Abstract
The responses of anurans to past and future climatic scenarios serve a great importance to recognise the biodiversity crisis and conservation priority in East Asia.The three main goals of this research study are: (1) to elucidate the past spatial and temporal patterns of the East Asian anurans using integrative approach phylogeography, advanced taxonomy and phylogenetic comparative methods, (2) to assess and understand the factors impacting the present evolutionary patterns, and (3) to recommend insightful knowledge that applicable for the future sustainment of the biodiversity of East Asian amphibian in the perspective of molecular and ecology. This dissertation is organised under three sections: (1) improvement in molecular sampling method, (II) assessments on evolutionary factors impacting the phylogeography, systematic and evolution of the East Asian anurans, and (III) the use of molecular phylogenies for conservation biology. The three sections are connected under six independent studies, interpreted as six chapters in this dissertation. All chapters are highlighting a variation of case studies related with the phylogeography, systematic and evolution of selected species groups representing the East Asian amphibians. First section includes chapter 1, a development of a standardisation methodology for a minimally invasive tail-clipping technique on anurans larvae. This molecular sampling procedure was examined on the larvae of species model, Rana huanrenensis. The results proved that the proposed minimum 0.3 mm tail-tip length was adequate to yield a high-quality genomic DNA, safe for tadpoles at Gosner stage 34 and above, with no fatality recorded during the experiment. Second section includes chapter 2 until chapter 5, where various evolutionary factors that potentially drive the current pattern of phylogeographic, systematic and evolution of East Asian anurans are discussed. Chapter 2 explores the phylogeography of microhylid frogs, genus Kaloula spp. in East Asia. This genus is critically understudied, suffered with the absence of fossils and lack accessibility of genetic data in public database. Simultaneously, the variation of past Yellow Sea in North East Asia has resulted in intra-clade divergence for at least six amphibian species, with no elucidation whether Boreal digging frog (Kaloula borealis) population affected by the same factor. This study is the first attempt to estimate mitochondrial molecular divergence dating between Kaloula spp. members in East Asia, highlighting the widespread northern population of K. borealis that distributed in two different margins of Yellow Sea. The results proved a strong impact of past paleogeological events: the Qinghai-Tibetan Plateau (QTP) reconstruction during Miocene, followed by Pleistocene glaciations on the northern most Kaloula populations. Chapter 