- Abstract
- Since novel coronavirus was reported in Wuhan, Hubei province, China, in December 2019, it has spread to the entire world. World Health Organization (WHO) declared Coronavirus Disease 2019 (COVID-19) outbreak as a pandemic on 11 March 2020. COVID-19 is a severe acute respiratory syndrome caused by SARS-CoV-2 infection and a threatening of an unprecedented scale to public health. Various efforts are being made to find effective therapies to curb the spread of this virus. The spike (S) protein of SARSCoV-2, especially, plays an important role in facilitating fusion and entry of the virus into target cells, and the angiotensin-converting enzyme 2 (ACE2) has been known as a cellular receptor for the S protein. Thus, spike protein is considered a target for developing antibodies and vaccines, and blocking the interaction between ACE2 and spike protein by antibodies is expected to be a viable therapeutic approach to this infectious disease. Here, we isolated a panel of antibodies targeting SARS-CoV-2 S protein from a human scFv phage library by using phage display technology. Several scFv were identified through enzyme-linked immunosorbent assay (ELISA). The neutralization activity of these clones for hACE2-S protein interaction was evaluated by competition ELISA. I suggest that these antibodies have potential for therapeutic agents targeting SARS-CoV-2.;2019 년 12 월 중국 후베이성 우한에서 신종 코로나바이러스가 보고된 이후 전 세계로 확산됐다. 또한 세계보건기구(WHO)가 2020 년 3 월 11 일 코로나바이러스 감염증-2019(COVID-19)를 유행병으로 선포했다. COVID-19 는 SARS-CoV-2 감염으로 인한 심각한 급성 호흡기 증후군으로 공중 보건에 전례 없는 규모의 위협이 되고 있다. 현재 이 바이러스의 확산을 억제하는 효과적인 치료법을 찾기 위한 다양한 노력이 이루어지고 있다. 특히 SARS-CoV-2 의 스파이크(S) 단백질은 대상 세포에 바이러스의 융합과 진입을 촉진하는 데 중요한 역할을 하며, Angiotensin-Converting Enzyme 2(ACE2)는 S단백질의 세포수용체로 알려져 있다. 따라서 스파이크 단백질은 항체와 백신 개발의 대상으로 여겨지고 있으며, 항체에 의한 ACE2 와 스파이크 단백질 간의 상호작용을 차단하는 것이 이에 대한 실행 가능한 치료적 접근이 될 것으로 기대된다. 여기서는 파지 디스플레이(Phage display)기술을 이용해 인간 scFv 파지 라이브러리에서 SARS-CoV-2 S 단백질을 대상으로 한 항체 패널을 분리하고 효소면역측정법(ELISA) 실험을 통해 여러 scFv 를 식별했다. ACE2-S 단백질 상호작용을 억제하는 클론의 중화활성은 Competitive ELISA 의해 평가되었다. 우리는 이 항체들이 SARSCoV-2 치료제의 잠재력을 가지고 있다고 제안한다.
