Abstract
뇌 외측고삐핵(lateral habenula, LHb)은 스트레스 및 우울증 조절의 핵심 뇌영역으로 주목받아 오면서 관련 기전으로서 LHb의 흥분성 신경전달의 장기 지속에 대한 전기생리학적 조사가 활발히 진행되어 왔다. 현재까지 LHb 영역의 흥분성 신경전달의 장기 지속적 억제(long-term depression, LTD)는 대사형 글루탐산 수용체 1형(metabotropic glutamate receptor1, mGlu receptor 1)의 활성화에 의해 매개되고, 우울증 유사 행동이 이러한 조절 메커니즘의 변조에 의해 야기된다는 것이 보고되었다(Park et al., 2017a; Valentinova & Mameli, 2016). 본 연구는 LHb 시냅스를 구성하는 글루탐산 수용체 중 잠재적인 칼슘 통로 역할을 할것으로 짐작되는 NMDA 수용체(NMDA receptor)와 칼슘 투과성 AMPA 수용체(Ca2+-permeable AMPA receptor)의 활성화가 흥분성 신경전달의 장기 억제를 조절 할 것인가에 관해 전기생리학적 조사를 진행하였다. 급성 및 만성 스트레스에 의해 야기되는 시냅스 가소성의 변조 메커니즘에서 LHb 영역에서 흥분성 신경전달의 장기 억제에 대한 잠재적인 생리적 기능을 확인하였다. LHb 영역으로 들어오는 흥분성 입력 신경다발(stria medullaris fiber, SM fiber)에 저빈도 전기 자극(low-frequency stimulation, LFS;150 μs, 1 Hz x 900 회)의 전달은 안정적으로LTD를 유도하였고, 이러한 흥분성 신경전달의 장기 억제는 NMDA 수용체 길항제 D-AP5에 의해서 억제되고, NMDA 하위 수용체(receptor subtypes) 특이적 조절을 보이는 것을 확인하였다. 뿐만 아니라, 시냅스 NMDA 수용체 길항제 MK-801과 글루탐산 수송통로(glutamate transporter) 길항제 DL-TBOA의 처리를 통해서 LHb 시냅스 영역과 시냅스 외 영역에서 LTD가 유도될 수 있다는 사실을 확인하였다. 이러한 LTD는 NR2A 포함 NMDA 수용체 길항제 NVP-AAM 077과 NR2B 포함 NMDA 수용체 길항제 Ro25-6981에 완벽하게 억제됨에 따라, NR2A포함 NMDA 수용체와 NR2B포함 NMDA 수용체가 시냅스 및 시냅스 외 영역에서 흥분성 신경전달의 장기 지속 조절에 관여할 것으로 보여진다. CP-AMPA 수용체는 코카인 노출에 의한 시냅스 가소성의 뱡향이 변조되는 메커니즘에 관여할 것으로 알려져 왔지만(Maroteaux & Mameli, 2012), LTD의 조절에 대한 보고는 없었다. 본 연구 결과는 CP-AMPA 수용체의 활성화는 NMDA 수용체형LTD를 조절하는데 핵심적인 조절자 역할을 하는 전기생리학적 근거를 제시하였다. 실제로, 저농도 Mg2+으로 변환된 인공뇌척수액에서 NMDA 수용체의 선활성화는 CP-AMPA 수용체를 억제하더라도 LTD 발생에 영향없이 안정적으로 유도되었다. 뿐만 아니라, NMDA에 의해서 유도되는 화학적 LTD는 CP-AMPA 수용체 길항제 NASPM에 의해서 완벽하게 억제됨에 따라 CP-AMPA 수용체의 활성화가 NMDA 수용체형 LTD를 유도하는데 필수적으로 보인다. CP-AMPA 수용체의 선행적 활성화는 시냅스 후 세포막의 탈분극을 야기하고, 결과적으로LTD를 유도하는데 영향을 준다는 선행 연구가 알려져 있다(Sanderson et al., 2016). 본 연구 결과에서도 NMDA 수용체형 LTD의 유도는 CP-AMPA 수용체의 활성화가 반드시 요구되고, 조절자로서 핵심적인 역할을 한다는 사실을 확인하였다. 시냅스 및 시냅스 외 영역의 LTD에 대한 생리적 기능을 확인하기 위해서 급성 및 만성 스트레스의 노출에 의한 영향을 조사하였다. 이전에 보고된 연구와 마찬가지로(Park et al., 2017b), 급성 및 만성 스트레스 모델의 LHb에서는 LFS에 의해 유도되는 LTD는 완벽하게 억제되는 것을 확인하였다. 그러나, DL-TBOA에 의해 시냅스 외 영역으로의 글루탐산 과잉을 야기하면 시냅스 외 영역에서 유도되는 LTD는 영향을 받지 않거나, 오히려 증가되어 유도된다는 사실을 확인하였다. 우리는 LTD 억제가 스트레스에 의해 증가된 코티코스테론(corticosterone) 호르몬에 의해 야기된 것을 확인하였다. 정상 쥐의 LHb에서 코티코스테론 약물처리는 LTD의 억제를 야기하지만, 시냅스 외 영역에서 기록되는 LTD에는 영향을 주지 않는 것을 확인하였다. 뿐만 아니라, 이러한 코티코스테론에 의해 유도되는 LTD 소거는 미네랄로코티코이드 수용체(mineralocorticoid receptor)의 활성화에 의해 조절되었다. 즉 급성 및 만성 스트레스에 의해 야기되는 시냅스 가소성의 변조는 스트레스 호르몬에 의해 야기되고, 미네랄로코티코이드 수용체의 활성화에 의해 조절되었다. 현재까지 LHb의 흥분성 신경전달의 장기 지속적 억제는 모노아민 시스템을 직접적 혹은 간접적 제어를 통해서 신경세포 활성의 과도하게 증가되는 것을 통제하고, 항상성을 유지하는데 중요한 기능을 할 것으로 보고되어 왔다(Valentinova and Mameli, 2016). 뿐만 아니라, 우울증 동물모델의 LHb에서는 이러한 LTD가 억제되고, LTP가 증가되는 시냅스 가소성의 변조 메커니즘이 무기력과 같은 우울증 유사 행동을 야기할 것으로 관련이 있을 것으로 보고되었다(Park et al., 2017a; Park et al., 2017b). 그러나, 본 연구 결과들에 따르면, LHb시냅스 및 시냅스 외 영역에서는 NMDA 수용체형 LTD가 유도되었다. 