Abstract
Respiratory syncytial virus (RSV) is an important cause of severe upper and lower respiratory disease in infants and the elderly. RSV has two subtypes, A and B, which are distinguished largely by differences in the patterns of recognition of mAbs and sequence variations within G protein. In this study, I put an emphasis on broad protective efficacy of vaccine candidates consist of two G core fragments from each subtype. The composite vaccine candidate, Gcf/AB, was generated by genetically linking fragment of amino acids 131 to 230 of G protein from RSV A2 and linking fragment of amino acids 131 to 230 of G protein from B1, expressed in E. coli, and purified as a fusion protein. In spite of the absence of any adjuvant, intranasal and intramuscular immunization with Gcf/AB induced strong G-specific antibody responses against both subtypes. Also, intranasal and intramuscular immunization of Gcf/AB induces potent and broad protective immunity against RSV-A (A2 strain) and RSV-B (KR/B/10-12) challenge without vaccine-induced weight loss. These data indicate that Gcf/AB with self-adjuvanticity could be a safe and efficacious vaccine candidate preventing both RSV A and RSV B virus infection.;호흡기 세포융합 바이러스(Respiratory syncytial virus)는 영유아 또는 면역력이 저하된 노인 연령층에서 상하부 호흡기 질환을 일으키는 것으로 중요한 병원체이다. 호흡기 세포융합 바이러스는 G 단백질의 염기 서열 변이와 단일클론항체의 인식 패턴에 따라 크게 A와 B 두 가지 아형으로 나뉜다. 본 연구에서 우리는 각 아형의 두 핵심 단편으로 구성 된 백신 후보물질의 폭 넓은 방어 효능에 중점을 두었다. 복합 백신(composite vaccine) 후보물질 Gcf/AB는 호흡기 세포융합 바이러스의 A2형으로부터 G 단백질의 아미노산 131번부터 230번까지의 단편과, B1형으로부터 G 단백질의 아미노산 131번부터 230번까지의 단편을 유전적으로 연결하여 박테리아에서 발현시키고 정제하여 융합 단백질로서 제작하였다. 면역 증강제 부재 하에서 Gcf/AB는 비강면역화와 근육 내 면역화로 두 아형 모두에 대하여 G 단백질에 특이적인 강한 항체 반응을 유도하였다. 또한 Gcf/AB 비강면역화와 근육 내 면역화는 백신으로 유도된 체중감소 없이 호흡기 세포 융합 바이러스 A형 (A2 바이러스)과 B형(KR/B/10-12 바이러스) 감염에 대해 잠재적이고 폭 넓은 방어면역을 유도하였다. 이러한 연구 결과는 자가면역증강 효과를 가진 Gcf/AB가 A형과 B형 호흡기 세포융합 바이러스 감염을 동시에 예방할 수 있는 안전하고 효과적인 백신 후보물질임을 보여준다.
