Abstract
Veratrum section Veratrum (Melanthiaceae) is composed of 6-15 species in temperate and boreal zone of the Northern Hemisphere, inhabiting deciduous forests and alpine meadows. In Korea and Japan, sect. Veratrum comprises eight taxa: V. alpestre, V. dahuricum, V. dolichopetalum, V. grandiflorum var. maximum, V. oxysepalum, V. patulum, V. stamineum var. micranthum, and V. stamineum var. stamineum. However, species delimitation and taxonomic treatments have long been controversial due to their highly variable vegetative and floral morphology. Especially, the six taxa of sect. Veratrum except V. stamineum have been treated as V. album complex because of their variations and/or overlaps in morphological features. To re-examine the taxonomic status and phylogenetic relationships within sect. Veratrum in Korea and Japan, maximum parsimony (MP), maximum likelihood (ML), and haplotype analyses were conducted based on a total of 4,856 bp of nrDNA ITS and cpDNA coding and noncoding regions (matK, psbA-trnH, rpL16, and trnS-trnG). In addition, microsatellite markers were developed to resolve species delimitation for V. album complex in Korea using Illumina MiSeq sequencing method. Furthermore, the physiological activities of V. oxysepalum were investigated of anti-oxidant, anti-atopic, anti-inflammatory, and immunostimulating that have not been studied yet for Veratrum oxysepalum in Korea were investigated. The MP and ML trees and median-joining haplotype network were poorly resolved among taxa within sect. Veratrum except V. stamineum and V. grandilforum var. maximum in Korea and Japan. Veratrum stamineum, which has outward spreading, exerted stamens beyond the tepals, diverged first within sect. Veratrum and the remaining taxa, which have straight shorter stamens compared to the tepals, formed a monophyletic group with poor species delimitation and phylogenetic relationships among them. Moreover, V. stamineum var. micranthum and V. stamineum var. stamineum could be treated as one their original taxa, V. stamineum Maxim. Because the haplotype of V. stamineum var. micranthum was derived from V. stamineum var. stamineum due to only one substitution of sequence of 4,007 bp of cpDNA. The incongruent phylogenetic position of V. grandiflorum var. maximum between ITS and cpDNA (it shares its most recent common ancestor with poorly resolved sect. Veratrum species in ITS, whereas it is sister to V. stamineum in cpDNA) suggested that it represents possible ancient hybrid origin between V. stamineum and V. oxysepalum. Thus, in this study the taxonomic status of V. grandiflorum var. maximum was assumed the elevated to a species rank, V. maximum not a variety of V. grandiflorum. The remaining five taxa of V. album complex, V. alpestre, V. dahuricum, V. dolichopetalum, V. patulum, and V. oxysepalum were very closely related each other based on the results of the MP, ML, and haplotype analyses. In addition, their morphological variation ranges were overlapped, so they are considered to be conspecific, V. oxysepalum, which is known to be distributed in the widest area in Artic coast from Norway to Alaska and Russia, China, Korea and Japan among V. album complex. Furthermore, to verify their relationships of the taxa of V. album complex, seven polymorphic microsatellite markers were developed and characterized in 50 accessions of V. album complex in Korea using Illumina MiSeq. Collectively, the polymorphic markers amplified 51 alleles in a range of five-11 with an average of 7.3 alleles per marker. The mean observed heterozygosity (Ho) and expected heterozygosity (HE) were 0.700 and 0.618, respectively. These markers will be effective for genetic diversity and species delimitation of V. album complex in Korea. The physiological