dc.description.abstractTopoisomerases are essential enzyme for the proliferation of cells. These enzymes release DNA topological problems during DNA transcription, replication, recombination and segregation. Topoisomerase Type II (Top II) is distinct from Topoisomerase Type I (Top I) which Top II acts as a homodimer, requiring Mg (II) and ATP hydrolysis for enzyme turn over and rapid kinetics. The topoisomerase enzymes have been research targets for the generation of cancer treatments because they play key role in cell division and proliferation. The Top II inhibitors are classified into two main classes based on their mode of action: Top II poison and Top II catalytic inhibitor. In this study, we report that JH-1-41 and JH-1-62 are promising anticancer agents that block topoisomerase catalytic cycle and not act as a poison. To demonstrate this, we systematically investigated the antiproliferative potential of JH-1-41 and JH-1-62 in various cancer cell lines and also clarified the mode of action on topoisomerase inhibitory activity. Based on the results obtained from diverse assays including decatenation, relaxation and unwinding assays of supercoiled DNA and non-stabilization of topo-DNA complex, it is revealed that the JH-1-41 and JH-1-62 act as DNA non intercalating human Top II catalytic inhibitors. Interestingly, JH-1-41 compounds inhibited both Top I and II while JH-1-62 inhibited only Top II significantly. We also elucidated that JH-1-41 and JH-1-62 induce G1 arrest. In summary, we have presented that JH-1-41 and JH-1-62 are potent anticancer agents that are expected to cause less adverse effects than other topoisomerase poison such as etoposide. However, further studies of the molecular mechanisms of anticancer effects of JH-1-41 and JH-1-62, including G1 arrest related signaling change and its mode of inhibition against human topoisomerase as well as DNA damage assay are needed. ;토포이소머라아제는 세포분화에 핵심적인 역할을 하는 효소로써 DNA 전사, 복제 과정에서 발생되어 DNA에 가해지는 긴장감을 해소시키는 역할을 한다. 토포이소머라아제는 크게 두 가지로 구분되며, 토포이소머라아제 I (Top I) 은 복제 시 DNA 의 한 가닥을 잘라 복제 시 발생되는 긴장감을 해소시키고 토포이소머라아제 II (Top II)는 Mg (II) 와 ATP를 소모하여 DNA 두 가닥을 잘라 DNA 가닥이 받게 되는 긴장감에 의해 발생될 수 있는 DNA 손상을 완화 시키는 역할을 한다. 이러한 토포이소머라아제의 역할에 의거하여 현재까지 토포이소머라아제를 표적으로 하는 많은 항암제들이 개발 되고 있다. Top II 를 저해하는 항암제로는 그 작용 기전에 따라 토포이소머라아제 독 과 Top II 촉매억제제로 나눌 수 있다. 본 연구에서는 화합물을 스크리닝 하여 선택한 JH-1-41 과 JH-1-62 화합물이 토포이소머라아제에 어떠한 기전으로 작용하는지 그 작용 메커니즘을 알아보고, 세포상에서 어떠한 손상을 주는지 확인하였다. JH-1-41과 JH-1-62가 토포이소머라아제에 작용하는 기전을 밝히기 위해 Decatenation, Relaxation, Cleavage complex, Unwinding 실험을 수행하였다. 위 실험들을 통하여서 JH-1-41과 JH-1-62는 DNA에 개재하지 않는 토포이소머라아제 II 촉매억제 제 임을 밝혔다. JH-1-41 은 토포이소머라아제 I 과 II를 모두 억제 하는 반면, JH-1-62 는 토포이소머라아제 II 만 억제 하는 특징을 확인하였다. 또한, JH-1-41과 JH-1-62 화합물이 세포상에서 미치는 영향을 알아보고자 세포독성 실험을 DU145(전립선 암세포), HCT15(대장암세포), HEK293 (간세포), T47D (유방암세포) 에서 진행하여 T47D에서 가장 좋은 효과를 보이는 것을 확인하였다. 유방암세포 MCF7 과 정상 유방세포 MCF10A에서의 세포독성 실험을 진행하여 etoposide와 달리 JH-1-41 과 JH-1-62는 MCF7에서 MCF10A에서 보다 강한 세포 독성을 보이는 양상을 보였다. 클론형성분석법을 수행하여 JH-1-41과 JH-1-62가 MCF7에서 MCF10A에서 보다 효과적으로 세포 재생능력을 떨어뜨리는 것을 확인하였다. Etoposide는 세포 독성 실험과 마찬가지로, 정상세포에서의 세포재생능력을 더 떨어뜨렸다. T47D 유방암 세포에서 세포주기 분석 실험을 통하여 JH-1-41과 JH-1-62 화합물 모두 G1 arrest 를 일으키는 것을 확인하였다. 앞으로의 실험에서, G1 arrest 에 관련된 단백질들을 단백질흡입법으로 확인하여 더 구체적인 메커니즘을 확인할 것이고, 코메트검사법을 통하여 DNA에 손상을 주는지를 알아 볼 것이다.-
