- Abstract
- The object of this study is to clarify the optimal processing condition that can maximize the antioxidant and α-amylase inhibition activity α of the coffee bean extract.
The coffee bean were prepared different degree of roasting (green bean, light, medium and dark), grinding temperature (ambient, cryogenic and cryogenic-fine), and extraction method (hot-water and microwave-assisted extraction). Physicochemical properties of beans and powder, total polyphenol contents, total flavonoid contents, antioxidant and α-amylase inhibition activity were investigated.
According to the degree of roasting, the weight decreased, L, a, and b value decreased, substantially. The antioxidant and α-amylase inhibition activity was highest in light-roasted beans. Then, the proper thermal
processing can increase the antioxidant activity of coffee beans. The total polyphenol contents, total flavonoid contents, ABTS, DPPH and a-amylase inhibition values of hot water extracted ground coffee via different processing conditions were in the ranges of 6.14 - 11.14 mg GAE/g, 18.85 - 33.23 mg QE/100g, 22.42 - 32.40 mM Trolox, 38.06 - 76.40 % and 21.21 - 68.91%, respectively. And light-roasted and cryogenic-fine ground coffee had the highest results among all samples. The microwave-assisted extraction (MAE) method increased the antioxidant capacity by 4.50 % (DPPH) and 28.73 % (ABTS) compared with the general hot-water extraction.
As a result, light-roasted and cryogenic-fine grinding is considered to be the most suitable processing condition for enhancing the bioactivity of coffee beans. And MAE (450W, 15s) method was more effective than conventional hot-water extraction method of the coffee bean extract.;본 연구의 목적은 커피추출물의 기능성을 향상시킬 수 있는 가공조건을 최적화 하는 것이다.
커피 원두는 항산화, 항당뇨 기능을 가지고 있으며, 폴리페놀 및 플라보노이드 등을 비롯하여 다양한 생리 활성 화합물이 항산화, 항당뇨 작용에 기여한다고 알려져 있다.
원산지 (브라질, 코스타리카, 콜룸비아, 케냐), 배전 정도(생두, 저배전, 중배전, 강배전), 분쇄 온도 (상온, 저온) 및 추출 방법 (열수추출, 마이크로파 추출)에 따라 커피 분말의 물리 화학적 특성, 항산화와 α-amylase 억제 기능을 분석하였다. 가공 방법에 따른 입자 특성을 확인하기 위해 주사 전자 현미경 (SEM)으로 가시화한 결과, 저온 분쇄한 시료는 상온 분쇄한 경우보다 균일하고 더 작은 입자로 나타났다. 또한 DPPH 및 ABTS 결과에 따르면, 배전 정도와 항산화능은 비례하지 않았다. 이는 커피에 존재하는 폴리페놀이 적절한 열가공에 의해 분해 또는 새로운 화합물이 생성되어 항산화능이 증가된 것으로 생각된다. 열수추출중 총 폴리페놀, 총 플라보노이드, ABTS, DPPH와 α-amylase 억제 기능을 측정한 결과는 각각 6.14 - 11.14 mg GAE/g, 18.85 - 33.23 mg QE/100g, 22.42 - 32.40 mM Trolox, 38.06 - 76.40 % and 21.21 – 68.91% 이다. 그중 저배전 및 저온분쇄로 가공한 커피가 제일 높은 결과를 보인다. 또한 마이크로파 보조 추출법 (MAE)을 사용하여 열수추출보다 4.50% (DPPH) 및 28.73% (ABTS) 항산화능을 향상시켰다.
본 연구를 통해 커피는 적절한 열가공으로 생성된 물질에 의해 항산화능이 증진됨을 확인하였고 커피의 기능성을 극대화하기 위한 가공 조건 (약배전, 저온미세분쇄 및 마이크로파 추출(450w, 15s) )을 확립하였다.
추후 가공 과정에 따라 형성되는 화합물에 대한 규명 및 각 물질의 생체 활성에 대한 더욱 깊은 연구가 필요하다고 사료된다.