3 elucidates the divergence times, dispersion mechanism and colonisation process of emerging invasive species, Asian Common Black-spined toad (Duttaphrynus melanostictus). This species model is often introduced through anthropogenic factor. I hypothesised that the widespread distribution of this species in Indomalaya and Eastern mainland Asia are resulted from past paleogeological events, Quaternary glaciations. The molecular dating and population expansion patterns match with colonisation of new landscapes during Last Glacial Maxima and. Despite so, a recent dispersion events of the species were also proved to be related to pre-historic anthropogenic activities. Chapter 4 investigated the impact of climate variability and the evolution of homoplasious life history trait within the highly-radiated Holarctic true frogs (Rana spp.). The results clarified the convergence of latitudinal expansibility trait in the subarctic R. amurensis clade, provided support on the clade adaptability to cold temperature condition of the subarctic, in concordance with the estimated time of the clade extension towards the northern-most latitude in Siberia (c. 7.0 Mya, Late Miocene). Chapter 5 explores complex case relates with anurans taxa distributed over large areas that display contradicting phylogeographic structures and hidden diversity. I tested both possibilities in the taxonomically unstable members of the true toad family (Bufonidae) distributed in the Holarctic. The results demonstrated a detailed phylogeographic patterns of Holarctic bufonids following the dispersion of several genera post breakdown of the West Gondwana in the Paleocene, to the emergence of the Asiatic true toads (Bufo sp.) in North-east Asia during Miocene. The end results resolve the hidden species boundaries within the North-eastern clades of the Eastern Palearctic Bufo, and provided support on local adaptation pattern across latitudinal gradient within this population. Third section includes the final chapter 6, discussing the hidden biological invasion scenario detected from brown frogs imported to Republic of Korea through market-trade. The crypticity in spatio-temporal patterns, phenotypic and genetic sequences has been assessed through a combination of commonly-used neutral marker with computational-based integrative taxonomy and phylogeography, and morphometry. A key morphometric trait relates with the webbed-toes structure has been proposed to help future species identification. This chapter also provided insight on the implication of trading as the bio-invasion driver that potentially disrupting the evolutionary unit of native species. The conclusions achieved for all chapters provide a detailed insights into the past and present patterns of phylogeography, systematic and evolution of selected anurans models distributed in the East Asia, to serve for the major purpose of sustainability of regional and global biodiversity. ;과거와 미래의 기후 시나리오에 대한 양서류의 반응은 동아시아의 생물 다양성 위기와 보존 우선 순위를 인식하는 데 매우 중요한 지표로 사용된다. 