뿐만 아니라, 급성 및 만성 스트레스를 경험한 동물의 LHb에서 시냅스 영역의 LTD는 억제되지만, 시냅스 외 영역의 LTD는 영향을 받지않거나, 오히려 증폭되는 것을 고려하면 LHb에서 이러한 시냅스 영역의 LTD 소거 현상에 대한 보상 메커니즘이 존재한다는 사실을 확인하였다. 즉, 스트레스에 의해 야기되는 LHb의 흥분성 신경세포의 과활성화는 우울증 유사 행동을 야기하게 되는 LTD 소거를 일으키지만, 증가된 시냅스 외 영역의 LTD를 유도함으로써 잠재적으로 모노아민 시스템의 과도한 억제를 막는 안전 장치로서 역할을 할 것으로 판단된다. 본 연구 결과는 LHb 시냅스 외 영역에서의 LTD 연구가 반복적인 스트레스에 의해 우울증으로 진행되는 병인 기전을 이해하는데 중요한 세포적 모델이 될 수 있음을 시사한다. ;As the lateral habenula (LHb) has been reported to play a critical role in the regulation of stress and depression, considerable electrophysiological research has been conducted on the long-term depression (LTD) of glutamatergic transmission. However, only a few studies have reported that LTD of glutamatergic transmission in the LHb is mediated by the activation of Gp1 mGluR and depressive-like behavior (learned helplessness) is caused by the alteration of modulation mechanism (Park et al., 2017; Valentinova & Mameli, 2016). The purpose of this study is to electrophysiologically investigate whether the activation of NMDAR and CP-AMPAR, which is probable to play a role as potential Ca2+ sources, among glutamate receptors in the LHb plays a functional role in LTD of the glutamatergic transmission. This study also confirms a potential physiological function in long-lasting suppression of the glutamatergic transmission in the LHb by exploring alteration mechanism of synaptic plasticity caused by acute and chronic stress. The results showed that LFS delivery (150 μs, 1 Hz, 900 pulse) to SM fiber into the LHb induced robust-LTD. Long-lasting suppression of the excitatory transmission was induced by D-AP5, NMDAR antagonist and LTD of the excitatory transmission was specifically modulated by NMDAR subtypes of NR2A and NR2B. In addition, the results revealed that treatment with MK 801 and DL-TBOA induced LTD in both synaptic and extrasynaptic regions in the LHb. It is highly probable that NR2A and NR2B-containing NMDAR, which are expressed in the synaptic and extrasynaptic regions, may involve in LTD of the excitatory transmission in that treatment with NVP-AAM 077 and Ro25-6981 fully blocked LTD. Although it has been suggested that CP-AMPAR is involved in altering the direction of synaptic plasticity from LTD to LTP (Maroteaux & Mameli, 2012), there have been few functional studies regarding LTD modulation. However, the results of this study revealed that the activation of CP-AMPAR was found to be a key regulator in modulating NMDAR-dependent LTD. Even under the condition of a suppression of CP-AMPAR, the preactivation of NMDAR was steadily induced by low Mg2+ modified aCSF without affecting the induction of LTD. In addition, CP-AMPAR activation was found to be essentially required for the induction of NMDAR-dependent