Abstract
Obesity is exposed to everyone and the typical risk factor of various chronic diseases. Association obesity and chronic disease is already well known. Obesity is occurred by lifestyle, environmental factors and genetic factors. Recently, genetic studies have been made actively with completion of human genome sequencing Fat mass and obesity-associated (FTO) gene is one of the famous obesity-associated genes, which is founded through Genome wide association study (GWASs). Recent studies discovered that the region covering obesity-related the single nucleotide polymorphisms (SNPs) at FTO locus regulates transcriptional expression of homeobox genes, called IRX3 and IRX5. There is controversy about the correlation between FTO and obesity that were mentioned in the previous studies. This controversy is even more grows because they have not been fully identified the biochemical function of FTO. So, by comparing various phenotype of a mouse model produced by manipulating the Fto gene in this point, Fto is investigated that what functions associated with growth and development of obesity. There are shown different phenotypes by mouse models depending on target tissue and gene manipulation. Generally, knocked out Fto gene expression is showed that increased postnatal mortality, decreased anthropometric data and changed eating behavior, food intake, various hormone levels. Above all, there are different results compared germline knock out mouse model and adult onset mouse model. It is happened only timing of Fto gene is knocked out. Repressing Fto expression in adult onset mouse model decreased mortality compared with germline knock out mouse model, but showed the same pattern increased body fat mass and body weight. This result imply that FTO gene have an important additional function not only obesity but also growth in early development period. Further studies are required to determine the effect of FTO gene detailed. From these studies, obese people might have help and apply for obesity-related disease prevention.;비만은 누구에게나 노출되어 있는 대표적인 만성질환의 위험 요인 중 하나이다. 비만은 생활습관 및 환경, 그리고 유전적 요인에 의해 발생한다고 알려져 있으며, 최근 인간 게놈 염기서열의 완성과 함께 관련 유전자에 대한 연구가 활발히 이루어져 왔다. 그 중에서도 전장유전체 상관성 분석(GWAS: Genome wide association study)을 통해 발견된 FTO(Fat mass and obesity-associated protein) 유전자를 빼놓을 수 없다. FTO 유전자는 대표적인 비만유전자로, 이 유전자 내에 위치한 다양한 단일핵산염기 다형성(Single nucleotide polymorphisms: SNPs)이 특히 체질량 지수, 체중, 체지방량과 상관성이 높다는 사실이 많은 인종에서 공통적으로 도출됨으로써 주목을 받았다. 그러나 최근 FTO 유전자 내에 존재하는 비만과 관련된 단일핵산염기 다형성의 염기서열 위치가 FTO가 아닌 그 유전자 가까이 존재하는 IRX3, IRX5라고 불리는 호메오박스 유전자들의 전사발현을 조절한다는 것이 밝혀지면서 기존에 언급되던 FTO와 비만의 상관성에 대해 논란의 여지가 있다. 이러한 논란은 FTO의 생화학적 기능이 완전하게 규명되지 않았기 때문에 더욱 가중되고 있다. 그러므로 이 시점에서 FTO 유전자를 조작하여 만들어진 다양한 종류의 생쥐 모델들의 표현형들을 비교분석하여 FTO가 성장 발달 및 비만과 관련된 어떤 기능을 하는지 정리해보고자 하였다. 생쥐 모델을 살펴보면, 유전자 조작의 세포 및 조직이 어디인지에 따라, 그리고 조작 방법에 따라 조금씩 다른 표현형을 보인다. FTO의 발현 억제는 생쥐의 출생 후 사망률을 증가시키고, 신체구성이나 체중 등의 신체계측치의 감소를 보인다. 또한 신체의 항상성 유지와 대사작용에 영향을 끼치고, 식행동 변화를 보이거나 식욕에 영향을 주기도 한다. 여기서 주목해 보고자 하는 것은 FTO 유전자를 조작시키는 시점에 따라서 다른 표현형을 보인다는 것이다. 생식세포 수준에서 조작했을 때에 비해 세포의 발달이 모두 끝난 시점에서의 FTO 발현 억제는 사망률을 낮추나, 체지방의 증가 및 체중 증가에서는 생식세포 수준의 조작한 모델과 동일하였다. 이는 초기 발달 과정에서 FTO가 체구성이나 비만뿐 아니라 성장에 중요한 추가적 기능이 있다는 것을 시사한다. 이후 자세한 생화학적 분석 및 생쥐모델의의 추가적 분석을 통해 밝혀지리라 기대한다. 이와 같은 추가적 연구는 FTO가 성장발달과 비만과 같은 질환에 미치는 기전을 제공하여 건강한 성장과 질병예방에 적용할 수 있는 기초자료를 제공하리라 사료된다.