본 연구의 구체적인 목적을 다음과 같이 세가지로 나누어 살펴 볼 수 있다: (1) 계통생물지리학, 계통분리학특론, 계통학적비교방법을 이용한 통합적 접근을 통해 동아시아 양서류의 과거 공간 및 시간적 패턴을 설명, (2) 현재의 진화 패턴에 영향을 미치는 요인들에 대한 평가 및 이해, 그리고 마지막으로 (3) 분자생물학적과 생태학적 관점에서 동아시아 양서류 생물 다양성의 미래 지속을 위한 통찰력과 정보 제공이 있다. 본 논문은 (1) 분자 샘플링 방법의 개선, (II) 동아시아 양서류의 계통지리와 분류및진화에 영향을 미치는 진화적 요인에 대한 평가, (III) 보존 생물학을 위한 분자 계통발생학 사용의 세 가지 섹션으로 구성하였다. 모든 장에서는 동아시아 양서류를 대표하는 선별된 종들의 계통학, 분류학, 진화에 관련된 다양한 사례 연구들의 중요성을 강조하고 있다. 첫 번째 섹션은 제1장, 양서류 올챙이에 대한 최소 침습적 꼬리 자르기 기술을 위한 표준화 방법론의 개발이다. 이 분자 샘플링 방법은 Rana huanrenensis의 올챙이를 통해 조사되었다. 연구 결과최소 0.3mm 꼬리끝 길이는 고품질의 유전적 정보를 산출하기에 적합하였고, Gosner stage 34이상 시기의 올챙이에게 안전하며, 실험 중 사망한 개체는 발견되지 않았다. 두 번째 섹션은 동아시아 양서류의 계통학적, 분류학적, 진화적 현재 패턴의 형성에 잠재적으로 기여하는 다양한 진화적 요인들에 대한 논의 부분으로 2장부터5장까지를 포함한다. 제2장에서는 맹꽁이과(Microhylidae) 맹꽁이속(Kaloula)의 동아시아에서의 계통지리학을 다룬다. 맹꽁이속에 대한 연구의 부족함, 화석의 부재 및 공공 데이터베이스의 유전 데이터 접근의 어려움으로 인해 해당 종의 연구의 진전에 어려움을 겪고있다. 동시에, 과거 동북아시아 황해의 변화에 따라 적어도 여섯종의 양서류 종에 대한 종내 분화가 이루어졌으며, 맹꽁이(Kaloula borealis) 개체군이 동일한 요인에 의해 영향을 받는지 여부는 규명되지 않았다. 본 연구는 동아시아의 맹꽁이속(Kaloula) 중들 사이의 미토콘드리아 분자의 분기 연대 측정을 하기 위한 첫 번째 시도이며, 황해의 두 지역의 대륙주변주에 광범위하게 서식하는 맹꽁이의 북방 개체군을 강조하고 있다. 본 연구의 결과는 연구 대상 종에 대한 과거 지질학적 사건들의 심대한 영향을 입증했다. 중신세(Miocene)시대의 칭하이-티베트고원(Qinghai-Tibetan Plateau) 복원과 이후에 최북단에 위치한 맹꽁이 개체군에서의 빙하기가 이를 뒷받침한다. 제3장에서는 새로운 침입종인 아시아검은안경두꺼비(Duttaphrynus melanostictus)의 분기 시점, 분산 메커니즘 및 군체 형성 과정을 설명한다. 이 종 은 종종 인위 적인 요인을 통해 도입된다. 본 연구는 이 종의 광범위한 분포가 동양구와 동 아시아 대륙에서 과거의 고생물학적인 사건인 제 4기 빙하로부터 비롯되었다고 가정하고있다. 분자 연대 측정과 인구 확장 패턴은 마지막빙하기의 최성기(Last Glacial Maxima) 기간의 새로운 환경으로의 이주의 식민지화와 일치한다. 최근에 일어난 이중의 분산은 선사시대 인류 활동과도 관련이 있는 것으로 입증되었다. 제4장에서는 다양한 형태로 진화한 전북구 참개구리속종(Rana spp.) 내에서 기후 변동성과 진화적 상동(成因相同; homoplasy)인 생활사 특징의 진화가 끼치는 영향에 대해 다루고있다. 연구 결과는 아북극 아무르산개구리(Rana amurensis) 분기군에서 위도 팽창 특성의 수렴을 명확히 밝히고, 분기군의 아북극에서의 저온 적응성을 뒷받침하며, 이는 시베리아 최북단 위도(c. 7.0 Mya, 후기 중신세)를 향한 분기군의 확장 추정 시기와 일치한다. 제5장에서는 넓은 영역에 걸쳐 분포된 양서류 분류군에 관련된 복잡한 사례의 고찰을 통해 모순된 지리학적 구조와 숨겨진 다양성을 보여주며 다양한 가설을 제시한다. 본 연구는 전북구(Holartic)에 분포하는 두꺼비과(Bufonidae)중 분류학적으로 불안정한 종에서 두 가지 가능성을 제실험한다. 그 결과는 팔레오세(Paleocene)동안의 서쪽에 위치한 곤드와나(Gondwana) 대륙의 분해와 중신세동안의 동북아시아의 두꺼비 출현으로 발생한 여러 종의 분산 후에 일어난 아시아 두꺼비속 개구리의 상세한 지리학적 패턴을 보여주었다. 최종 결과는 Eastern Palearctic Bufo의 북동 분기군 내의 숨겨진 종 경계를 해결하고, 이 개체군 내의 위도 (기울기 따른)에 걸친 지역적 적응 패턴에 대한 증거를 보여준다. 세 번째 섹션의제 6장 에서는 시장 무역을 통해 한국에 수입된 산개구리류에서 검출된 잘 알려지지 않은 생물학적 침략 시나리오를 논의하였다. 산개구리 보호색의 시공간적 패턴, 표현형 및 유전자 염기서열 분석에 일반적으로 사용되는 중성 유전자 마커와 계산 기반 통합 분류법 및 계통학, 형태 측정법의 조합을 통해 평가하였다. 또한, 향후의 종 식별을 위해 물갈퀴-발톱 구조와 관련된 주요 형태학적 특성을 제시하였다. 이 장은 무역이 토착종의 진화 단위에 지장을 줄 가능성이 있는 생물침략적인 영향에 대한 통찰력을 보여준다. 모든 장에서 다루고 있는 최종적인 결론은 지역적뿐만이 아닌 전 세계적으로 생물 다양성의 지속 가능성의 주요 목적에 기여하기 위해 동아시아에 분포하는 선별된 양서류 의 계통학, 분류학과 진화의 과거와 현재 패턴에 대한 상세한 통찰력을 제공한다.
