Association of plasma adiponectin and leptin levels with ovarian cancer

Abstract

Decreased adiponectin and increased leptin concentrations are found to be associated with the development of obesity-related cancers such as endometrial cancer and breast cancer. It has been suggested that ovarian cancer might have similar risk factors like nulliparity, genetic mutations. The purpose of this study was to explore the association of plasma adiponectin and leptin level with ovarian cancer. In this study, we compared the plasma adiponectin and leptin concentration in ovarian cancer patients with those of normal control. In addition, we investigated whether there are differences in the concentration of adiponectin and leptin between subgroups classified by surgical stages and histologic types. We measured adiponectin and leptin levels of patients and controls using ELISA technique; we ascertained anthropometric data and clinicopathologic data by review of medical record. The mean adiponectin and leptin concentrations in ovarian cancer patients were lower than those of the control group with statistical significance. However, there was no significant difference in adiponectin and leptin levels in the subgroup analysis; early-stage (I/II) versus advanced-stage (III/IV) disease, serous type versus non-serous type. We found that not only plasma adiponectin level but also leptin level was decreased in ovarian cancer patients, which is different from the previous reports with other cancers. Additional study with larger sample size is necessary in order to explicit the role of adiponectin and leptin in the development of ovarian cancer.;목적: 비만과 관련있는 혈중 adiponectin 및 leptin은 자궁내막암 및 유방암의 발생과 진행에 연관되어 있는 것으로 알려져 있다. 본 연구는 비만과 유관한 또 다른 암종인 난소암 환자에서서 혈장 내 adiponectin 과 leptin 농도가 난소암 발생과 연관 있는지 확인 하고자 하였다. 방법: 난소암 환자와 대조군의 혈장 내 adiponectin 과 leptin 농도를 ELISA방법으로 측정하고, 환자의 임상정보는 의무기록을 검토하여 수집하였다. 혈중 adiponectin, leptin 농도와 난소암의 발생, 병기 및 조직형과의 연관성을 분석하였다. 결과: 난소암 환자의 혈장 내 adiponectin과 leptin 농도는 대조군에 비해 유의하게 낮았다. 각 (8.25μg/mL vs 11.44μg/mL, p=0.026) (7.09 ng/mL vs. 15.4 ng/mL, p=0.001). 그러나, 초기 난소암환자(I/II기)와 진행된 난소암 환자(III/IV기)간, 장액성 난소암과 비장액성 난소암 사이간에 혈장 내 adiponectin, leptin 농도 사이에는 통계적으로 유의한 차이는 보이지 않았다. 결론: leptin 농도가 다른 암종의 연구에서와는 달리 난소암에서 대조군에 비해 반대로 유의하게 낮아지는 현상을 보이며 이러한 현상을 명확히 규명하기 위해 더 많은 난소암환자를 대상으로 한 추가 연구가 필요하다고 본다.