activities of anti-oxidant, anti-atopic, anti-inflammatory, and immunostimulating of 70 % ethanol of two parts (aerial and underground) of the Korean V. oxysepalum were investigated from 10 populations in Korea. The anti-oxidant activity was more efficient in the extracts of V. oxysepalum of underground part than aerial part, especially the extracts of V. oxysepalum of HA (Mt. Hwaak) was most efficient (IC50 – 336.80 µg/ml). The anti-atopic activity was efficient in the extracts of V. oxysepalum of aerial and underground part of GJ (Mt. Geumjeong), JJ (Jeju island), and SB (Mt. Sobaek) at 200 µg/ml concentration. The anti-inflammation activity was most efficient in the extracts of V. oxysepalum of aerial part of JJ. The immunostimulating activity was most efficient in the extracts of V. oxysepalum of aerial part of JJ. In conclusion, the 70% ethanol extract of Korean V. oxysepalum complex of JJ-aerial part promoted anti-atopic, anti-inflammatory, and immunostimulating activities.;멜란지움과 (Melanthiaceae) 여로속 (Veratrum)의 박새절 (section Veratrum)에는 6 내지 15종이 속해 있으며, 이들은 북반구의 온대 및 아한대 지역의 활엽 수림과 고산 초원 지대에 서식한다. 한국과 일본에 분포하는 박새절에는 V. alpestre, V. dahuricum, V. dolichopetalum, V. grandiflorum var. maximum, V. oxysepalum, V. stamineum var. stamineum, 그리고 V. stamineum var. micranthum의 8 분류군이 처리되어 있지만, 이 분류군들은 매우 다양한 형태적 변이를 나타내기 때문에 종의 구분과 분류학적 처리에 있어서 오랫동안 논란이 되어왔다. 특히 V. stamineum을 제외한 한국과 일본 박새절의 6 분류군은 변이와 형태적 특징의 중복으로 인해 V. album의 복합체로 처리되어 왔다. 이에 한국과 일본의 박새절 분류군을 대상으로 핵 DNA의 ITS와 엽록체 DNA의 4 개 구간 (matK, psbA-trnH, rpL16, trnS-trnG)에서 총 4,856 개의 염기서열을 maximum parsimony (MP), maximum likelihood (ML) 및 haplotype으로 분석하여 그들의 분류학적 위치와 계통발생학적 관계에 대하여 알아보고자 하였다. 그리고 Illumina MiSeq 시퀀싱 방법을 사용하여 한국의 V. album 복합체에 대한 microsatellite 마커를 개발하여 그 특성을 평가하였다. 더 나아가, 국내의 박새 (V. oxysepalum)에 대해 아직 연구되지 않은 생리활성인 항산화 및 항아토피, 항염증, 면역활성 효과에 대해 연구하였다. 총 4,856 개의 염기서열을 바탕으로 한 MP 및 ML, haplotype 분석에서 V. stamineum과 V. grandiflorum var. maximum을 제외한 한국과 일본의 박새절 분류군들은 각각의 종으로 지지되지 않았다. 화피보다 길게 위로 튀어나오는 수술을 가진 V. stamineum은 한국과 일본의 박새절에서 가장 먼저 분지되었으며, 화피보다 짧은 길이의 수술을 가진 나머지 분류군들은 단계통 그룹을 형성하였다. 또한, haplotype 분석 결과는 V. stamneum var. micranthum의 haplotype이 V. stamineum var. stamineum의 haplotype에서 단 한 개의 염기치환으로 연결되었기에, 이 두 분류군을 별개로 처리하기 보다는 단일한 분류군인 V. stamineum으로 처리하는 것을 지지하였다. 다음으로, V. grandiflorum var. maximum은 핵 DNA와 엽록체 DNA 구간의 계통발생 tree에서 불일치한 계통적 위치를 나타내었는데, 이것은 V. grandiflorum var. maximum이 V. stamineum과 V. oxysepalum과의 잡종으로 인해 형성된 분류군임을 지지하였다. 따라서 V. grandiflorum var. maximum을 V. album 복합체에서 분리함과 동시에, 이를 V. grandiflorum의 변종이 아닌 하나의 종인 V. maximum으로 처리함을 지지한다. 마지막으로, MP 및 ML, haplotype 분석 결과에 따르면 V. album 복합체의 나머지 분류군인 V. alpestre 및 V. dahuricum, V. dolichopetalum, V. patulum, V. oxysepalum은 서로 매우 밀접하게 연관되어 있으며, 이들의 형태학적인 변이의 범위가 중복되어 나타나기 때문에 이들을 각각의 분류군으로 처리할 수 없었다. 따라서 이들을 모두 V. album 복합체에서 가장 넓게 분포하며 가장 다양한 형태적 변이를 보이는 V. oxysepalum으로 처리하고자 한다. 또한, Veratrum album 복합체를 대상으로, 형태적 변이의 다양성으로 인하여 종의 구분이 어려운 분류군들의 유전적 다양성을 알아볼 수 있는 microsatellite 마커를 개발하고자 하였다. 그 결과로 Illumina사의 MiSeq 시퀀싱 방법을 사용하여 한국의 V. album 복합체의 유전적 다양성에 대해 알아볼 수 있는 7 개의 microsatellite 마커를 개발하여 50 개체에 대해 그 특성을 평가하였다. 그 결과, 7 개의 마커당 평균 7.3 개, 총 51 개의 allele을 확인하였다. 마커의 평균 observed heterozygosity (HO)와 expected heterozygosity (HE) 값은 각각 0.700과 0.618으로 유전적 다양성이 높은 것으로 나타났다. 이 마커들은 앞서 분석한 방법에 비해 한국의 V. album 복합체에 대하여 더 자세한 유전적 다양성의 정보를 줄 것으로 기대한다. 마지막으로, 한국의 박새의 생리활성 효과를 알아보고자 전국 10개 지역에서 채집한 박새를 지상부와 지하부로 나누어 70 % 에탄올을 가하여 그 추출물로 항산화 및 항아토피, 항염, 면역활성 효과를 알아보았다. 항산화 활성은 HA (화악산)의 박새추출물에서 가장 높은 효과를 나타내었으며 (IC50 – 336.80 µg/ml), 항아토피 활성은 200 µg/ml 농도의 GJ (금정산) 및 JJ (제주도), SB (소백산)의 지상부와 지하부 박새추출물에서 가장 높은 효과를 나타내었다. 항염증과 면역증강 활성은 모두 JJ (제주도)의 지상부 박새추출물이 다른 지역의 추출물에 비해 가장 높은 효과를 나타내었다. 결론적으로 한국의 10 개의 지역 중에서 JJ (제주도)의 지상부 박새추출물은 세포에 대한 독성이 거의 나타나지 않는 동시에 항아토피 및 항염, 면역활성 효과를 촉진시키는 것으로 나타났다.
