dc.description.abstractPeptides have been widely evaluated as possible therapeutic drug candidates against protein-related diseases since they are representative biomolecules that possess a broad spectrum of physiological activities in vivo. Nevertheless, peptides are poor cell permeable and vulnerable to enzymatic degradation. Peptoids are oligomers of N-substituted glycines that have advantages such as easy synthesis, resistance to proteolytic degradation, and improved cell permeability. In order to provide peptoids with structural and functional diversity, many different types of derivatization strategies have been investigated. Peptoids were effectively derivatized using Sonogashira cross-coupling reaction strategy between terminal alkynes and aryl iodide on peptoid in the presence of Pd(0)/Cu(I) catalysts and DIPEA. Solid-phase Sonogashira cross-coupling reaction were optimized by screening the reaction conditions. Linear peptoids could be derivatized in excellent yields on solid-phase. Various acetylene surrogates were also tested to give adequate results. Our strategy allows straightforward incorporation of a variety of the side chains to broaden the chemical functionalities of peptoids. The formation of the peptoid derivatives was thoroughly confirmed, based on HPLC chromatogram and tandem mass spectrometry to determine the conversion yield and sequence of the desired peptoids. We have successfully developed novel conditions for solid phase peptoid derivatization using Sonogashira cross-coupling reaction. Furthermore, we also extended the reaction scope for more attractive cyclization on solid-phase by the same strategy. Cyclic peptoids were attained in good conversion yields. Our strategy for derivatization and cyclization of peptoids by Sonogashira coupling reaction should provide a robust platform for the construction of various peptoid derivatives and cyclic peptoids as useful chemical tools in drug development, chemical biology, recognition and related studies.;펩타이드는 생체 내에서 광범위한 바이오 활성을 갖는 대표적인 생분자이기 때문에 단백질 관련 질환에 대한 가능한 치료 약물 후보로서 널리 알려 져왔다. 그럼에도 불구하고 펩타이드는 세포 투과성이 좋지 않고 단백질분해효소에 취약하다. 펩토이드는 용이 한 합성, 단백질 분해에 대한 내성 및 개선 된 세포 투과성과 같은 대표적인 이점을 갖는 N- 치환 된 글린신의 올리고머이다. 또한 이산 다량 체로의 조립, 2차 구조로 접을 수 있는 기능, 분자 수송 체, 나노 물질 등으로 작용하는 등 다양한 구조적 및 기능적 특징이 부여된다. 또한 페토이드는 안정적인 구조를 갖고 있고 고리화 후 단백질분해효소에 더 강하다. 펩토이드에 구조적 및 기능적 다양성을 제공하기 위해, 많은 다른 유형의 유도체 화 전략이 연구되었다. 이러한 연구에 대한 노력 외에도, 우리는 소노가시라 커플링 반응을 결합방법을 사용했다. 고상에서 아이오도 페닐기를 갖는 몇몇 모델 펩토이드를 팔라듐 촉매로 소노가시라 반응 조건하에서 다양한 알킨으로 처리하여 매우 우수한 수율로 유도체 화 된 펩토이드를 수득하였다. 원하는 생성물에 대한 최대 수득률을 제공하도록 최적화된 반응 조건을 철저히 조사하였다. 우리를 또한 펩토이드의 고리화에 대한 위의 전략을 적용했으며 결과는 합리적 했다. 우리는 이 전략이 펩토이드의 유도체 화 및 고리 화 방법에 추가 될 수 있다고 믿는다.-
