Abstract
Inflammatory response is an essential cause of ischemia stroke damage. Especially the key signaling molecules, reactive oxygen species (ROS), that play a pivotal role in the induction disorder of inflammatory response. Under the inflammatory conditions, oxidative stress leads to infiltrate inflammatory cells across the blood brain barrier (BBB). Several studies suggested that immune responses induced by T cells and macrophages have essential roles in the ischemic stroke. However, there are no study that Peroxiredoxin 1 (Prdx1) antioxidant protein can initiate the antioxidant response and inhibit the inflammatory brain damage during ischemic stroke. Here I demonstrated that deficiency of Prdx1 induced more severe damage at the ischemic stroke. To explore the role of Prdx1 in the development of acute ischemic brain injury, I subjected Prdx1 deficient and WT mice to 1hr of middle cerebral artery occlusion (MCAO) followed by 24hrs of reperfusion Prdx1 deficient exhibited increased neurological deficits and enlarged brain infarction after ischemia reperfusion compared with Prdx1 WT mice. To identify relationship between Prdx1 and immune responses, I analyzed the immune cells population of the immune organs and brain by using FACS. I set up the single cell sorting and gating strategy of immune organs and brain cells. As the results, identified that infiltrated macrophages increased at the stroke (MCAO) induced severe damage of brain. Whereas found that, in the Prdx1 deficient mice, infiltrated macrophages were decreased at the Prdx1 deficient mice compared with Prdx1 WT mice. Next, I examined the mRNA expression of brain inflammatory cytokines by qPCR. As the typical T cell cytokines were shown increased tendency at the MCAO brain and more increased at the Prdx1 deficient mice. However, the expression of the activated inflammatory macrophages cytokines had decrease pattern at the Prdx1 deficient mice compared to Prdx1 WT mice which is correlated with the results of infiltrated macrophages population. In conclusion, I confirmed Prdx1 has protective role in the ischemic stroke. The regulation of Prdx1 at the immune responses shown different pattern that infiltrated macrophages were decreased at the severe damage brain compared to previous study. Expression pattern of inflammatory cytokines also supports this results. I suggest that Prdx1 affects the cell death mechanisms of the brain. Although more research are needed to elucidate a Prdx1 mediated protective mechanism against stroke, prdx1 can be an essential target protein to reduce damage of ischemic stroke.;뇌졸중은 우리나라에서 사망원인 2위를 차지하는 질병으로 뇌에 혈액을 공급하는 혈관이 막히거나 터져서 뇌 손상이 오고 그에 따른 신체장애가 나타나는 질환이다. 주로 노인 질환으로 인식되었지만 최근 젊은 세대에서도 흔히 발병하는 질환으로 알려져 있다. 뇌에 적절한 산소 공급이 이뤄지지 않아 후유증이 많이 남는 만큼 뇌졸중에 있어 혈액순환장애로 인한 활성산소의 증가는 세포구조를 손상 시키는 데에 중요한 역할을 할 것이라 여겨진다. 퍼록시레독신-1 은 이 활성산소 중에서 H2O2를 제거해주는 대표적인 항산화 효소로 알려져 있다. 아직까지 뇌졸중 마우스 모델에서 퍼록시레독신-1 이 어떤 역할을 하는지 정확하게 밝혀진 바가 없다. 본 연구에서는 뇌졸중을 유발 시켰을 때 퍼록시레독신 - 1 이 발현되지 않는 마우스는 정상적인 마우스보다 뇌 손상이 증가되는 것을 확인할 수 있었다. 이후에 퍼록시레독신-1이 없는 경우 어떠한 변화가 있는지 확인하기 위해 면역시스템을 확인해 보았다. 면역시스템을 조절하는 대표적인 면역조직인 비장과 림프절, 혈액 내의 면역 세포의 분포를 확인해 본 결과 뇌졸중을 유발하지 않았을 때 퍼록시레독신-1 의 유무에 관계없이 면역세포의 분포에 큰 변화가 없음을 알 수 있었다. 또한 뇌졸중을 유발한 마우스 모델에서도 퍼록시레독신-1은 면역세포의 분포에 크게 영향을 미치지 않음을 알 수 있었다. 반면에 뇌에 존재하는 면역 세포들을 확인해본 결과 뇌졸중이 유발되지 않은 상태에서는 다른 면역 조직과 유사하게 면역세포분포의 변화가 없었지만 뇌졸중이 유발된 경우 뇌로 유입된 면역세포인 마크로파지의 분포가 더 많은 손상을 입은 퍼록시레독신-1이 없는 마우스의 뇌에서 대조군 마우스에 비해 유의적으로 감소하는 것을 확인할 수 있었다. 면역세포가 분비하는 사이토카인의 경우에도 뇌졸중이 유발되었을 때 뇌에서 더 증가되는데 퍼록시레독신-1 이 없는 마우스의 경우에 증가가 억제되는 경향을 보였다. 이 연구결과를 통해 퍼록시레독신-1 이 제거가 되었을 때 뇌세포 사멸에 더 많은 영향을 끼칠 것이라 유추할 수 있었고 추후에 뇌세포 손상을 저해하는 뇌졸중 치료 연구에 중요한 타깃 유전자가 될 수 있을 것으로 생각한다.
