Abstract
Part I. Calpain 2 Enhances Mesenchymal Shift Through the HIF1α-Twist Axis in Triple Negative Breast Cancer Cells Breast cancer is highly ranked as one of the most common cause of cancer death. Although Basal-Like Breast Cancer (BLBC) is not the major subtype of breast cancer in population, it is responsible for a disproportionate share of mortality with higher grade tumors and higher rates of distant recurrence. Because most of the BLBC patients represent resistance to conventional chemotherapies, new approaches are urgently needed to improve therapeutic outcome in BLBC treatment. Calpain, a family of calcium-dependent cysteine proteases, has a wide range of cellular functions related to cancer, including functions in apoptosis, hypoxia, proliferation, migration and invasion. The correlation between m-calpain (calpain 2) expression and the clinical outcome in BLBC was investigated using the database analysis. Calpain 2 knock-down cellular model was constructed using mesenchymal BLBC cell lines. Changes in the cellular morphology, proliferation rate, colony forming ability and migration and invasion capacities of calpain 2 knock-downed (KD) cells were compared to those of empty vector transduced cells. Calpain 2 KD cells showed cobblestone like shape with increased cellular circularity, reduced anchorage-dependent and -independent growth, and downregulation of migration and invasion compared to empty vector transduced cells. EMT related gene expression was also investigated. EMT was reversed by calpain 2 KD. Among the EMT related transcription factors, only Twist showed consistent reduction in protein and mRNA levels. HIF1α, a transcription factor which is known to directly bind to Twist promoter, was investigated for its subcellular localization and binding to HRE (Hypoxia-Response Element) in Twist promoter using the immunofluorescence assay and ChIP assay, respectively. The percentage of HIF1α in the nuclear fraction was decreased by calpain 2 KD. The interaction between HIF1α and HRE of Twist promoter was also inhibited by calpain 2 KD. Hypoxia-induced Twist expression was diminished by calpain 2 downregulation. Calpain 2 re-expression by transient transcription recovered mesenchymal property with downregulation of ZO-1 level. Moreover, filamin A cleavage was also decreased by calpain 2 KD. The interaction between HIF1α and C-terminal fragment of filamin A was decreased. Anoikis sensitivity in calpain 2 KD cells was investigated using WST assay, Annexin V/PI double staining, and immunoblotting analysis. Calpain 2 downregulated cells obtained anoikis sensitivity. The results indicate that calpain 2 could play an important role in EMT process via HIF1α translocation, followed by Twist expression. Calpain 2 might serve as a novel target for therapeutic strategies of BLBC patients. Part II. Development of Dual Inhibitor Against μ-Calpain Enzymatic Activity and Aβ Aggregation μ-Calpain, a calcium-dependent cysteine protease, is hyper-activated when the intracellular calcium homeostasis is disrupted in Alzheimer’s disease. Hyper-activation of μ-calpain increases the expression level of β-secretase (BACE1) and, consequently, enhances the accumulation of β-amyloid (Aβ) peptide leading to extracellular senile plaques. Also, activated μ-calpain induces hyper-phosphorylation of tau along with the formation of neurofibrillary tangle (NFT) through p35 cleavage into p25. The accumulation of Aβ peptide and the formation of NFT are the major mechanisms of neurodegeneration in AD. To find the novel μ-calpain inhibitor, we screened the chalcone derivatives. Among them, CNa 5 showed inhibitory activity to μ-calpain in experiment using recombinant enzymes and the enzyme-overexpressed cellular lysate. CNa 5 effectively inhibited expression of BACE1, formation of Aβ peptide, cleavage of p35 to p25, and phosphorylation of tau under the condition of oxidative stress. μ-Calpain inhibitory activity of CNa 5 was confirmed by docking study, as the compound was suited at the active site of μ-calpain. CNa 5 had exceptionally strong neuroprotective activity when considering the calpain inhibitory potency. For this reason, inhibitory activity of Aβ aggregation was evaluated. CNa 5 inhibited β-sheet aggregates formation which was confirmed using ThT (Thioflavin T) staining and CD (Circular Dichroism) spectroscopy and docking study showed the possible binding mode of CNa 5 to Aβ pentamer. Thus, we concluded the novel chalcone derivative, CNa 5, is a potent drug candidate for the treatment of AD by inhibiting both μ-calpain and Aβ aggregation.;기저형 유방암은 유방암의 아형 중 높은 전이 및 재발률과 더불어 가장 좋지 않은 예후를 가지는 아형이다. 기저형 유방암 환자는 표적 치료를 위한 표적 단백질을 발현하지 않으며 그 예후가 나쁘기 때문에 새로운 치료 전략의 수립이 필요한 실정이다. Calpain은 칼슘 의존적 프로테아제로, 종양과 관련된 다양한 세포 기능 전반에 걸쳐 주요한 역할을 담당하는 것으로 알려져 있다. 본 연구에서는 calpain 2 발현과 기저형 유방암의 임상 결과와의 상관성을 데이터베이스 분석을 통하여 확인하였다. 데이터베이스 분석에서 calpain 2의 발현은 기저형 유방암의 아형 특이적으로 위험(risk)를 증가시켰으며 상피간엽이행 관련된 다양한 유전자의 발현과 상관관계를 나타냄과 동시에 기저형 유방암의 생존과도 밀접한 연관성을 나타내었다. 데이터베이스 분석 결과를 바탕으로 간엽성 특징을 가지는 기저형 유방암 세포주에서 calpain 2을 선택적으로 침묵시킨 세포 모델을 확립하였고 그에 따른 표현형의 변화를 확인하였다. Calpain 2를 침묵시킨 세포 모델에서 세포의 형태와 부착 의존적 및 비의존적 세포 증식 속도의 변화를 확인하였다. 이와 더불어 calpain 2 침묵 세포에서 세포의 이동성 및 침습성의 감소를 확인하였으며 이를 바탕으로 상피간엽이행과 관련된 표지 단백질의 변화를 확인하였다. Calpain 2 침묵 세포에서 상피간엽이행 관련 단백질 중 상피성 표지 단백질은 증가하고 간엽성 단백질은 감소하는 양상이 확인되었고 해당 단백질의 mRNA 수준에서도 유사한 결과를 나타내었다. 또한, 상피간엽이행 표지 단백질의 발현을 조절하는 다양한 관련 전사인자들의 발현을 조사하였을 때 Twist의 발현만이 특이적으로 calpain 2 침묵 세포에서 감소하는 것이 관찰되었다. Twist 발현의 변화는 mRNA 수준에서도 확인되었기 때문에 Twist의 발현을 조절하는 전사인자인 HIF1α의 세포 내 분포를 조사하였다. HIF1α는 세포질에서 핵으로 이동함으로써 그 활성이 증가하게 되는데 calpain 2를 침묵시킨 세포에서 핵 내 HIF1α의 분포가 현저히 낮게 나타났다. 이러한 변화는 CoCl2로 유도된 HIF1α의 활성화에 의한 Twist의 mRNA 수준 변화로도 확인되었는데, calpain 2 침묵 세포에서는 CoCl2에 의한 Twist mRNA 증가가 관찰되지 않았다. 이와 더불어 calpain 2 침묵 세포에서는 HIF1α와 Twist의 promoter 상의 HRE 부분과의 상호작용이 현저히 줄어드는 것이 확인되었다. Calpain 2 침묵 세포에서 다시 calpain 2를 발현시켰을 때 상피성 표지 단백질인 ZO-1의 발현이 감소하였으며 HIF1α의 세포 내 분포를 결정짓는데 중요한 단백질인 filamin A의 절단도 calpain 2 침묵 세포에서 감소하였던 것이 다시 증가되는 양상을 확인하였다. 절단된 filamin A와 HIF1α의 상호작용이 calpain 2 침묵 세포에서 감소되는 것이 관찰되었으며 실제 종양 조직에서와 유사한 배양 조건인 삼차원적 배양에서도 Twist의 발현이 감소되는 것을 관찰할 수 있었다. Calpain 2와 종양 전이와 관련하여 상피간엽이행 뿐만 아니라 비부착 상태에서 유도되는 세포 사멸인 아노이키스와의 연관성도 확인하였다. Calpain 2 침묵 세포는 부유 배양 시 생존율이 감소되는 양상을 나타내었으며 세포 사멸 관련 단백질의 발현변화와 세포 사멸을 확인하기 위한 Annexin V/PI 염색을 이용한 실험 결과에서도 calpain 2 침묵 세포에서 더 많은 세포 사멸이 유도됨을 관찰하였다. 이는 calpain 2 를 감소시킴으로써 아노이키스에 대한 감수성을 획득할 수 있음을 시사하는 결과이다. 결론적으로 calpain 2의 발현을 조절하는 것은 상피간엽이행에서의 Twist를 조절하고 아노이키스에 대한 감수성을 획득시킴으로써 새로운 치료 표적으로써의 높은 개발 가능성을 보여준다. μ-Calpain (Calpain 1)은 알츠하이머 병에서 무너진 칼슘 항상성에 의해 과활성화되는 것으로 알려져있다. μ-Calpain의 과활성화는 β-secretase (BACE1)의 발현을 증가시킴으로써 β-amyloid의 축적을 유도한다. 또한, CDK5의 조절자인 p35 단백질을 p25로 잘라 CDK5의 활성을 지속적으로 유지시키고 그에 따라 tau 단백질의 과인산화를 초래함으로써 neurofibrillary tangle (NFT)의 형성을 유도한다. β-Amyloid 축적에 의한 β-amyloid의 응집과 NFT의 형성은 알츠하이머 병에서 가장 잘 알려진 두 가지 발병 기전으로 μ-calpain은 그 두 가지 기전을 유기적으로 연결하는 주요한 치료 표적이다. 새로운 μ-calpain 저해제를 개발하기 위하여 일련의 chalcone 유도체를 스크리닝 하였고 그 중 CNa 5는 μ-calpain 저해능을 나타냄과 동시에 매우 높은 신경세포 보호 효과를 나타내었다. CNa 5는 BACE1의 발현을 억제하고 β-amyloid의 형성을 저해하였으며 p35가 p25로 절단되는 것을 막아 tau의 인산화를 억제하였다. 이러한 효과는 μ-calpain 억제 효능을 통하여 나타나는 것으로 예상되며 μ-calpain의 직접적 억제 효과는 docking study를 통하여 μ-calpain의 활성부위에 직접 CNa 5가 작용함으로써 나타나는 효과임을 규명하였다. CNa 5의 신경세포보호효과는 μ-capain의 억제 효능을 고려할 때 매우 강력했기 때문에 β-amyloid의 응집에도 관련이 있을 것으로 예상되어 이를 조사하였다. CNa 5는 β-amyloid가 응집되면서 나타내는 베타병풍구조(β-sheet)의 형성을 억제하였고 이를 Thioflavin T 염색과 원편광이색성(Circular Dichroism)을 조사함으로써 평가하였다. 또한 docking study를 통하여 CNa 5가 β-amyloid의 오량체(pentamer)와 결합하는 양상을 확인함으로써 그 가능성을 평가하였다. 결과적으로 새로운 chalcone 유도체인 CNa 5가 알츠하이머 병에서 μ-calpain의 효소 활성을 억제함과 동시에 β-amyloid의 응집을 저해함으로써 새로운 치료제로써의 가능성을 나타내었다.
