in pQE80L. As a result of the previous experiment, thick bands were obtained in the order of pET30b, pColdI, and pBAD30, but as a result of a 1 minute transporter uptake assay using [14C]Gln, a valid transport value was obtained only in pBAD30 cloning induced to L-arabinose 40mM. SLC1A5 v2 cloned pBAD30 was expressed as an active protein in E. coli. Based on this, v1 and v3 were expressed through a pBAD system and confirmed through a Western blot. In this study, pBAD30 was identified as a vector capable of expressing SLC1A5 as an active form in E. coli. The selection of inhibitors for the SLC1A5 strain in E. coli is expected to be possible with the expression of the SLC1A5 through pBAD30. It is possible to confirm whether the SLC1A5 inhibitor predicted through structural simulation really functions. In addition, although the structure of v1 and the predictive structure of v2 and v3 by in silico are different, studies on the substrate and structure of the three-variant bodies are needed in that they all transport glutamine.
