Abstract
As globalization, trade, and overseas travel have continued to increase, humans gained great economic benefits, but were affected by various invasive species. Invasive species migrated to non-primary regions due to human activity, and the distribution of invasive species expanded more rapidly due to global warming. Many invasive species have a major impact on human life for reasons including spreading of vector-borne diseases, crop damage, and economic loss from ecological disturbances. In many countries, studies to control losses due to the invasive species, as well as population genetic studies, are being conducted. This study attempted to confirm the genetic structure and diversity of invasive invertebrates living in Korea. In this study, I tried to understand the genetic status of invasive species inhabiting Korea by confirming their genetic structure and diversity. In the first chapter, I introduced general information about the background of this study and about the target species. The target species, Aedes albopictus and Armadillidium vulgare were selected as subjects of this study because they are considered an introduced species. In the second chapter, I have dealt with the basic content of the population genetic analysis used in this study. In the third chapter, I compared A. albopictus population to confirm the introduction of the species into Korea. The Asian tiger mosquito is an invasive species that mediates disease and has recently expanded its distribution rapidly due to human activities. For this reason, a number of studies are being conducted in various countries to confirm the introduction of A. albopictus, but there is insufficient information on the Korean population. In this study, I analyzed the genetic diversity and structure of two mitochondrial genes and 16 microsatellites in mosquitoes inhabiting Korea, Japan, and Laos, and as a result, I found the possibility that the Korean population is a native population. The results of mitochondrial genetic analysis showed that the genetic diversity of the Korean and Japanese populations was quite low, but the Lao population did not share haplotypes with the former two groups, indicating that the Korean A. albopictus was not derived from the Lao population. In the microsatellite results, all of the Korean, Japanese, and Lao populations showed high diversity, and even though there were mixed groups, it was confirmed that there are independent clusters unique to Korea. It means that there is a genotype unique to Korea. Continuous international trade and overseas travel require constant monitoring of species that can cause mosquito-borne diseases. In addition, genetic studies will be useful basis for understanding the genetic status of vectors and managing vector diseases. In the fourth chapter, The Korean A. vulgare population was introduced and confirmed the genetic evidence forming the species complex. A. vulgare is distributed worldwide and due to their low mobility in nature, the expansion of their distribution have been depended on human activities. Although many studies have been conducted on A. vulgare as a model species for studies related to whether mitochondrial genetic diversity is affected by Wolbachia