LTD by confirming that LTD chemically induced by NMDA was fully blocked by CP-AMPAR antagonist, NASPM. This may be due to the fact that Ca2+ influx triggered by the preactivation of CP-AMPAR leads to NMDAR activation and LTD induction (Sanderson et al., 2016). In other words, the results showed that CP-AMPAR activation was found to induce NMDAR-dependent LTD and play a critical role in modulating NMDAR-dependent LTD as a regulator. To confirm physiological function of synaptic and extrasynaptic LTD in the LHb, this study explored the effects of acute and chronic stress exposure. As previously suggested (Park et al., 2017), the results revealed that LFS-induced LTD was completely extinct in the LHb of rats exposed to acute and chronic stress. However, stress did not affect extrasynaptic LTD when DL-TBOA caused glutamate spillover in the extrasynaptic region. Rather, it facilitated the induction of extrasynaptic LTD. Also, it was found that CORT hormone increased by stress led to LTD extinction. For normal rats, treatment with CORT caused LTD to be extinct but did not have an effect on extrasynaptic LTD. LTD extinction which was induced by CORT was modulated by MR activation as well. Taken together, these findings suggest that the alteration of synaptic plasticity triggered by acute and chronic stress is caused by CORT hormone and regulated by MR activation. According to electrophysiological researches conducted over many years, it has been suggested that long-lasting suppression of glutamatergic transmission in the LHb plays critical roles in preventing monoamine system from the excessive increase in neuronal activity through the direct or indirect regulation (Park et al., 2017; Valentinova & Mameli, 2016). In addition, several studies alteration of synaptic plasticity involved in LTD inhibition and LTP facilitation is likely to cause depressive-like behavior such as learned helplessness (Park et al., 2017; Park et al., 2017). However, the results of this study indicated that NMDAR-dependent LTD was induced in the synaptic and extrasynaptic regions in the LHb. Considering the result that stress did not affect extrasynaptic LTD and rather facilitated LTD induction in the extrasynaptic region in the LHb of acute and chronic-stressed rats, it is highly probable that there may be a compensation mechanism for synaptic LTD extinction. Hyperactivity of excitatory neuron caused by stress in the LHb is likely to lead to LTD extinction causing depressive-like behavior and may serve as a safety box to prevent the excessive inhibition of monoamine system. Future study focusing on extrasynaptic LTD will have implications for pathogenesis of depressive disorders evoked by repeated stress in the extrasynaptic region in the LHb.
