Abstract
최근 많은 고분자와 생체물질들이 다양한 산업분야에 활용되어지고 있습니다. 이 논문에서는 분자 동역학 시뮬레이션 방법을 사용하여 고분자와 생체물질의 구조 변화 뿐만 아니라 다른 분자 간의 상호작용을 관찰하였습니다. 분자 단위에서의 고분자 및 생체물질 관찰은 근본적인 과학 지식에 기여할 수 있을 것이라고 생각합니다. Chapter Ⅰ에서는 PNIPAM의 가역적인 펴짐 – 뭉침 현상을 관찰하기 위해 분자 동역학 시뮬레이션을 사용하였습니다. PNIPAM은 수용액 상태에서 약 32℃ 에서 LCST를 보이는 잘 알려진 온도 민감성 고분자입니다. 생체 온도와 근접한 온도에서 일어나는 펴짐 – 뭉침 성질 때문에 PNIPAM은 여러 분야, 특히 생체의학 분야에서 각광받고 있습니다. 이 논문에서는 임계점 근처에서 고분자의 뭉침 정도에 따른 3가지의 서로 다른 구조로부터 시작하여 1마이크로초에 이르는 긴 시뮬레이션을 수행하였습니다. 시간에 따른 뭉침 정도를 파악하여 가역적인 구조 변화를 관찰하였고 추가적으로 시간에 따른 소수성 상호작용, 물과 고분자간의 수소결합, 고분자 내의 수소결합 등을 분석했습니다. Chapter Ⅱ 에서는 세라마이드가 O/W 에멀전의 안정성에 미치는 영향을 관찰하기 위해 세라마이드를 포함하는 지질 이중층과 단일층 구조를 모델링하여 분자 동역학 시뮬레이션을 수행하였습니다. 세라마이드는 피부 각질층의 필수 성분으로 외부 유해물질의 피부침투를 방지하고 피부 속 수분증발을 막는 역할을 하는 것으로 알려져 있습니다. 이와 같은 특징때문에 세라마이드는 최근 화장품과 피부 연고 등에 활용되어지고 있지만, 아직까지 에멀전에서의 세라마이드 거동 메커니즘이 밝혀지지 않은 상태입니다. 지질 이중층과 단일층 내에서 세라마이드가 구조적 안정성에 미치는 영향을 알아보기 위해 우리는 지질 층 내에서의 세라마이드3b와 계면활성제의 비율을 다양하게 변화시켜가며 리피드 당 면적 값과 밀도를 계산하였습니다. 그 결과, 지질 이중층과 단일층 모두 100 나노초 동안 안정한 구조를 유지한다는 것을 관찰할 수 있었습니다.;In recent years, a lot of polymers and biomaterials have been used in a variety of industries. Here, we present molecular dynamics (MD) simulations of polymer and biomaterial to observe not only structural change but intermolecular interaction. Our observations on a molecular scale for polymer and biomaterial may contribute to many kinds of scientific interests fundamentally. In chapterⅠ, MD simulations are employed to investigate the reversible swelling-collapse transitions of a single poly (N-isopropylacrylamide) (PNIPAM) chain. PNIPAM is a well known temperature responsive polymer which shows LCST around 32℃ in aqueous solution. Due to this temperature-responsive coil-globule transition near physiological temperature, a variety of PNIPAM-based materials have been proposed, specially in various biomedical applications. In this study, we performed simulations at different temperatures near the LCST starting with three different conformations: extended, loosely collapsed, and tightly collapsed conformations. We observe the reversible swelling-collapse transition during we analyze simulation trajectories in terms of radius of gyration, intrachain distances, hydrophobic contacts, and chain-water and intrachain hydrogen bonding. In chapter Ⅱ, we perform MD simulations of bilayer and monolayer structures containing ceramide 3b to investigate the effect of ceramides on the oil-in-water (O/W) emulsion from a view of formulation stability. Ceramides, one of the major lipid components in the stratum corneum, is known to be effective in maintaining the moisture of the skin and enhancing the vitality of the skin. Therefore, ceramides have been given increasingly attention to in several topical applications including cosmetics and medications. Despite its usefulness, however, their microscopic behaviors in emulsions remain elusive. To investigate the compositional effect of ceramide 3b on structural stability of bilayers and monolayers, we varied compositions of ceramide 3b and C16-alkyl glucosides in layers and calculate the area per lipid (APL) and density profiles. It is found that the both bilayers and monolayers remained stable during 100 ns simulations. This work suggests the stability of ceramide-containing bilayer and monolayer structures in emulsions.