infection, no genetic studies at the Korea scale, and information on the genetic substance was inadequate. In this study, the mitochondrial analysis of the Korean, Russian, and Japanese populations showed very low diversity, but it was confirmed that a group was formed regardless of geographic factors. Such results suggest that the introduced population formed species complex. In addition, in the tree results, it was confirmed that they are placed in the same cluster as multiple overseas groups. Microsatellite results show relatively higher diversity than mitochondria, this is believed to be due to A. vulgare's unique reproduction system. As active global trade progresses, crops, compost, etc. are carried and invading such as A. vulgare, which can harm agriculture and landscape architecture, are increasing, and continuous monitoring is required. Genetic studies can also help to understand the genetic status of the species that can cause economic loss. In this study, identified the genetic diversity and structure of invasive invertebrates living in Korea and found out whether they were introduced or not. Such studies can be used not only to help understand the evolutionary history of organisms, but also as a basis for preemptive control strategies for managing invasive species. ;세계화가 증대되고 무역과 해외여행이 지속적으로 증가하면서 인간은 큰 경제적 이득을 얻게 되었지만 다양한 침입종의 영향을 받게 되었다. 침입종들은 인간의 활동으로 인해 원집단이 아닌 지역으로 이동하게 되고 기후변화의 영향으로 분포를 더욱 빠르게 확장하게 되었다. 많은 침입종들은 질병 매개, 농작물 피해, 생태교란으로 인한 경제적 손실 같은 이유로 인간의 삶에 큰 영향을 주고 있다. 여러 국가에서는 침입종으로 인한 손실을 방제하기 위한 연구들이 수행되고 있고 집단유전학적 연구 또한 이루어지고 있다. 본 연구에서는 한국에 서식하는 침입종 무척추동물을 대상으로 유전적 구조와 다양성을 확인하여 이들의 유전적 상태를 파악하고자 하였다. 첫 번째 장에서는 본 연구의 배경이 되는 일반적인 내용들을 다루었고 선정한 대상 종들의 정보를 제공하였다. 두 번째 장에서는 본 연구에서 사용되는 집단유전학적 분석에 관한 기본적인 내용들을 다루었다. 세 번째 장에서는 흰줄숲모기 집단을 비교하여 한국 흰줄숲모기의 도입 여부를 확인하였다. 흰줄숲모기는 질병을 매개하는 침입종으로 인간의 활동으로 인해 최근 급속도로 분포를 확장하였다. 이러한 이유 때문에 여러 국가에서는 흰줄숲모기의 도입 여부를 확인하기 위한 다수의 연구가 수행 중이지만 한국의 집단에 대한 정보는 미미했다. 이 연구에서는 한국, 일본, 라오스의 흰줄숲모기를 미토콘드리아 유전자 2개, 16개의 마이크로새틀라이트로 유전적 다양성과 구조를 분석하였고 그 결과 한국 집단이 원집단이라는 가능성을 발견했다. 미토콘드리아 유전자 분석 결과에서는 라오스를 제외한 한국, 일본 집단의 유전적 다양성은 상당히 낮은 것으로 나타났지만 라오스 집단이 다른 집단과 haplotype을 공유하지 않아 한국의 흰줄숲모기가 라오스 집단에서 유래된 것은 아니라고 보였다. 마이크로새틀라이트 결과에서는 한국, 일본, 라오스 집단 모두 높은 다양성을 보여주었고 혼합된 집단도 존재하지만 한국만의 독립적인 클러스터가 있는 것을 확인하여 한국만이 가지는 유전형이 있는 것을 밝혀냈다. 지속되는 국제 무역과 해외여행으로 인해 모기 매개 질병을 유발할 수 있는 종에 대한 모니터링이 꾸준히 필요하다. 또한 유전학적 연구는 매개체의 유전적 상태를 파악하여 매개 질병관리의 유용한 기초자료가 될 것이다. 네 번째 장에서는 한국의 공벌레 집단이 도입되었고 species complex를 형성하고 있다는 유전적 증거를 확인했다. 공벌레는 전 세계적인 분포를 나타내고 있으며 자연적으로는 낮은 이동성을 가져 인간의 활동에 의해 분포를 확장한 것으로 알려져 있다. 미토콘드리아 유전자 다양성이 Wolbachia 감염의 영향을 받는지에 관련된 연구의 모델 종으로 많은 연구가 이루어져 있지만 한국에서의 유전적 연구는 전무하여 유전적 실체에 대한 정보가 부족했다. 이 연구에서는 한국, 러시아, 일본 집단의 미토콘드리아 분석에서는 아주 낮은 다양성이 나타났지만 지리적 요소와 관계없이 차이가 큰 그룹을 형성하는 것을 확인했다. 이와 같은 결과는 도입된 집단이 species complex를 형성하게 된 것을 유추하게 해준다. 또한 tree 결과에서는 여러 해외 집단과 동일한 클러스터에 놓이는 것을 확인할 수 있었다. 마이크로새틀라이트 결과에서는 미토콘드리아보다 비교적 높은 다양성이 나타났는데 이는 공벌레의 독특한 번식 시스템에 의한 것으로 보인다. 활발한 해외무역이 진행됨에 따라 농작물, 퇴비 등이 운반되어 농업, 조경업에 해를 끼칠 수 있는 공벌레와 같은 침입종이 증가하여 이에 대한 지속적인 모니터링이 필요하다. 또한 유전학적 연구는 유전적 상태를 파악해 경제적 손실을 줄 수 있는 종에 대한 이해를 도와줄 수 있다. 본 연구에서는 한국에 서식하는 침입종 무척추동물들의 유전적 다양성과 구조를 파악하여 도입 여부와 유전적 상태에 대해 밝혀냈다 이와 같은 연구는 생물의 진화적 역사에 대한 이해를 도울 뿐만 아니라 침입종을 관리하기 위한 선제적 방제 전략의 기초적 자료로 활용될 수 있다.
