Abstract
본 연구에서는 혐기성 발효공정을 이용하여 고농도 유기성 폐수인 당밀폐수를 기질로 사용하여 다양한 운전조건하에서 수소와 메탄 생산 특성 및 미생물 군집특성을 살펴보고자 하였다. 또한 혼합미생물 배양액 사용시 발생할 수 있는 수소생산 공정내 메탄생성 제어방법과 수소 및 메탄 그리고 미생물 연료전지 공정을 이은 3단 통합공정의 가능성을 살펴보고자 하였다. 각 공정내 활성 미생물의 군집변화는 대용량 염기서열 기법을 통하여 분석하였고, 이러한 미생물 군집특성과 운전인자들과의 상관성을 네트워크 분석법을 통해 규명하였다. 당밀폐수를 이용하여 가수분해/산생성 공정에서 수소생산 성능과 미생물 군집과의 상관성을 조사하였다. 우점종은 Clostridiales와 Lactobacillales이었으나, 수소생산속도는 Pseudomonadaceae와 Micrococcineae와 양의 상관관계를 나타내었다. 총 유기산, acetate, butyrate와 propionate 농도는 Bacillales, Sporolactobacillus와 Lactobacillus 등의 젖산발효균과 양의 상관관계를 보였다. 메탄생성균 억제를 통한 가수분해 효율을 높이기 위한 방법으로 간헐적 탄소원 공급법을 평가하였다. 간헐적 탄소원 공급법 운전으로 인하여 반응조내 우점미생물은 Clostridiaceae에서 Acetobacteraceae로 바뀌었다. 간헐적 탄소원 공급법으로 운전한 이후 연속식 탄소원 공급법으로 전환하여 운전한 뒤에도 메탄농도는 0.3%이하로 유지되었고 메탄생성균도 전체 미생물의 0.2% 이하를 차지하였다. 수리학적체류시간(HRT) 조절방법도 당밀폐수를 이용한 수소생산공정의 장기운전중에 발생하는 메탄생성반응을 억제하는 효과가 있음을 확인하였다. 메탄생산조에서 유입되는 당밀폐수 농도변화에 따른 메탄생산 성능과 활성미생물 군집사이의 상관성을 UASB 반응조에서 평가하였다. 최고 메탄생산속도와 COD 제거율은 당밀폐수 유입농도 16 g-COD L-1 조건에서 각각 0.48 L-CH4 L-1 d-1와 91.2%였다. 메탄생산속도는 Methanosarcina와 Methanosaeta 등을 포함하는 대부분의 메탄생성세균과 양의 상관관계를 나타내었다. 가장 우점한 Methanosarcina는 Bifidobacterium 및 Methanosaeta와 음의 상관관계를 나타낸 반면 Methanosaeta는 Bifidobacterium과 양의 상관관계를 가지는 것으로 나타났다. 동일한 농도의 당밀폐수가 유입되는 메탄생산조에서 HRT 조건변화에 따른 메탄생산 성능과 활성미생물 군집사이의 상호작용 특성에 대하여 조사하였다. 메탄생산 성능과 미생물군집 사이의 상관성분석에 의해 Lactococcus와 Methanosaeta가 네트워크의 거점 주체(hub node)이면서 상호간에도 양의 상관관계를 갖는 것으로 나타났다. 또한 두 균종 모두 메탄생산성과 유기물 부하량과도 양의 상관관계를 가지며 서로 다른 미생물 네트워크들을 이웃하여 공유하고 있는 것으로 나타났다. 수소 생산(BioH2), 메탄 생산 공정(BioCH4) 및 미생물 연료전지(MFC) 공정을 연계하는 3단 통합 공정을 통해 당밀폐수를 처리함으로써 각 공정의 유기물 제거율 및 에너지 생산성에 대한 기여도를 평가하였다. 총 유기물제거량에 대한 BioH2와 BioCH4 및 MFC의 각 공정별 기여도는 각각 20.8%, 72.2%, 7.0%였고, 총 에너지 생산량에 대한 각 공정의 기여율은 각각 18.7, 81.7%, 0.16%이었다. HRT 고려시 BioH2와 BioCH4 및 MFC 공정의 유효용적이 1:2:3의 비율이 적절하였다. 이러한 결과들은 통합 공정 시스템이 에너지 회수 뿐만 아니라 유기성 폐기물 처리에 의한 환경오염 저감 효과도 얻을 수 있으며, 처리대상폐수에 따라 다양한 연계공정 구성이 가능함을 시사한다. ;Global attention has recently been paid to bioenergy production of hydrogen and methane through the anaerobic digestion process using organic wastewater due to fossil fuel depletion and global warming. Molasses wastewater as a sugar-rich waste is one of the most valuable raw material for bioenergy production due to its high organic strength and bioavailability. Interpretation for complex interactions of microbial community structures and operational parameters can help to establish stable biogas production. RNA-based approach for biogas production systems is recommended for analysis of functionally active community members which are significantly underestimated. In this thesis, hydrogen and methane production using molasses wastewater were investigated under variable operational conditions as well as control methods of unfavorable methanogenesis in mixed cultures in biohydrogen production. In addition, the active microbial community was analyzed using RNA-based massively parallel sequencing technique, and their correlation patterns were analyzed using networking analysis. Microbial community associated with hydrogen production performance was