Abstract
PINK1, linked to familial Parkinson’s disease (PD), is known to affect mitochondrial function. Parkinson’s disease is the second most common neurodegenerative disorder in the world. However, current medicine can only call on symptomatic treatment for dopamine depletion, and no drugs that rescue mitochondrial dysfunction are yet available. Lack of appropriate phenotypic models and poor understanding of pathogenesis of mitochondrial deficits have restrained the development of mitochondrial drugs. Thus, the present study is focused on finding compound and its mechanism of rescuing mitochondrial enzyme activity in PINK1 null dopaminergic cell model. Drug-screening identified ginsenoside Re as a compound capable of reversing the deficit in complex IV activity in PINK1 null cells. Importantly, the pharmacological effects of ginsenoside Re could be reversed by treatment of the pan-NOS inhibitor, L-NAME, and could be reproduced by low level NO treatment. In our pervious study, Hsp60, LRPPRC, and Hsp90 were identified as upstream regulators of complex IV activity. Treatment of ginsenoside Re enhances functioning of the defective PINK1-Hsp90/LRPPRC-Hsp60-complex IV signaling axis in PINK1 null neurons by restoring NO levels, providing potential for new therapeutics targeting mitochondrial dysfunction in PD.;PINK1은 가족성 파킨슨씨병을 유발하는 유전자 중 하나로서, 미토콘드리아의 기능과 연관 되어있음이 알려져 있다. 파킨슨씨병은 전세계적으로 두번째로 가장 흔한 퇴행성 뇌질환이다. 하지만, 현재 의학에서는 부족한 도파민을 공급해주는 등의 임시적인 치료만 가능하며, 더 근본적인 미토콘드리아 기능 이상을 회복시키는 치료제들이 아직 개발되지 않고 있다. 그 동안 미토콘드리아 기능 회복 약물 발굴이 어려웠던 점으로서 좋은 표현형을 가진 모델이 부족한 점 과, 미토콘드리아 기능 이상은 어떤 부분을 타켓으로 하는지 그 병리적인 기전이 제대로 밝혀지지 않은 점을 제시할 수 있다. 따라서, 본 연구에서는 PINK1 유전자가 결여 된 도파민 신경세포 모델을 이용하여 미토콘드리아 기능이상을 회복시키는 약물과 그 기전을 찾는 데에 중점을 두었다. 약물 스크리닝을 통해 ginsenoside Re 가 PINK1이 결여된 세포에서 전자전달계 complex IV (cytochrome c oxidase) 장애를 회복시킬 수 있음을 발견했다. 중요하게도, ginsenoside Re의 약물학적 효과는 nitric oxide (NO)를 생산하는 모든 종류의 nitric oxide synthase 저해제인 L-NAME으로 차단되고, 낮은 농도의 NO 공급으로 약물학적 효과가 재현되는 것을 발견하였다. 본 실험실의 과거 연구 결과에서, Hsp60, LRPPRC 와 Hsp90이 complex IV의 위 단계 조절 단백질임을 밝혔다. PINK1이 결여된 신경세포에서 3μM Ginsenoside Re의 24시간 처리는 Nitric oxide 공급을 통해 손상된 PINK1-Hsp90/LRPPRC-Hsp60-complex IV signaling axis가 상당히 회복되는 우수한 효과를 나타내며 앞으로 파킨슨씨병의 미토콘드리아 기능 이상 치료제 약물로 개발 될 가능성을 제시한다.
