dc.description.abstract암은 생명의 기본 단위인 세포의 성장 및 분열과정에 이상이 생겨 발생하는 질환으로 주요 사망원인으로 알려져 있다. c-Myc은 이러한 암의 발생에 관여하는 다양한 암유전자 중 하나로, 다양한 세포기능을 조절하는 데 관여하는 전사인자이다. c-Myc은 정상적인 세포의 생명활동에도 중요한 역할을 수행하므로 암세포 특이적으로 c-Myc의 활성을 조절하는 것이 매우 중요하다. 본 연구내용은 c-Myc이 과발현된 세포에서 특이적으로 세포사멸을 유도하는 것으로 확인된 화합물 C604, ((Z)-ethyl2-(4-acetoxy-3-methoxybenzylidene)-7-methyl-5(-3-nitrophenyl)-3-oxo-3,4-dihydro-2H-thiazole [3,2-a]pyrimidine-6-carboxylate) 의 작용기전을 다양한 암세포에서 규명하는데 초점을 두고 있다. 본 화합물은 불멸화된 사람섬유아세포에 c-Myc을 과발현한 세포 (STF-cMyc) 에서만 특이적으로 세포사멸을 유도했다. 세포사멸 기전을 규명하기 위해 C604를 처리한 세포주와 대조군 세포주 간 다양한 단백질들의 양적변화를 항체를 이용하여 조사하였고, apoptosis 과정에 중요한 caspase 3와 Poly (ADP-ribose) polymerase (PARP)의 cleavage를 확인하였다. C604에 의한 apoptosis는 c-Myc이 과발현 된 HCT116, SW620과 NCI-H23 등 다양한 암세포에서도 확인되어 c-Myc 이 과발현된 암세포주에 특이적으로 작용할 가능성을 규명하였다. C604를 처리한 세포주들에서 세포주기를 분석한 결과, G2 phase 분포가 증가함을 관찰함으로써 이 화합물은 c-Myc이 과발현된 세포에서 G2/M phase 세포주기 차단 현상을 야기시키고, 세포사멸이 이어짐을 확인하였다. 또한 CDC25A와 c-Myc의 단백질양이 증가하는 것을 관찰함으로써 C604에 의한 세포사멸 작용기전에 중요한 역할을 수행할 가능성을 확인하였다. 또한 C604이 p53이 비활성화 되어 있는 세포에서 세포사멸 효과가 상승한 것을 관찰하였다. 본 연구결과는 C604와 관련 응용물질들이 c-Myc이 과발현된 암세포의 특이적 사멸을 유도함을 보임으로서, 새로운 c-Myc 특이적 후보물질로써의 가능성을 제시한다.;Cancer is among the leading causes of morbidity and mortality. Various transcription factors including cellular myelocytomatosis oncogene (c-Myc) regulate the cellular pathways and cell cycles. c-Myc is a multifunctional transcription factor whose deregulated expression is associated with malignancy and poor clinical outcome in various cancer. In this study, I have tried to identify a new chemical for specifically targeting c-Myc. Initial screening was done with library compounds of 4000 chemicals. By conducting MTS assays, HJYou’s lab in National Cancer Center (NCC) identified a compound, (Z)-ethyl12-(4-acetoxy-3-methoxybenzylidene)-7-methyl-5(-3-nitrophenyl)-3-oxo-3,4-dihydro-2H-thiazole[3, 2-a] pyrimidine-6-carboxylate), C604 to induce cell death in cells with c-Myc overexpression. C604 was shown to induce apoptotic cell death in c-Myc-overexpressing Standardization of Human Diploid Fibroblast (STF) cells, but not in control cells by western blot analysis. Apoptotic proteins including cysteine-dependent aspartate-directed protease 3 (caspase 3) and poly (ADP-ribose) polymerase (PARP) were cleaved in response to C604 in STF-cMyc cells, but not in STF-Control cells. Consistent with the results from STF-cMyc, C604 induced apoptotic cell death in HCT116, SW620 and NCI-H23 cell lines which overexpress c-Myc, suggesting a role of C604 as an apoptosis inducer in cancer cells with c-Myc overexpression. In addition, flow cytometry assays and western blot analysis showed that C604 induced cell cycle arrest at G2/M phase and increased the expression of cell division cycle 25A (CDC25A) and c-Myc at protein level, respectively. The results implied that C604 could induce cell cycle arrest at G2/M phase, followed by apoptotic cell death through activation of caspase 3-dependent apoptotic pathway. In addition, C604 induced apoptotic cell death is done via p53-independet pathway as well as p53-dependent pathway. Taken together, these data suggest that C604 has the potential value as a novel therapeutic candidate for cancer patients with c-Myc overexpression.-
