dc.description.abstractAdipose tissue is an endocrine organ that regulates the energy homeostasis and the inflammatory reaction. The risk for Type 2 diabetes can be increased in a failure of adipocyte differentiation. Peroxisome proliferator-activated receptor gamma (PPARɤ) agonists that drive the differentiation, have been shown for their therapeutic potentials of type 2 diabetes. Cell differentiation is fundamentally influenced by epigenetic mechanisms, but the ones contributing to adipogenesis remain to be elucidated. Epigenetic mechanism is regulation of gene expressions through changes in chromatin structure, not in DNA sequences, using DNA methylation and histone modification. A previous study from our laboratory showed that adipogenesis was blocked in absence of ten-eleven translocation methyl-cytosine dioxygenase 2 (Tet2) gene, active DNA demethylases, via DNA methylation-mediated downregulation of the Pparɤ expression. Interestingly, when the Tet2 gene was knockdown after the adipogenic induction, the effect was not observed, suggesting that TET2 plays an important role in the early stage of adipogenesis. We analyzed the genome-wide mRNA expression profile at 4 hours after adipogenic differentiation induction to confirm the role of Tet2 gene at that time. The knockdown of Tet2 gene altered transcriptional regulations related with cell cycle, DNA replication, and ribosome. The Tet2 knockdown interrupted transcriptional alterations of the adipogenesis initiation. The Ddx5 expression was upregulated at 4 hours after adipogenic differentiation induction, while the knockdown of Tet2 gene marginally blocked this induction (p = 0.06). Level of 5hmC was decreased upon adipogenesis on the putative promoter and enhancer at the Ddx5 locus (p < 0.05), whereas it was highly enriched on the gene body after adipogenesis. The knockdown of Ddx5 prior to adipogenic differentiation induction also downregulated lipid accumulation and the Pparɤ expression (p < 0.05). Taken together, the Ddx5 expression regulation by TET2 might be vital for the beginning of adipogenesis.;지방은 에너지 저장소이자 혈당 항상성 유지와 면역 반응을 조절하는 내분비 기관이다. 지방 분화가 제대로 일어나지 않으면 제 2형 당뇨병의 발병률이 높아질 수 있고 지방 분화의 핵심 조절 인자인 PPARγ의 작용제는 제 2형 당뇨병의 치료제로 사용된다. 분화 과정은 후생유전학적 기전과 연관이 깊다고 알려져 있지만 지방 분화와 후생유전학적 기전의 관련성에 대한 연구는 미미하다. 후생유전학적 기전이란 DNA의 염기 서열의 변화 없이 유전자 발현을 조절하는 것을 뜻한다. 대표적인 조절 기전으로는 DNA 메틸화와 히스톤 변형이 있다. 본 연구에서는 DNA 메틸화 제거에 관여하는 효소인 TET2의 발현 변화가 지방 분화에 영향을 미치는 기전을 연구하고자 하였다. 지방 분화 여부는 중성지방 축적량과 PPARγ 및 그 표적 유전자 발현으로 측정되었다. Tet2 유전자 발현 억제한 뒤 지방 세포 분화 유도하면 분화가 억제되었지만, 분화 유도 시점이나 3일 이후에 Tet2 유전자 발현 억제 시에는 지방 분화가 정상적으로 일어났다. 이를 통해, TET2는 초기 지방 분화에 영향을 준다는 것을 알 수 있다. 따라서, 초기분화 시 Tet2의 표적이 되는 인자를 살펴보고자, 발현 억제 2일 후 지방 분화를 유도 시키고 4시간 뒤에 mRNA 발현 변화를 전장적으로 보고자 Microarray를 통해 측정하였다. 지방 분화에 의해 발현이 올라간 유전자 중 97개는 Tet2 발현 억제로 인해 발현이 감소되었고 지방 분화에 의해 발현이 감소한 유전자 중 42개는 Tet2 발현 억제로 인해 발현이 증가하여 Tet2 발현 억제는 분화 전 발현상태와 산관성이 높음을 나타냈다. Tet2 유전자 발현 억제로 인해 변한 유전자들은 대부분 세포 주기, DNA 복제, 리보솜 구조 구성과 관련 있었다. RNA 헬리카제로 알려져 있는 Ddx5 유전자 발현 변화도 발견되었다. Tet2 유전자 발현을 분화 2일 전 억제한 경우, Tet2 유전자 발현 억제한 군과 대조군 간의 Ddx5 유전자 발현 정도의 차이가 분화시점에는 차이가 없었지만, 분화 후 4시간 때에는 Tet2 유전자 발현을 억제한 군에서 Ddx5 유전자 발현 정도가 낮아졌다. 지방 분화가 일어나면 Ddx5 유전자의 발현 조절 구간인 프로모터와 인헨서 추정 구간에서 5hmC의 수준이 감소하였다. Tet2 유전자 발현을 조절했을 때와 마찬가지로 지방 분화 유도 전 Ddx5 유전자 발현을 억제한 경우 지방 분화가 일어나지 않았다. 이러한 결과들을 통해 초기 지방 분화 시 TET2가 DNA 탈메틸화 작용을 이용해 Ddx5 유전자 발현을 조절할 수 있다는 것을 추정할 수 있다. Tet2에 의한 Ddx5 유전자 발현 조절로 Pparγ 유전자 발현을 조절할 수 있으므로 본 연구의 결과는 지방세포 분화 초기의 후생유전적 기전을 제시하며, 이후 새로운 제 2형 당뇨병 치료제 발굴의 기초 자료로 사용될 수 있다.-