investigated in acidogenic hydrogenesis process using molasses wastewater. Although Clostridiales and Lactobacillales were relatively abundant, the hydrogen production rate was positively associated with Pseudomonadaceae and Micrococcineae. Total VFA and acetate, butyrate, and propionate concentrations were positively correlated with lactic acid bacteria such as Bacillales, Sporolactobacillus, and Lactobacillus. Intermittent feeding was evaluated as an appropriate method for selective enrichment of hydrogenesis through methanogen suppression. Intermittent feeding of substrate shifted the dominants in the reactor from Clostridiaceae (70.5%) and Lactobacillaceae (11.0%) to Acetobacteraceae (62.0%) and Clostridiaceae (38.0%). In the reactor operated in continuous feeding mode after intermittent feeding, methane concentration was below 0.3% and the portion of methanogens in the bacterial community was maintained below 0.2%. Controlling the hydraulic retention time (HRT) also had an effect on methanogenesis suppression. The interaction between methane production performance and active microbial community dynamics was characterized at different loading rates by increasing influent substrate concentration in an upflow anaerobic sludge blanket (UASB) reactor using molasses wastewater. The UASB reactor achieved a maximum methane production rate of 0.48 L L-1 d-1 with a chemical oxygen demand (COD) removal efficiency of 91.2% at an influent molasses concentration of 16 g-COD L-1 (OLR of 5.5 g-COD L-1 d-1). The network analysis between operation parameters and microbial community indicated that MPR was positively associated with most methanogenic archaea, including the relatively abundant Methanosarcina and Methanosaeta, except Methanofollis. The most abundant Methanosarcina was negatively associated with Bifidobacterium and Methanosaeta, whereas Methanosaeta was positively associated with Bifidobacterium. The methane production and active microbial community were characterized at different loading rates by changing HRT in an UASB reactor using molasses wastewater. Based on network analysis, Lactococcus and Methanosaeta were network hub nodes and positively correlated each other. In addition, they were positively correlated with methane production and organic loading rate, and they shared the other microbial hub nodes as neighbors. The contribution of individual processes to COD removal and energy production in a three-stage integrated process was evaluated using molasses wastewater. The contribution of hydrogenic process (BioH2), methanogenic process (BioCH4), and a microbial fuel cell (MFC) reactor was 20.8%, 72.2%, and 7.0% of the total COD removal, and 18.7%, 81.2%, and 0.16% of the total energy production, respectively. Hydraulic retention times for BioH2, BioCH4, and the MFC reactor were 1 d, 2 d, and 3 d, respectively. Collectively, the results of the experiments in this thesis can be utilized to design the energy recovery processes of BioH2 and BioCH4 from molasses wastewater and improve the efficiency.
